The objekt of this work was the detection of genetic polymorphisms which impact the efficacy of gemcitabine and cytarabine, two nucleoside analogues employed as cytostatic drugs in treatment of cancer. As investigational material of this study, isolated human leucocytes of 72 healthy donors and 54 lymphoblastoid cell lines were used and the variability of the cytotoxicity of these two drugs was measured. The projekt aims to contribute to the optimisation and individualisation of the chemotherapy with gemcitabine and cytarabine.

Gegenstand dieser Arbeit ist die
Identifizierung genetischer Polymorphismen, welche die Wirksamkeit
der als Zytostatika eingesetzten Nukleosid-Analoga Gemcitabin und
Cytarabin beeinflussen. Als Untersuchungsmaterial dienten isolierte
Leukozyten von 72 gesunden Spendern sowie 54 lymphoblastoide
Zelllinien, an denen jeweils die Variabilität der Zytotoxizität
dieser beiden Substanzen gemessen wurde. Das Ziel ist die
Optimierung der Chemotherapie mit Gemcitabin und Cytarabin in der
Behandlung von Krebserkrankungen.
