Insl3 is a testisspecific hormone which governs the development of the gubernaculum ligament needed for the migration of the embryonal testis to its adult scrotal position. We analysed, if the Insl3-controlled gubernaculum development depends on the presence of androgens and if overexpression of Insl3 in female embryos induces gubernaculum development leading to the descensus of the ovaries. Interestingly female transgenic mice developed bilateral inguinal hernia. Therefore the fertility of transgenic mice was measured. Furthermore we examined the effect of the Insl2/Insl3 transgenic allele on Insl3-deficient males. In addition to that questions concerning the regulation of Insl3 were covered. Within the promoterregion of Insl3 a cis-acting element was analysed and the corresponding transacting factor was identified as Apg-1, a member of the hsp110 related family of heatshock proteins. Luciferaseassays revealed its role regulating the Insl3 gene. Subcellular localisation provided more evidence, that the identified transacting element acts as a transcription factor. We also analysed the expression pattern of apg-1 during postnatal testis development. Northern Blots revealed the temperature dependend nature of Apg-1 expression. To verify the exact length of the 5`-prime UTR in vivo, transgenic mice were generated and the expression of the transgenic allele was examined.

Die Aufgabe der vorliegenden Arbeit war es zu untersuchen, ob die Insl3-gesteuerte Gubernaculumentwicklung von Androgenen abhängig ist und ob die Überexpression von Insl3 in weiblichen Embryonen zu Entwicklung des Gubernaculums führt und damit den Descensus der Ovarien induziert. Zu diesem Zweck wurden transgene Mäuse hergestellt, die Insl3 im Pankreas überexprimieren. Interessanterweise zeigten sich bei weiblichen transgenen Mäusen bilaterale Inguinalhernien. Daher wurde die Fertilität transgener Weibchen und die Wirkung des Insl2/Insl3 Transgens bei Insl3-defizienten Männchen untersucht. In dieser Arbeit sollten auch Fragen zur Regulation des Insl3 Gens geklärt werden. Innerhalb der Promotorregion des Insl3 Gens sollte durch Gel Retardation Assays ein cis-ständiges Element charakterisiert und der daran bindende transagierende Faktor als Mitglied der hsp110 verwandten Heatshock-Proteinfamilie identifiziert werden. Durch Luciferaseassays sollte die Beteiligung des transagierenden Faktors an der Regulation des Insl3 Gens untersucht werden. Zur Bestimmung der subzellulären Lokalisation des transagierenden Faktors wurden GFP-Fusionsproteine herstellt, in kultivierte Zellen transient transfiziert und fluoreszensmikrokopisch untersucht. Außerdem wurde in diesem Zusammenhang das Expressionsmuster des transagierenden Faktors in der postnatalen Testisentwicklung untersucht und Untersuchungen zur temperaturabhängigen Expression durchgeführt. Um die Transfektionsexperimente zur Ermittlung des 5`-flankierenden Bereich des Insl3 Gens der Maus in vivo zu bestätigen, wurden transgene Mäuse erzeugt und in Northern-Blot-Analysen auf Expression des transgenen Allels untersucht.
