dc.description.abstractDas Makrolid-Antibiotikum Roxithromycin und zwölf Solvate von neun verschiedenen Fluorochinolon-Antibiotika werden mit Einkristallröntgenbeugung untersucht. Als Modell der Elektronendichte werden asphärischen Streufaktoren aus der Invariom-Datenbank verwendet. Die Invariom Nomenklatur wird systematisch auf heteroaromatische Verbindungen ausgeweitet. Für die Auswahl von Modellverbindungen werden neue Regeln aufgestellt und validiert. Die Antibiotika werden mit der kleinsten Quadrate Prozedur strukturell verfeinert. Elektrostatische Potentiale und Dipolmomente werden aus der Elektronendichteverteilung berechnet und verglichen. Für das Verständnis der Wirksamkeit von Antibiotika ist Elektrostatik -zusätzlich zum sterischen Raumbedarf- von besonderer Bedeutung.de

dc.description.abstractengThe macrolide antibiotic roxithromycin and twelve solvates of nine different fluoroquinolone antibiotics are investigated by single-crystal X-ray diffraction. Aspherical scattering factors from the invariom database are used to model the electron density. The invariom nomenclature is systematically extended to heteroaromatic systems. New rules for selecting model compounds are established and validated. From the electron-density distribution of least-squares refined structures of antibiotics molecular electrostatic potentials and dipole moments are calculated and compared. In addition to sterical information electrostatics have important implications for understanding antibiotic activity.de
