The part of purinergic (ATP-) receptors in the pathogenesis of amyotrophic lateral sclerosis (ALS) is so far not thoroughly understood. Current knowledge about the pathogenesis of ALS mostly depends on research with transgenic rodents, which show mutations in the gene encoding the Cu/Zn-superoxide-dismutase (mSOD1). Purinergic receptors regulate the activity and motility of microglia as well as calcium-depended processes and the transmembrane potential in motoneurons by calcium-influx and -release. We examined the expression of the ionotropic purinergic P2X4-receptor in different points of time during the course of disease in the ventral horn of lumbal spinal cord sections of WT and SOD G93A-mice and human postmortal thoracal spinal cord per immunohistochemistry and Western Blot analysis. In WT-mice a low expression of the receptor was mostly found on axons of MNs during the postnatal period whereas the P2X4-like immunoreactivity (P2X4-LIR) was stronger and mainly located on the somata in WT-adults. Compared to the WT, SOD-G93A-mice showed a strong expression on somata and their axons especially on cells with morphological signs of degeneration. A specific P2X4-like immunoreactivity in terms of a receptor-upregulation could also be shown on microglia in transgenic tissue. From Western Blot-Analysis resulted the quantitative determination of the P2X4-expression. A strong and increasing expression of P2X4-monomers during the course of disease could be shown. By immunohistochemical analysis of human postmortal slices of the thoracal spinal cord the observations of the mouse-model could be reproduced. We clearly show that the P2X4-upregulation correlates with morphological signs of degeneration during the course of disease in ALS and can be used as a marker for damaged motoneurons. Under functional aspects the upregulation of the P2X4-receptor is probably because of a local extracellular increase of ATP. Our examinations show the upregulation on motoneurons as well as on microglia in ALS-tissue. Chronic microglial n euroinflammation with increased production of neurotoxic substances is well documented in ALS; that is why we must assume that the P2X4-receptor is a important part of the intercellular cross-talk that occurs as a inflammatoric process next to the neurodegeneration in ALS.

Die derzeitigen Kenntnisse zur Pathogenese
der Amyotrophen Lateralsklerose (ALS) stützen sich auf
Untersuchungen von transgenen Mäusen, die Mutationen im humanen
Cu/Zn-Superoxid-Dismutase (mSOD1)-Gen tragen und ein etabliertes
Modell für die familiäre Form der ALS darstellen. Purinerge (ATP-)
Rezeptoren steuern durch intrazelluläre(n) Ca++-Einstrom und
-Freisetzung sowohl Aktivität und Motilität von Mikroglia-Zellen
als auch Ca++ -abhängige Prozesse und das Membranpotential in
Motoneuronen. Die Rolle von Purin-Rezeptoren in der Pathogenese der
ALS ist bisher nicht geklärt. Wir untersuchten
Paraffin-eingebettete Rückenmarksschnitte von Wildtyp- und
SOD1G93A-Mäusen immunhistochemisch auf die Expression des
liganden-gesteuerten P2X4-Purin-Rezeptors zu unterschiedlichen
Krankheits-Zeitpunkten. Eine Western-Blot-Analyse sowie die Messung
und Auswertung der Pixelintensität wurde zur (semi-) quantitativen
Bestimmung der Rezeptorexpression durchgeführt. Zusätzlich wurde
humanes postmortales Rückenmarksgewebe zur Expressionsanalyse des
P2X4-Rezeptors untersucht. In Wildtyp-Mausgewebe fand sich eine
schwache Expression von P2X4-Rezeptoren in Motoneuronen der grauen
Substanz während der postnatalen Entwicklung; das entsprechende
immunhistochemische Signal zeigte sich in der Folge hochreguliert.
In SOD1G93A-transgenen Mäusen zeigte sich im Vergleich zu
Kontrollen eine verstärkte Expression von P2X4-Rezeptoren auf
Motoneuronen, Axonen sowie in der Vorderwurzel des Rückenmarks,
insbesondere auf morphologisch bereits geschädigten Zellen. Eine
Hochregulation auf Mikrogliazellen im Rahmen der Mikrogliose wurde
ebenfalls beobachtet. In der Western-Blot-Analyse fand sich eine
verstärkte Expression von P2X4-Monomeren im Krankheitsverlauf. In
humanem Rückenmarksgewebe von postmortalem Kontroll- und ALS-Gewebe
ließen sich die Befunde der Expressionsanalyse von P2X4 aus dem
Mausmodell reproduzieren. Die Hochregulation von P2X4-Rezeptoren
auf Motoneuronen während des Krankheitsverlaufs korreliert mit
morphologis chen Schädigungszeichen von Motoneuronen und kann als
Marker für geschädigte Motoneurone eingesetzt werden. Unter
funktionellen Aspekten ist die Hochregulation von P2X4
möglicherweise die Folge einer lokalen, extrazellulären
ATP-Erhöhung mit konsekutiver Membrandepolarisation und
intrazellulärem Ca++-Influx und in einen vermutlich vorliegenden
inflammatorischen Aspekt der neuronalen Degeneration i. R. der ALS
involviert.
