dc.description.abstractBei an Amyotrophische Lateralsklerose (ALS) erkrankten Patienten werden bestimmte Motoneurongruppen in Hirnstamm und Rückenmark teilweise beschädigt. Dies wird mit einem gestörten Kalzium-Metabolismus und mitochondrialen Schäden in Verbindung gebracht. Im Rahmen der vorliegenden Arbeit werden diese Pathomechanismen mithilfe von patch-clamp Messungen und Calcium imaging untersucht, dies an selektiv vulnerablen und resistenten Motoneuronen in Scheiben des Maushirnstamms. Bei facial motoneurons (FMNs), die vulnerabel gegen ALS sind, bewirkten Störungen des mitochondrialen elektrochemischen Potentials durch bath application des mitochondrialen uncoupler FCCP (Carbonyl cyanide 4-trifluromethoxyphenylhydrazone) eine signifikante Verzögerung der zytosolischen Kalziumentfernung sowie eine substantielle Kalziumfreisetzung aus Mitochondrien. Diese Ergebnisse ähnelten den Beobachtungen von vulnerablen hypoglossalen Motoneuronen (HMNs), unterschieden sich jedoch signifikant von Untersuchungen, die an selektiv resistenten okulomotorischen Neuronen (OMNs) und trochlearen Motoneuronen (TMNs) durchgeführt wurden. Sowohl OMNs als auch TMNs zeigten nur eine schwache Kalziumfreisetzung nach FCCP-Anwendung, wobei die höchste Amplitude um ein Vierfaches niedriger lag als bei FMNs und HMNs gemessenen Werten. FCCP hatte auch keinen signifikanten Einfluss auf die zytosolische Kalziumentfernung in TMNs und OMNs. Eine ausführliche Untersuchung der FMNs und TMNs offenbarte deren unterschiedliche Pufferkapazität von Kalzium, wobei FMNs den niedrigen Wert von 46, TMNs jedoch einen Wert von 392 hatten. Weitere Messungen des Kalziummetabolismus von facial motoneurons bewiesen, dass sich die niedrige Kalzium-Pufferkapazität von facial motoneurons in den allerersten Tagen der postnatalen Entwicklung entwickelt. Dementsprechend zeigten facial motoneurons von P0-Tieren den relativ hohen Wert von 132 bei der Kalzium-Pufferkapazität. Im Gegensatz hierzu hatten die FMNs von Tieren in P4 und P10 den wesentlich niedrigeren Ks von 46.Im Ganzen weisen diese Untersuchungen darauf hin, dass ALS-vulnerable Motoneuronen durch eine niedrige Kalzium-Pufferkapazität charakterisiert sind. Dies bedingt eine enge Interaktion von Mitochondrien und intrazellulärem Kalzium, die eine Verbindung der metabolischen und elektrischen Aktivitäten optimiert. Unter pathophysiologischen Bedingungen führt diese Spezialisierung zu einem Anstieg der zellulären Energie und beschleunigt die Verschlechterung des schon geschwächten Metabolismus.de

dc.description.abstractengDefined motoneuron populations in the brain stem and spinal cord are selectively damaged during Amyotrophic Lateral Sclerosis (ALS), which has been linked to perturbed Ca2+ metabolism and mitochondrial damage. In this dissertation, the pathomechanisms are studied by performing patch-clamp recordings and Calcium imaging in selectively vulnerable and resistant motoneurons in slice preparations from mouse brainstem. In the case of facial motoneurons (FMNs), which are vulnerable in ALS, disruption of the mitochondrial electrochemical potential by bath application of the mitochondrial uncoupler FCCP (Carbonyl cyanide 4-trifluromethoxyphenylhydrazone) provoked a significant retardation of cytosolic calcium clearance rates and substantial Ca2+ release from mitochondrial stores. These results were similar to the observations from vulnerable hypoglossal motoneurons (HMNs), but were significantly different from observations made in selectively resistant oculomotor neurons (OMNs) and trochlear motoneurons (TMNs). Both the OMNs and TMNs only displayed minor Ca2+ release after FCCP application, where peak amplitudes were ~4-fold smaller compared to those in the FMNs & HMNs. FCCP also did not significantly affect cytosolic Ca2+ clearance rates in TMNs and OMNs. A detailed investigation on the FMNs and TMNs revealed their contrasting Ca2+ buffering capacities, where FMNs showed a low value of 46 but TMNs had a value of 392. Further study of the calcium metabolism of facial motoneurons revealed that the low Ca2+ buffering property of facial motoneurons is achieved during the very first few days of post natal development. Accordingly, in P0 animals, facial motoneurons displayed a relatively high calcium buffering capacity value of 132. In contrast, FMNs in animals at P4 & P10 displayed substantially lower Ks of 46.Taken together, these observations suggest that ALS vulnerable motoneurons are characterized by a low Ca2+ buffering capacity, leading to an intimate interaction of mitochondria and [Ca2+]i that optimizes the coupling of metabolic and electrical activity. Under pathophysiological conditions, this specialisation creates an augmentation in cellular energy and accelerates the deterioration of an already weakened metabolic system.de
