As a natural product, natural rubber (cis-1,4-polyisoprene) is subjected to a continuous biological process of degradation in the biosphere. For this reason it is astonishing that the exact biological and chemical mechanisms of degradation are still unknown. In this thesis natural rubber degradation was examined by two different Streptomyces species as well as an organism already described as Xanthomonas sp. 35Y. Morphological, physiological and molecular biological analyses revealed, that the natural rubber degrading organism known as Xanthomonas sp. 35Y in fact produces a yellow xanthomonadin-like pigment, characteristically of xanthomonads, but does not actually belong to the genus Xanthomonas. For this reason the classification of this bacterium to the new genus Pseudoxanthomonas is proposed. Proteinbiochemical investigations lead to the identification of a 65 kDa protein, which Pseudoxanthomonas sp. 35Y specifically produces in high concentrations during growth on natural rubber.Other Languages
Als Naturprodukt unterliegt Naturkautschuk (cis-1,4-Polyisopren) in der Biosphäre einem langsamen aber kontinuierlichen biologischen Abbauprozeß. Daher ist es auch erstaunlich, daß die genauen biologischen und chemischen Vorgänge des mikrobiellen Naturkautschuk-Abbaus immer noch weitgehend unbekannt sind. Im Rahmen dieser Arbeit wurde der NR-Abbau durch zwei Streptomyces-Stämme sowie einem als Xanthomonas sp. beschriebenen Organismus eingehender untersucht. Morphologische, physiologische wie auch molekularbiologische Analysen haben jedoch gezeigt, dass der als Xanthomonas sp. beschriebene NR-Abbauer zwar ein für Xanthomonaden charakteristisches gelbes Xanthomonadin-ähnliches Pigment bildet, jedoch keinesfalls zur Gattung Xanthomonas gehört. Daher wurde die Einordnung dieses Organismus in die neue Gattung Pseudoxanthomonas vorgeschlagen. Proteinbiochemische Untersuchungen haben zur Identifizierung eines 65 kDa-Proteins geführt, das Pseudoxanthomonas sp. 35Y spezifisch beim Wachstum auf NR- oder DPNR-Latex in hoher Konzentration bildet.
