The epigenetic regulation of gene expression plays a very important role in development and is often mediated by the methylation of histones. Trimethylation of histone 3 lysine 9 (H3K9me3) leads to chromatin silencing and the formation of heterochromatin by recruitment of heterochromatin protein 1 (HP1). Eggless (Egg), the ortholog of the human methyltransferase SETDB1, is required for H3K9 trimethylation in the female germ line of Drosophila. Here we show that Windei (Wde), the Drosophila homolog of mouse mAM and human MCAF binds to Egg and precisely colocalizes with Egg in ovaries. Wde localizes to euchromatin, is enriched on chromosome 4 and binds to the Painting of fourth (POF) protein. Null mutations in wde are homozygous lethal. Germ line cells mutant for wde show strongly reduced H3K9me3 staining and degenerate by apoptosis. Since the mutant phenotypes of wde and egg are indistinguishable, we propose that Wde is an essential binding partner of Egg and is indispensable for H3K9 trimethylation in germ line cells, which is essential for the establishment of the germ line specific chromatin state.

Die epigenetische Regulation der Genexpression spielt eine sehr wichtige Rolle in der Entwicklung und wird häufig über die Methylierung von Histonen vermittelt. Die Trimethylierung von Histon 3 am Lysin 9 (H3K9me3) führt zur Ruhigstellung des Chromatins und zur Bildung von Heterochromatin durch die Rekrutierung von Heterochromatin Protein 1 (HP1). Eggless (Egg), das Ortholog der humanen Methyltransferase SETDB1, ist für die H3K9-Trimethylierung in der weiblichen Keimbahn von Drosophila notwendig. Hier zeigen wir, dass Windei (Wde), das Drosophila-Homolog des murinen mAM und des humanen MCAF1, an Egg bindet und präzise mit Egg in Ovarien colokalisiert. Wde lokalisiert am Euchromatin, ist am vierten Chromosom angereichert und bindet an Painting Of Fourth (POF). Nullmutationen in wde sind homozygot letal. Wde mutante Keimzellen zeigen eine starke Reduktion der H3K9me3-Färbung und degenerieren durch Apoptose. Da die mutanten Phänotypen von wde und egg nicht voneinander zu unterscheiden sind, schlussfolgern wir, dass Wde ein essenzieller Bindungspartner von Egg und unverzichtbar für die H3K9-Trimethylierung in Keimbahnzellen ist, was für die Etablierung des Keimbahn-spezifischen Chromatinstatus notwendig ist.
