The final step in hepatitis b virus formation is the envelopment of the pre-assembled nucleocapsid at the membrane of the endoplasmatic reticulum which contains the viral surface proteins. It is already shown that amino acids within the range of 98 to 124 of the large envelope- protein are essential for the envelopment. In the present dissertation the phenotypical characterization of deleted constructs from the large envelope-protein shall reveal the influence on virus formation of the so far not well investigated domain between amino acids 170 to 253 of the large envelope-protein which is arranged to the cytosolic side of the endoplasmatic reticulum.

Der die Morphogenese des Hepatitis B Virus
abschliessende Schritt ist die Umhüllung des
cytoplasmatisch vorgebildeten Nukleokapsids an der
Membran des Endoplasmatischen Retikulums, welche die
viralen Oberflächenproteine enthält. Es konnte gezeigt
werden dass der Bereich zwischen den Aminosäuren 98 und
124 des großen Hüllproteins für die Umhüllung
essentiell ist. In der vorliegenden Arbeit soll durch
phänotypische Charakterisierung deletierter Konstrukte
des großen Hüllproteins aufgeklärt werden, welchen
Einfluss die bisher noch nicht untersuchte, zum Cytosol
gerichtete Domäne zwischen den Aminosäuren 170 und 253
des großen Hüllproteins auf die Virusbildung hat.
