dc.description.abstractDa vorliegende Studien unserer Arbeitsgruppe auf eine Beteiligung von Connexinen an der oralen Plattenepithelkarzinogenese hindeuten, war es Ziel dieser Arbeit in-vivo am Hamstermodell die Expression der Connexine 26, 43 und 45 im Verlauf der Karzinogenese zu untersuchen. Die durch 9,10-Dimethyl-1,2-Benzanthrazen (DMBA) induzierten Karzinome wurden makroskopisch, histologisch, sowie auf ihren Inflammationsgrad hin untersucht, um die Effektivität des Tiermodells zu prüfen. Weiterhin erfolgte sie Genexpressionsanalyse mittels RNA-Isolation, PCR-Analyse und statistischer Auswertung. Es konnte insgesamt gezeigt werden, dass die Applikation von DMBA in Abhängigkeit von Behandlungsdauer makroskopisch, histologisch und inflammatorisch einen deutlichen karzinogenen Effekt hatte und, dass das histopathologische Grading signifikant mit der Länge der Behandlungsdauer korrelierte. Im Rahmen der Genexpressionsanalyse konnte im Verlauf der DMBA-Behandlung für Connexin 26 und 45 eine signifikante Überexpression beschrieben werden. Connexin 43 zeigte sich als nicht differentiell exprimiert. Ein Zusammenhang zwischen histologischem Grading und Genexpression war nicht nachvollziehbar.de

dc.description.abstractengExisting studies by our study group have indicated connexins to be involved in oral carcinogenesis of the squamous cell carcinoma, so the objective of this trail was to investigate the gene expression of connexin 26, 43 and 45 in the course of carcinogenesis by using hamsters for an in-vivo study design. By applying 9,10-Dimethyl-1,2-Benzanthrazen (DMBA) into the hamster pouches, we were able to induce carcinomas that could be examined macroscopically, histologically, as also the scale of inflammation to evaluate the effectiveness of this technique. Furthermore we analyzed gene expression by performing RNA isolation, PCR and applying statistical analysis. Overall we were able to demonstrate a distinct carcinogenic effect of DMBA application in accord with the length of treatment by inspecting the macroscopic changes, the histology and inflammation. We found significant correlation between the histopathological grading and duration of treatment. Considering gene expression, we detected a significant increase in gene regulation of connexin 26 and 45. Connexin 43 showed no changes in gene regulation.
