Members of mammalian uncoordinated 13 protein family (Munc13s) are essential for SV exocytosis in neurons and have also been implicated in LDCV exocytosis in chromaffin cells. However, the C.elegans ortholog Unc-13 appears to be dispensable for LDCV exocytosis, raising the question whether SNARE-mediated exocytosis of SVs and LDCVs is controlled by distinct regulatory proteins. We therefore analyzed LDCV exocytosis in cultured chromaffin cells taken from knockout mouse lines of four members of the Munc13 protein family, Munc13-1, Munc13-2, Munc13-3 and Baiap3. Baiap3/Bap3 (brain specific angiogenesis inhibitor 1 associated protein 3), was identified and named as an interaction partner of BAI 1 (brain specific angiogenesis inhibitor-1), and is a Munc13 homologue of unknown function. Its closet known relative, the non-neuronal Munc13-4, has been shown to be essential for the secretion of lymphocyte cytolytic granule content. Unlike Munc13-4, Baiap3 expression is largely restricted to brain and adrenal gland, with low levels also present in lung. To determine whether LDCV exocytosis is impaired in the absence of Munc13-1, Munc13-2, Munc13-3 or Baiap3, we combined flash photolysis of caged Ca2+ and membrane capacitance measurements in cultured chromaffin cells. Our analysis of exocytosis in chromaffin cells shows that Munc13-1 and Munc13-2 act as positive regulators of LDCV exocytosis and play a more critical role in the release of this vesicle type than their C.elegans ortholog Unc-13. Based on sequence similarity, Baiap3 is a member of the Munc13 protein family. However, unlike Munc13-1, -2, -3 and -4, which function as essential positive regulators of SNARE-mediated exocytosis, our data indicate that Baiap3 negatively regulators exocytosis, at least in chromaffin cells. In this study, we found that exocytosis in chromaffin cells from Munc13-1 deficient mice and mice lacking both Munc13-1 and Munc13-2, showed a dramatic reduction in LDCV exocytosis. In contrast to this, chromaffin cells deficient for Baiap3 showe d increased exocytosis and Baiap3 overexpression in WT cells suppressed LDCV release. The exact molecular mechanism by which Munc13s regulate SNARE-mediated exocytosis remains to be elucidated. However, we found that Baiap3 interacted with both Munc13-1 and Syntaxin in co-sedimentation assays, which indicates that competition for binding to SNARE complex components is a possible explanation for the opposing functions of Munc13-1/2 and Baiap3 LDCV exocytosis.

Munc13 Proteine sind eine Familie von essentiellen
Regulatoren der Vesikel-Exozytose. Munc13 steht für
mammalian uncoordinated 13 , und spiegelt den Phänotyp
der C. elegans Mutante unc-13 (uncoordinated 13)
wieder. Fehlen Munc13s in den Neuronen von Säugetieren
(M. musculus) oder die entsprechenden Orthologe in
Invertebraten (C. elegans und D. melanogaster) wird die
Fusion der synaptischen Vesikel (SVs) mit der
Plasmamembran unterbunden. Basierend auf
Überexpressionsstudien wurde eine ähnliche
Kontrollfunktion bei der Exozytose von
Large-Dense-Core-Vesikeln (LDCVs) für
Säugetier-Chromaffinzellen postuliert.
Überraschenderweise scheint aber Unc-13 in C. elegans
keine essentielle Rolle bei der Exozytose von LDCVs zu
spielen, was die Frage aufwirft, ob die Freisetzung von
SVs und LDCVs von unterschiedlichen Regulatoren
kontrolliert wird. Um diese Frage zu beantworten, haben
wir LDCV-Exozytose in Chromaffinzellen von
Deletionsmutanten (Knockout-Mäusen) von vier
Mitgliedern der Munc13-Familie (Munc13-1, Munc13-2,
Munc13-3 und Baiap3) untersucht. Baiap3/Bap3 (brain
specific angiogenesis inhibitor 1 associated protein 3)
ist ein Munc13-Homolog unbekannter Funktion, welches
ursprünglich als Interaktionspartner von BAI1 (brain
specific angiogenesis inhibitor 1) isoliert wurde. Der
nächste Verwandte von Baiap3 ist Munc13-4, welches
nicht in Neuronen exprimiert wird, aber ein
essentieller Regulator der Exozytose der zytolytischen
Vesikel von Lymphozyten ist. Baiap3 wird im Gegensatz
zu Munc13-4 im Gehirn und im Nebennierenmark exprimiert
und es kommt in geringeren Mengen auch in der Lunge
vor. Um festzustellen ob LDCV-Exozytose durch die
Abwesenheit von Munc13-1, Munc13-2, Munc13-3 oder
Baiap3 beeinträchtigt wird, kombinierten wir
Flash-Photolyse von caged Ca2+ mit
Membrankapazitätsmessungen in isolierten
Chromaffinzellen der entsprechenden
Knockout-Mauslinien. Unsere Analyse dieser Experimente
zeigt, dass Munc13-1 und Munc13-2 positive Regulatoren
der LDCV-Exozytose in Chromaffinzellen sind. Damit
scheinen Munc13-1 un d Munc13-2 eine kritischere Rolle
bei der LDCV-Exozytose zu spielen als ihr Orhtolog in
C. elegans, Unc-13. Baiap3, obwohl es aufgrund seiner
Domänenstruktur und Sequenzhomologie zur Munc13-Familie
dazugehört, scheint eine der den anderen Homologen
entgegengesetzte Rolle bei der LDCV-Exozytose zu
spielen. In der Abwesenheit von Munc13-1 und Munc13-2
war die LDCV-Exozytose drastisch reduziert, während in
Abwesenheit von Baiap3 verstärkt LDCV-Exozytose
stattfand und Baiap3-Überexpression zur Abschwächung
der Exozytose führte. Damit ist Baiap3 das erste
Mitglied der Munc13-Familie, für das eine negative
regulatorische Funktion der SNARE-vermittelten
Exozytose gezeigt werden kann. Der genaue molekulare
Mechanismus durch den Munc13-Proteine Vesikel-Exozytose
regulieren ist nicht bekannt. Wir konnten aber
feststellen, dass Baiap3 sowohl mit dem SNARE-Protein
Syntaxin als auch mit Munc13-1 interagiert, was eine
Konkurrenz um die Bindung von Komponenten des
SNARE-Komplexes als wahrscheinlich erscheinen
lässt.
