Small conductance, Ca2+ - activated K+ channels (SK) are responsible for afterhyperpolarizations (AHPs) regulating neuronal excitability and thus contribute to learning and memory. Here, we determined the regulation of SK channel gene expression and describe their possible role in cognitive processes during aging and in response to stress. We showed that transcription of SK2 channels is regulated by glucocorticosteroid (GRs), mineralocorticocoid receptors (MRs) and transcription factor nuclear factor KappaB (NF-kappaB) in PC12 cells and mouse hippocampus. In chapter 2 we found that selective agonists for GRs and MRs, corticosterone, dexamethasone and aldosterone increased SK2 transcription about two fold. This effect was blocked by pretreatment of their selective antagonists mifepristone and spironolactone. C2-ceramide, activating NF-kappaB, doubled the amount of SK2 mRNA. Treatment of antisense probes against the NF-kappaB subunits p50 and p65 successively reduced C2-ceramide induced increase of SK2 transcription in PC12 cells. In chapter 3, we showed that the amount of SK2 transcripts increased two hours after one-hour immobilization in Balb/c mouse hippocampus using semi-quantitative RT-PCR. Subcutaneous injection of spironolactone blocks stress-induced increase of SK2 mRNA 2 hours after one-ho ur immobilization. Intracerebroventricular injection of SN50, a selective peptide blocker against NF-kappaB, decreased the amount of SK2 mRNA in the hippocampus of Balb/c and C57BL/6J mice. Age-related decrement in the performance of some learning tasks is correlated with substantial changes in neuronal signal processing in the hippocampus. Chapter 4, we showed that elevated expression of SK3 channels in hippocampi of aged mice contributes to reduced long-term potentiation (LTP) and impaired trace fear conditioning, a hippocampus-dependent learning task.

Kalzium-aktivierte Kaliumkanäle (SK) sind
verantwortlich für die sogenannte
"afterhyperpolarization" (AHP) und regulieren somit die
neuronale Erregbarkeit. Damit spielen sie auch eine
entscheidende Rolle bei Lernvorgängen. In der
vorliegenden Arbeit wurde die Regulation der Expression
von SK Kanälen während Alterungsprozessen und im
Zusammenhang mit Stresserfahrungen beschrieben. Wir
konnten zeigen, dass die Transkription des SK2 Kanals
durch Rezeptoren für Glucocorticoide (GRs),
Mineralocorticoide (MRs) und NF-kappaB in PC12 Zellen
und dem Hippokampus der Maus reguliert wird. Im Kapitel
2 wird beschrieben, dass selektive Agonisten für GRs
und MRs wie Corticosterone, Dexamethasone und
Aldosterone die Transkriptionsrate von SK2
verdoppelten. Die jeweiligen Antagonisten Mifepristone
und Spironolactone konnten diesen Effekt blockieren.
Der NF-kappaB Agonist C2-Ceramide führte ebenfalls zu
einer Verdopplung der Transkription von SK2. Die
Behandlung mit Antisense-Proben gegen die NF-kappaB
Untereinheiten p50 und p65 hatte eine schrittweise
Reduktion des C2-Ceramide Effektes zur Folge. Im
Kapitel 3 konnte mit Hilfe von RT-PCR nachgewiesen
werden, dass zwei Stunden nach Immobilisierung für eine
Stunde SK2 mRNA im Hippokampus von Balb/c Mäusen erhöht
war. Diese Erhöhung wurde durch subkutane Injektion von
Spironolactone verhindert. Die basale Expression von
SK2 mRNA konnte im Hippokampus von Balb/c und C57BL/6J
Mäusen durch die intracerebroventriculare Injektion des
NF-kappaB Antagonisten SN50 reduziert werden. Die
altersabhängige Abnahme von Lernleistungen ist mit
Veränderungen in Signalvorgängen des Hippokampus
deutlich korreliert. Im Kapitel 4 konnte nachgewiesen
werden, dass die erhöhte Expression von SK3 Kanälen im
Hippokampus von alten Mäusen zu reduzierter Lang-Zeit
Potenzierung (LTP) und verminderter Furcht
Konditionierung führt.
