Intinsic optical signals (IOS) regularly occur together with hypoxia induced spreading depression (HSD). IOS show a characteristic developement. First tissue reflectance becomes a little bit lower than during normoxic conditions. Cell schwelling during neuronal activity is a possible reason for the reduced tissue reflectance. A short time after the peak of HSD tissue reflectance becomes more intensive, especially in the wavelength of λ=380-560nm. Only at λ=440nm the reflectance is lower than in the other wavelength. NADH is possibly responsible for the increase of reflectance. Arround the wavelegth of λ=440nm mitochondrial cytochromes a, a3 and c have their maximal absorption under hypoxic conditions. After reoxygenation tissue reflectance returns nearly to the normoxic begining of the experiments. At next the reflectance increases a little bit and stays at this achieved intensity till the end of the registration period. Irreversible damage in dendrits is supposed to lead to this second reflectance increase (dendritic beading). Ion substitution experiments and drug treatment caused characteristic changes in the complex reflectance profile. Ca2+substitution intensifies, whereas Cl-substitution reverses the IOS. Hypotonicity intensifies the IOS, too. In hypertonic ACSF (artificial cerebrospinal fluid) the IOS become attenuated. Mitochondria play a key role in the developement of IOS, especially in the wavelength of λ=340-440nm. Glia cells take part in the initial reflectance decrease during early hypoxia. Wavelength λ>460nm are well suited for registration of IOS.

Intrinsische optische Signale (IOS) entstehen grundsätzlich im Zusammenhang mit einer hypoxieinduzierten Spreading Depression (HSD). In der Entwicklung der IOS sind mehrere markante Phasen zu unterscheiden. Bevor es im Anschluss an die HSD zu einer Intensitätssteigerung der IOS kommt, ist zunächst eine geringe Abschwächung der optischen Signale zu beobachten. Diese lässt sich auf die Schwellung von Neuronen und Gliazellen während der neuronalen Erregung zurückführen. Der HSD folgt unmittelbar eine deutliche Intensitätszunahme der IOS, die vor allem die Wellenlängenbereiche λ=380-560nm erfasst. Das kann teilweise mit dem Anstieg der Autofluoreszenz von NADH unter hypoxischen Bedingungen begründet werden. Betroffen sind vorrangig die Wellenlängen λ=400-460nm. Ein tiefer Einschnitt im Bereich um λ=440nm im Reflexionsprofil ist auf die gesteigerte Absorption der mitochondrialen Cytochrome a, a3 und c zurückzuführen. Nach Reoxygenierung nimmt die IOS-Intensität wieder ab. Das Ausgangsniveau vor der Hypoxie wird aber nicht erreicht. Die IOS werden danach im gesamten beobachteten Wellenlängenbereich λ=320-680nm erneut kräftiger. Dafür ist vermutlich die partiell irreversible Schädigung der Dendriten verantwortlich (dendritic beading). Dieser charakteristische Verlauf in der IOS-Entwicklung ist am gleichen Gewebe mehrfach reproduzierbar. Kalziumionen limitieren den Intensitätszuwachs der IOS. Zudem sind IOS strikt chloridabhängig. Hypotone ACSF-Lösung intensiviert die optischen Signale. Im Gegensatz dazu werden die IOS in hypertoner Lösung abgeschwächt. Gliazellen tragen offenbar zum initialen Reflexionsrückgang vor der HSD bei, dennoch haben die IOS überwiegend einen neuronalen Ursprung. Die IOS haben zu großen Teilen ihr morphologisches Korrelat in den Mitochondrien. Der Wellenlängenbereich λ>460nm ist am Besten geeignet für IOS-Messungen, da hier die stabilsten Signale auftreten.
