Injections of proctolin into the protocerebrum can elicit species-specific stridulation in both male and female grasshoppers of the species Chorthippus biguttulus. The stimulated behavior is similar to the natural stridulation with respect to both the temporal structure and patterns of stridulatory movements of the hindlegs. Compared to stimulation of stridulation by muscarine, proctolin generally elicited shorter stridulation after a shorter latency. Muscarinic AChRs-activation may lead to prolonged calling singing. In contrast, proctolin, when injected to the same site within the brain as muscarine, elicited song sequences of shorter duration and additional elements (wing flapping, precopulatory movements etc.) normally seen during courting. This suggested that proctolin release might be associated with courtship singing and could probably trigger switches between calling and courtship singing when a con-specific female responds acoustically. In addition, mAChRs-activation (blocked by scopolamine) is not necessary for proctolin-stimulated stridulation.Stimulatory effects of proctolin are terminated by desensitization of proctolin receptors, that appeared with short inter-stimulus intervals (<30 sec). Proctolin mediates its excitatory effects in the neural circuits that control stridulation through receptors that activate a phospholipase C (blocked by neomycine and Li+), protein kinase C (phorbol ester activated) and ryanodine-insensitive Ca2+ release from intracellular stores. Extracellular Ca2+ influx might be also involved (blocked by 5 mM Ni2+) in proctolin mediated excitation. Receptor specific activation of PLC by muscarine (mAChRs), proctolin (PRs), glutamate (mGluRs(1,5)) within the control circuit is postulated. Activation of adenylate cyclase (blocked by SQ22536) had only weak effects on proctolin-stimulated singing. Stridulation could also be stimulated through mGluRs(1,5)-activation (DHPG), while dopamine seemed to suppress proctolin-stimulated singing. Co-stimulation of proctolin with a second transmitter could alter the time course of proctolin-stimulated stridulation (glycine, histamine, glutamate) or lead to the appearance of other courtship elements - wing flapping and precopulatory movements (glutamate). Possible mechanisms and functions of co-transmission in the stridulation-controlling neuronal circuit are discussed.

Das artspezifische Gesangsverhalten der Feldheuschrecke Chorthippus biguttulus läßt sich sowohl bei Männchen als auch bei Weibchen durch Injektionen des Pentapeptids Proktolin in das zentrale Protozerebrum auslösen. Das stimulierte Verhalten entspricht bezüglich der seiner zeitlichen Struktur und der Bewegungs- und Lautmuster weitgehend der natürlichen Stridulation. Verglichen mit der durch Muskarin ausgelösten Stridulation tritt das durch Proktolin induzierte Verhalten zwar nach kürzerer Latenz auf, ist aber auch von kürzerer Dauer. Gesangsstimulation durch Proktolin erfolgt dabei unabhängig von der Aktivierung muskarinischer ACh-Rezeptoren, die experimentell durch Skopolamin blockiert werden können. Während Aktivierung muskarinischer ACh-Rezeptoren zumeist Werbegesang auslöst führt Proktolin an identischem Stimulationsort zur Durchführung zusätzlicher Balzelemente (Flügelflattern, Anspringbewegungen etc.) die dem Werbeverhalten der Feldheuschrecken zuzuordnen sind. Endogene synaptische Ausschüttung von Proktolin könnte daher den Übergang von Rufgesängen zu Werbegesängen initiieren, welcher möglicherweise durch den Gesang eines artgleichen Weibchens in unmittelbarer Nähe ausgelöst wird.Der Gesang stimulierende Effekt von Proktolin verschwindet bei wiederholter Applikation in kurzen Intervallen (<30s), was auf eine Desensitisierung der Proktolinrezeptoren hindeutet. Proktolin vermittelt Erregung im Gesang kontrollierenden Hirnneuropil durch Aktivierung von Phospholipase C (hemmbar durch Neomycin und Lithiumionen), Proteinkinase C (direkt aktivierbar durch Phorbolester) und nicht Ryanodin sensitive Kalziumausschüttung aus intrazellulären Speichern. Auch Kalziumeinstrom aus dem Extrazellulärraum (hemmbar durch 5 mM Ni2+) könnte zur Gesamterregung beitragen. Hemmung der Adenylatzyklase durch co-injiziertes SQ22536 führte zu kleiner Veränderung der durch Proktolin stimulierten Gesangsaktivität. Desweiteren konnte spezifisches Stridulationsverhalten durch Aktivierung metabotroper Glutamatrezeptoren (mGluRs(1,5) durch DHPG) induziert werden, wogegen Dopamin das durch Proktolin stimulierte Verhalten inhibiert. Durch Proktolin ausgelöste Stridulation konnte auch durch Co-Stimulation anderer Transmitter (Glyzin, Histamin, Glutamat) in ihrem Zeitverlauf und/oder in Bezug auf die durchgeführten Stridulationsbewegungen moduliert werden. Mögliche Funktionen und Mechanismen solcher Co-Trasmissionen im Stridulation kontrollierenden Hirnneuropil von Feldheuschrecken werden diskutiert.
