dc.description.abstractengThe occurrence of intracytoplasmic membranes (ICM) in Gluconobacter oxydans was first described by Claus et al. in 1975. The presence of the ICM was observed during the early stationary growth phase. It was proposed, that an increased turnover of the used substrate glycerol at the same time is related to the presence of the ICM. Concerning the possible use of the effect for the biotechnological application, these issues are examined in this dissertation by using Western-Blot analysis, the immunogold-labeling technique for the localization of membrane-bound enzymes via transmission electron microscopy (TEM) and the determination of the specific activity of membrane bound, pyrroloquinolin quinine (PQQ) dependent dehydrogenases. Overall, the obtained data indicates that there is no clear connection between the occurrence of the ICM and the specific activity of membrane bound, PQQ dependent dehydrogenases and therefore it seems unlikely that the ICM could be useful in the improvement of the biotechnological application of Gluconobacter oxydans. In the end however, further investigations are proposed to obtain additional insides into the ICM and the possibility of using them as a platform for membrane-bound dehydrogenases in Gluconobacter oxydans.de
