dc.description.abstractDie regulierte Exozytose wird in Neuronen und neuroendokrinen Zellen durch die SNARE-Proteine Synaptobrevin/VAMP, SNAP-25 and Syntaxin1 vermittelt. Während der Fusion bilden diese SNARE-Proteine einen stabilen Komplex, der anschließend in einer konzertierten Aktion von α-SNAP und der AAA-ATPase NSF dissoziiert wird. In dieser Arbeit wurde ein zellfreier Exozytose-Assay entwickelt.Auf beschichteten Objektträgern wachsende PC12-Zellen wurden durch einen Ultraschallpuls in einer kleinen Kammer direkt auf dem Objekttisch eines Fluoreszenzmikroskops aufgeschlossen. Durch Ca2+ konnten eGFP-beladenen Vesikel der so hergestellten "membrane lawns" zur Exozytose und damit zur Ausschüttung ihres Inhalts gebracht werden. Durch den entwickelten Assay konnte die Zeitspanne zwischen dem Zellaufschluß und der vorgenommen Exozytose-Messung verringert und damit der biochemische "run-down" der Membranen reduziert werden.Durch diese Methode konnte eine Vielzahl verschiedener Faktoren auf frisch hergestellte in Ihren Eigenschaften intakten Membranen stark ähnelnden "membrane lawns" getestet und dadurch die späten Schritte der Exozytose für Untersuchungen zugänglich gemacht werden. Die Cytosol- und ATP-Mg-Abhängigkeit dieses Assays wurde addresiert. Übereinstimmend mit vorhergehenden Beobachtungen an permeabilisierten Zellen ist ATP-Mg von entscheidender Wichtigkeit für die Fähigkeit des Systems, auf eine Erhöhung der Ca2+-Konzentration zu reagieren. Im Gegensatz zur Ausschüttung von Noradrenalin in permeablisierten PC12-Zellen allerdings, ist das in dieser Arbeit beschriebene System nicht abhängig von Cytosol.In der folgenden Studie konnte mit dem beschriebenen zellfreien Assay gezeigt werden, dass die Ca2+-abhängige Exzytose von bereits "gedockten" sekretorischen Granulen unabhängig von NSF stattfindet, jedoch durch die Zugabe von rekombinatem α-SNAP stark inhibiert wird. Desweiteren wurde festgestellt, dass diese Inhibition mit der Bindung von α-SNAP an kleine mit Syntaxin 1 co-lokalierende "cluster" auf der cytosolischen Seite der Plasmamembran einhergeht. Durch Inkubation mit Botulinum Neurotoxin C1, einer Protease die spezifisch Syntaxin 1 schneidet, konnte diese Bindung unterbunden werden. Sowohl die Bindung an die Membran, als auch die Inhibition der Exozytose wurden durch die Gegenwart von NSF verhindert. Für eine Mutante von α-SNAP, die NSF nicht mehr aktiveren kann, war dies nicht der Fall. Wir können daher folgern, dass α-SNAP die Exozytose dadurch inhibiert, dass es die Interaktion von Syntaxin mit seinen SNARE-Partner verhindert, ein Befund, der eine unerwartet Role von α-SNAP in der Exozytose enthüllt.de

dc.description.abstractengRegulated exocytosis in neurons and neuroendocrine cells is mediated by the SNARE proteins synaptobrevin/VAMP, SNAP-25, and syntaxin 1. During fusion, these SNAREs form a stable complex that is disassembled by the concerted action of α-SNAP and the AAA-ATPase NSF.During this work I have developed a cell-free assay for exocytosis. Ultrasonic disruption of PC12 cells grown on coated coverslips was performed on the microscope stage in a small chamber and results in the generation membrane lawns that could be triggered with Ca2+ to exocytose their eGFP content. Reduction of the delay between the loss of cell integrity and the measurement of exocytosis reduced biochemical run-down on membranes. With this assay it was possible to access and to probe the late steps in the exocytotic pathway by applying a wide range of various factors onto freshly made lawns that had resembled intact membranes.In these studies I addressed the questions of cytosol and ATP-Mg-dependence in this assay and could conclude that in agreement with preceding observations on permeabilized cells, ATP-Mg was vitally required to enable the system to respond to Ca2+-trigger. However, the system described in this thesis was not dependent on cytosol what made it different from noradrenaline release in permeabilized PC12 cells. In the following study, I have used the above cell-free assay to show that Ca2+-dependent exocytosis of pre- docked secretory granules was independent of NSF but it was potently inhibited by addition of recombinant α-SNAP. Furthermore, I found that inhibition correlated with binding of α-SNAP to small clusters on the inside of the plasma membrane, which co-localized with syntaxin 1. Binding was prevented by incubation with botulinum neurotoxin C1, a protease selectively cleaving syntaxin 1. Both membrane binding and inhibition of exocytosis were abolished in the presence of NSF but not when an α-SNAP mutant defective in NSF-activation was used. I could conclude that α-SNAP inhibits exocytosis by preventing the interaction of syntaxin with its SNARE partners, revealing an unexpected site of action for α-SNAP in the exocytotic pathway.de
