To generate a stable respiratory activity from birth to death is one of the most important functions of the brain stem. In the adult brain stem g-aminoacid(GABA)-mediated inhibition is essential for this function, however it is controversial whether and to what extend GABAergic inhibition is necessary to produce a respiratory rhythm in neonatal animals. This study investigates the developmental changes of the GABAergic system and their influence on respiratory activity using the respiratory active brain stem preparation of NMRI mice. The results suggest an important role of GABAergic inhibition at all tested ages, mainly mediated via GABAB receptors during the first three postnatal days. During this period of time, the intracellular chloride-concentration decreases from ca 100 mM at P0 to 4-7 mM at P4 and later. Only after it has reached its final level, GABAA receptor mediated chloride currents gain an increasing influence on the respiratory activity. This switch in the dominance of GABAB to GABAA receptor-mediated inhibition is accompanied by an intracellular redistribution of GABAB receptors. Until P4, GABAB receptor immunoreactivity is distributed diffusely in somata and dendrites. In older animals, an increasing number of additional immunopositive granules become visible in the cytoplasm. The granules are GABAB receptor-containing vesicles as revealed by electron microscopy. They were not only detected in the brain stem but in neurons throughout the central nervous system. The time course of their appearance reflects the delayed maturation of higher brain centres (brain stem P5, hippocampus P7, cortex P8). GABAB receptors colocalise with the transcription factor ATF4 in 5-10% of these vesicles. Using co-immunoprecipitation, the strength of interaction between the GABAB receptor subunits R1 and R2 and ATF4 reveals distinct developmental time courses. The results support the hypothesis that non-functional chloride-mediated inhibition in neonatal animals is compensated by cation-mediated inhibition. Furthermore, the direct interaction of the GABAB receptor with a transcription factor suggests a novel mechanism in signal transduction and transport of GABAB receptors.

Die Generierung einer lebenslang-stabilen respiratorischen Aktivität gehört zu einer der wichtigsten Hirnstammfunktionen. Um diese zu gewährleisten ist im adulten Hirnstamm die g-Aminobuttersäure(GABA)-vermittelte Inhibition von essentieller Bedeutung. Es ist bislang nicht eindeutig geklärt, in wie weit diese Inhibition im frühpostnatalen Hirnstamm eine funktionelle Rolle spielt. Die vorliegende Arbeit untersucht anhand des respiratorischen Hirnstammpräparates der NMRI Maus die entwicklungsbedingten Veränderungen des GABAergen Sytems und deren Einfluß auf die respiratorische Aktivität. Die Daten deutet darauf hin, daß GABAerge Inhibition unmittelbar nach der Geburt eine wichtige Rolle spielt, welche in den ersten postnatalen Tagen (P0-P3) weitgehend von GABAB-Rezeptoren ausgeübt wird. Die GABAA-Rezeptor-vermittelte Inhibition spielt erst nach P4 eine funktionelle Rolle, wenn die intrazelluläre Chloridkonzentration von ca. 100 mM an P0 auf das adulte Nieveau (4-7 mM) gesunken ist. Diese funktionelle Verschiebung der frühpostnatalen GABAB-Dominanz zu einer adulten GABAA-Dominanz ist begleitet von einer intrazellulären Umverteilung des GABAB-Rezeptors. Seine Immunoreaktivität ist bis P4 in Soma und Dendriten diffus verteilt, während ab P5 in zunehmender Anzahl intensiv immunopositiver Granula im Zellplasma auftreten. Diese erwiesen sich elektronenmikroskopisch als GABAB-Rezeptor-haltige Vesikel. Sie sind außer im Hirnstamm in vielen weiteren Hirnregionen zu finden. Der Zeitpunkt, ab welchem sie auftreten, spiegelt weitgehend die ontogenetische Reife der Hirnzentren wieder (Hirnstamm P5, Hippocampus P7, Cortex P8). In 5-10% dieser Vesikel sind die GABAB-Rezeptoren mit dem aktivierenden Transkriptionsfaktor ATF4 kolokalisiert. Während GABAB R1 und ATF4 ihre stärkste Interaktion zwischen P0 und P3 aufweisen, erreicht die Interaktionstärke von R2 und ATF4 ihr Maximum zwischen P8 und P15. Die hier dargestellten Ergebnisse unterstützen die Hypothese, daß die perinatale Absenz der funktionellen Chlorid-getragenen Inhibition durch funktionelle Kationen-getragene Inhibition kompensiert werden kann. Darüber hinaus deutet die direkte Interaktion des GABAB-Rezeptors mit ATF4 auf einen neuen Mechanismus in der Signal-Transduktion und im Transport von GABAB-Rezeptoren hin.
