The sarcomeric Z-disc is the main structure of the decentralized model of the muscle mechanotransduction. Proteins, which are anchored to the Z-disc are associated with cardiomyopathy. An example is the muscle LIM protein, MLP. Mutations in MLP lead to dilated cardiomyopathy in humans and mice. To elucidate the underlying molecular mechanisms, MLP interacting proteins have been sought and thereby the Myc interacting zinc finger protein 1 (Miz1) was identified. For this reason the investigation of unexplored Miz1 function in the heart has been set as goal of this work. Genetic screening analysis in this study showed that a substitution mutation in exon 6 of the Miz1 gene (A188V-Miz1) can cause cardiomyopathy. Coimmunoprecipitation and subcellular localization experiments showed localization of Miz1 to the Z-disc, where it interacts with MLP and calcineurin. In an in vitro luciferase assay Miz1 was able to activate the calcineurin promoter. The results presented here suggest for the first time that overexpression of Miz1 in isolated neonatal rat cardiomyocytes leads to an activation of NFATc2 and to cellular hypertrophy. In addition, the overexpression of Miz1 in the same ex vivo models has antiapoptotic effect. The fact that the Miz1 expression is upregulated in heart hypertrophy animal model of aortic constriction (TAC), confirms the hypothesis of the involvement of Miz1 in the formation and the development of cardiomyopathy. For the in vivo Analysis of the Miz1 function, Miz1 cardiac-specific conditional knockout and Miz1 transgenic mice were generated. The analysis of Miz1 conditional knockout animals pointed to a protective effect of Miz1. Indeed Miz1 knockout mice were unable to accomplish compensatory mechanisms four weeks after TAC surgery. It can be concluded that Miz1 is a Z-disc protein whose overexpression leads to cellular hypertrophy and which has, thanks to its anti-apoptotic character, a cardio-protective effect.

Die sarkomerische Z-Scheibe ist die
wichtigste Struktur des dezentralisierten Modells der
Mechanotransduktion. So fand man Kardiomyopathien verursachende
Mutationen in mehreren Genen, die für die Z-Scheibe assoziierte
Zytoskelettproteine kodieren. Ein Beispiel hierfür ist das
Muskel-LIM-Protein MLP. Um die zugrunde liegenden molekularen
Mechanismen aufzuklären, wurden MLP interagierende Proteine gesucht
und dadurch das Myc interagierende Zinkfinger Protein 1 (Miz1)
identifiziert. Daraufhin wurde die Analyse der Miz1-Funktion im
Herzen als Zielsetzung dieser Arbeit festgelegt. Genetische
Screening-Analysen im Rahmen dieser Arbeit zeigten, dass eine
Substitutionsmutation im Exon 6 des Miz1-Gens (A188V-Miz1)
kardiomyopathieauslösend ist. Koimmunopräzipitation und
subzelluläre Lokalisationsexperimente zeigten eine Lokalisierung
von Miz1 an der Z-Scheibe, wo es mit MLP und Calcineurin
interagiert. In einem in vitro Luciferase-Assay konnte Miz1 den
Calcineurin-Promotor aktivieren. Die hier dargestellten Ergebnisse
legen zum ersten Mal nahe, dass die Überexpression von Miz1 in
isolierten neonatalen Ratten-Kardiomyozyten zu einer Aktivierung
von NFATc2 und zur zellulären Hypertrophie führt. Zusätzlich wirkt
die Miz1-Überexpression in den gleichen ex vivo Modellen
antiapoptotisch. Durch die Tatsache, dass im Tiermodell der
Aortenkonstriktion (TAC) die Miz1-Expression hochreguliert ist,
bestätigt sich die Hypothese der Beteiligung von Miz1 an der
Entstehung und an der Entwicklung von Kardiomyopathien. Für die
Untersuchung der in vivo Miz1-Funktion wurden im Laufe dieser
Arbeit herzspezifische konditionale Miz1-Knockout und Miz1
transgene Mäuse generiert. Die Analyse der konditionalen
Miz1-Knockout-Tiere deutete auf einen protektiven Effekt von Miz1
hin: Die Miz1-Knockout-Mäuse waren nicht in der Lage die
kompensatorischen Mechanismen vier Wochen nach einer TAC-Operation
zu leisten. Stattdessen entwickelten diese Tiere direkt eine
Herzinsuffizienz. Zusammenfassend lässt sich feststellen, dass es
sich bei Miz1 um ein Z-Scheibe-Protein handelt, dessen Expression
bei Herzhypertrophie hochreguliert ist, dessen Überexpression zur
zellulären Hypertrophie führt und welches, dank seines
antiapoptotischen Charakters, kardioprotektiv wirkt.
