Apoptosis is a form of programmed cell death, which is regulated by numerous gene products. Many of these genes are mutated or deregulated in human carcinoma. Bax inhibitor-1 (BI-1) gene codes for an anti-apoptotic protein, which is upregulated in carcinogenesis and inhibits the effects of Bax. In the present study, we could show an upregulation of BI-1-expression in mammary-, uterus- and ovarian carcinoma. In addition, we were able to show an expression of BI-1 in human mammary carcinoma cell line MDA-MB-231, MDA-MB-453, MCF-7, HCC70, ZR-75-1 and T-47D. Down-regulation of BI-1-expression leads to apoptosis of 32% of MDA-MB-231 cells 96 h after siRNA transfection. In the second part of the study, DIO-1 could be identified as BI-1 interaction-partner and was chosen for further analysis because of its pro-apoptotic role in apoptosis. The analysis of subcellular colocalization shows, that DIO-1 is colocalized with BI-1 in the cytoplasma. The expression analysis with human mammary- and prostate carcinoma cell lines shows a 38% of nuclear localization, 26% of partial nuclear localization and 36% of cytoplasmatic localization of DIO-1 in PC-3 cells. The downregulation of expression of both BI-1 and DIO-1 using siRNA in prostate- and mammary carcinoma cell lines did not reveal any upregulation in caspase-3 activity in comparison to the control transfected cells. In the third part of the study, we investigated the effect of the apoptosis induction using BI-1 specific duplex-siRNA oligonucleotids in mammary carcinoma of nude mice. Two weeks after implantation of MDA-MB-231 cells in nude mice, tumors developed, which were treated with BI-1-siRNA/Atelocollagen (AC)-complexes. Using an eXplore Optix-System, it was possible to localize siRNA coupled with Cy5.5 in vivo. The siRNA/Ac complex was detectable and stable during the entire period in the tumor. Tumor development was observed with flat-panel volumetric computer tomography (fpVCT) every second day and analysed. It could be demonstrated that BI-1-siRNA/Ac treated tumors grew slower than Luc-siRNA/Ac treated tumors with fpVCT data (77 mm³ differences in tumor end volume). Taken together, our results indicate that BI-1 could play an important role in the tumor progression of mammary carcinoma. BI-1-siRNA/Ac treatment could be a possible approach for a tumor therapy. The usage of molecular optical system is an effective method for investigation of anatomical and physiological changes at the molecular level of the tumor-mouse-model.

Die Apoptose stellt eine Form des programmierten Zelltods dar, die von zahlreichen Genprodukten gesteuert wird. Viele dieser Gene sind in humanen Karzinomen mutiert oder fehlerhaft reguliert. Das Bax Inhibitor-1 (BI-1)-Gen kodiert für ein antiapoptotisches Protein, welches die Wirkung von Bax aufheben kann. Für das BI-1-Gen konnte eine erhöhte Expression bei der Progression von Tumoren nachgewiesen werden. In der vorliegenden Arbeit wurde eine Hochregulierung der BI-1-Expression bei Mamma-, Uterus- und Ovarialkarzinomen festgestellt. Weiterhin wurde die Expression von BI-1 in den humanen Mammakarzinomzelllinien MDA-MB-231, MDA-MB-453, MCF-7, HCC70, ZR-75-1 und T-47D nachgewiesen. Die Herunterregulierung der BI-1-Expression führte zur Apoptose in 32% der MDA-MB-231-Zellen 96 Stunden nach der siRNA-Transfektion. Im zweiten Abschnitt der Arbeit wurde DIO-1 als BI-1-Interaktionspartner identifiziert und aufgrund seiner pro-apoptotischen Rolle in der Apoptose weiter analysiert. Die Analyse zur subzellulären Kolokalisation zeigte, dass DIO-1 zusammen mit BI-1 im Zytoplasma lokalisiert ist. Die durchgeführte Expressionsanalyse an humanen Mamma- und Prostatakarzinomzelllinien zeigte, dass DIO-1 in allen Zelllinien exprimiert wird. Die subzelluläre Lokalisation von DIO-1 nach Herunterregulierung der BI-1-Expression in Prostatakarzinomzelllinien zeigte in 38% der PC-3-Zellen eine nukleäre Lokalisation, in 26% eine partiell nukleäre und in 36% eine zytoplasmatische Lokalisation von DIO-1. Bei der gemeinsamen Herunterregulierung der BI-1- und DIO-1-Expression mittels RNAi in den Prostata- sowie Mammakarzinomzelllinien wurde keine gesteigerte Caspase-3-Aktivität im Vergleich zu den kontrolltransfizierten Zellen gemessen. Im dritten Abschnitt der Arbeit wurde der Effekt der Apoptose-Induktion durch die Einführung von BI-1-spezifischen Duplex-siRNA-Oligonukleotiden in induzierten Mammakarzinomen bei Nacktmäusen untersucht. Etwa zwei Wochen nach der Implantation von MDA-MB-231-Zellen in die Nacktmäuse entwickelten sich Tumoren, die mit BI-1-siRNA/Atelocollagen (Ac)-Komplexen behandelt wurden. Über das eXplore Optix-System war es möglich, die mit Cy5.5 gekoppelte siRNA in vivo zu lokalisieren. Der siRNA/Ac-Komplex blieb über den gesamten Zeitraum hinweg im Tumor detektierbar und weitestgehend stabil. Über Flächendetektor-Volumetrische-Computertomographie (FD-VCT) wurde die Tumorentwicklung alle zwei Tage beobachtet und die Daten analysiert. Die mittels FD-VCT segmentierten Tumordaten belegen, dass die BI-1-siRNA/Ac-behandelten Tumoren langsamer wuchsen als die mit Luc-siRNA/Ac-behandelten Kontrolltumoren (77 mm³ Tumorendvolumenunterschied). Zusammenfassend zeigen die Ergebnisse der Arbeit, dass BI-1 eine wichtige Rolle in der Tumorprogression von Mammakarzinomen spielt. Daher kann eine BI-1-siRNA/Ac-Behandlung einen möglichen Ansatz für eine Tumortherapie darstellen. Die hier verwendeten molekularen optischen Bildgebungsverfahren stellen weiterhin eine effektive Methode zur Untersuchung von anatomischen und physiologischen Veränderungen bei Tumor-Maus-Modellen auf molekularer Ebene dar.
