dc.description.abstract | Drei aufeinanderfolgende Glucosylierungen
sind die letzten Stufen bei der Bildung der Dolichol-PP-Glycane
bevor diese an neugebildete Proteine im Endoplasmic Reticulum (ER)
übertragen werden. Diese drei Glucosylreste sind wichtig bei der
Proteinfaltung und Qualitätskontrolle im ER. Der dritte und zweite
Rest werden nacheinander abgespalten, um dann als
mono-Glucosyl-Glycane vom Calreticulin / Calnexin System erkannt zu
werden. Abschließend werden alle Glucosereste vor dem Austritt aus
dem ER abgeschnitten. Mit meinen Experimenten habe ich die Funktion
der Glucosylierungs Enzyme (Alg5/wol, Alg6/gny und Alg8/CG4542) bei
der Zellbewegungen während Gastrulation von Drosophila melanogaster
analysiert. Mein Fokus war die Funktion von E-Cadherin bei der
Zell-Interkalation. Ich konnte zeigen, dass E-Cadherin ein
relevantes Substrat der ER-glykolysierungsenzyme ist und dass die
E-Cadherin-Menge in diesen Mutanten reduziert ist. Die Reduktion
von E-Cadherin durch RNAi führt zu einen entsprechenden Phänotyp.
Ich habe beobachtet, dass sich die neuen Zell-Grenzen durch
periodische Expansion bilden, auf die E-Cadherin anreicherung
folgt. Die Intensität von E-Cadherin und die Länge der neuen
Grenzen sind anti-korreliert. Ich postuliere, dass E-Cadherin nicht
die treibende Kraft für die Erweiterung der neuen Zell-Grenzen ist,
aber die erweiterten Grenzen stabilisiert. | de |

dc.description.abstracteng | Three consecutive glucosylations are the
last steps in formation of the dolichol-PP-glycans before the
glycans are transferred to nascent proteins in the ER. These three
glucosyl residues are assumed to function in protein folding and ER
quality control since they are consecutively cleaved to allow
folding before the mono-glucosyl-glycan is recognized by the
calreticulin/calnexin system. Finally, all glucoses are clipped off
before ER exit. In my studies, the function of these glucosylation
enzymes (Alg5/wol, Alg6/gny and Alg8/X-330) has been analyzed in
the movement and morphogenesis of gastrulation and in Drosophila
embryonic development. I have focused on their function in cell
intercalation and found the expression of the integral membrane
protein E-Cadherin strongly reduced and partially glycosylated in
the mutants. Consistently, reduced expression of E-cadherin induced
by RNAi leads to a comparable phenotype, which indicates that
E-cadherin is a relevant down stream ta! rget of the X-330 mutant
for the cell intercalation defect. To study the mechanism of new
border formation in cell intercalation, I have observed the new
borders extended via pulsed manner with E-cadherin accumulated soon
after. E-cadherin intensity and the length of new borders are
anticorrelated. We propose that E-cadherin could not provide the
force for new border extension, but functions to stabilize the
