In patients with myelodysplastic syndromes (MDS) iron overload caused by long term red blood cell transfusions is an important factor for comorbity. In this report we present the results of our comparative study based on colony assays and FACS analyses of hematopoetic cells in MDS patients with and without iron overload. We can demonstrate, that iron overload suppresses the proliferation of erythroid progenitors cells (BFU-E) while the myeloid compartment (CFU-GM) was not found to be affected. Patients with slightly elevated ferritin values show an impaired proliferation capacity in comparison to patients with normal ferritin levels. Furthermore, we can show, that this negative impact is reversible by sufficient iron chelation therapy. Apoptosis of hematopoietic progenitor cells was not found to be affected by iron overload.

Bei Patienten mit myelodysplastischen Syndromen (MDS) ist die transfusionsbedingte Eisenüberladung ein wichtiger Faktor für Komorbiditäten. In dieser Dissertation werden Progenitorzelleigenschaften zwischen Patienten mit normalen und erhöhten Ferritinwerten verglichen. Dies basiert auf Colony Assays und durchflusszytometrischen Apoptosemessungen an hämatopoetischen Progenitorzellen. Eisenüberladung unterdrückt die Proliferation erythroider Progenitorzellen (BFU-E) signifikant, die myeloischen Vorläuferzellen sind nicht betroffen.Auch Patienten mit nur leicht erhöhten Ferritinwerten zeigten eine signifikant erniedrigte Proliferation (BFU-E) im Vergleich zu Patienten mit normalen Ferritinwerten. Weiterhin konnte gezeigt werden dass dies durch eine Chelattherapie zumindest partiell reversibel ist. Die Apoptose und Nekrose von CD 34 positiven Zellen wird durch eine Eisenüberladung nicht beeinflusst.
