Mechanisms of proton translocation in Methanosarcina mazei Gö1

dc.description.abstractDie Umsetzung von Methanol und Wasserstoff plus Kohlendioxid im Zuge der Methanogenese ist an den Aufbau eines elektrochemischen Ionenpotentials über die Cytoplasmamembran gekoppelt. Dieser Ionengradient dient der ATP-Bildung aus ADP und Pi durch ATP Synthasen.In dieser Arbeit konnten drei unterschiedliche protonentranslozierende Enzymsysteme des methanogenen Archaeons Methanosarcina mazei Gö1 analysiert werden: Die F420H2:Heterodisulfid-Oxidoreduktase, die H2:Heterodisulfid-Oxidoreduktase sowie eine protonentranslozierende Pyrophosphatase.Es konnte gezeigt werden, dass 2-Hydroxyphenazin den Elektronentransport zwischen den Schlüsselenzymen der H2:Heterodisulfid-Oxidoreduktase und der F420H2:Heterodisulfid Oxidoreduktase vermitteln konnte. Dieser Elektronenüberträger ist ein wasserlösliches Analogon des physiologischen Cofaktors Methanophenazin. Versuche mit invertierten Membranvesikeln des Organismus bewiesen, dass der Elektronentransport an eine Translokation von Protonen über die Cytoplasmamembran gekoppelt ist. Das maximale Verhältnis von Protonen zu Elektronen war dabei 4:0 für jede der beiden Elektronentransportketten. Der so aufgebaute Protonengradient diente zur ATP Regeneration aus ADP und Pi durch ATP Synthasen.Es wurde nachgewiesen, dass die untersuchten Phenazin-abhängigen Reaktionen duch Diphenylen Idodonium chlorid (DPI) spezifisch gehemmt werden konnten. Verschieden Experimente führten zu der Hypothese, dass DPI duch seine strukturelle Ähnlichkeit zum Phenazin-Grundgerüst als kompetitiver Hemmstoff wirkte.Die Sequenzdatenanalyse des Ms. mazei Gö1 Genomprojektes ergab unter anderem die Existenz eines 12 Gene umfassenden Genclusters fpo (für F420H2:Phenazin Oxidoreduktase). Es zeigte sich, dass die Gene in einem Operon organisiert sind. Elf dieser ORFs zeigen Homologien zur bakteriellen NDH-1 sowie zum mitochondriellen Komplex I (NADH:Ubichinon Oxidoreduktase) der Atmungskette. Dabei existierte in Ms. mazei kein NADH-Dehydrogenase-Modul, anstatt dessen wurde mit der F420H2 Dehydrogenase ein Polypeptid FpoF mitaufgereinigt, dass Ähnlichkeiten zu archaeellen F420-abhängigen Hydrogenasen zeigte. Dieses Ergebnis legte den Schluss nahe, das dieses FpoF Protein der Dehydrogenase als alternatives Input Modul zur Anpassung an den zentralen Elektronencarrier F420 dient. Dabei wurde das korrespondierende Gen fpoF nicht als Bestandteil des Operons identifiziert, sondern es stellte sich heraus, dass es auf einem anderen Teil des Chromosoms lokalisiert ist.Es wurde weiterhin über die Struktur und Synthese von Methanophenazin berichtet. Es konnte nachgewiesen werden, dass dieser Cofaktor den Elektronetransport zwischen den Schlüsselenzymen der obengenannten Elektronentransportketten von Ms. mazei vermittelt. Es zeigte sich, dass diese Substanz nicht nur das erste Phenazinderivat war, dass eine physiologische Funktion als Elektronenüberträger besaß, sondern es ist auch das erste Beispiel für Phenazine in Archaea überhaupt.Bioinformatische Untersuchungen des Ms. mazei Genoms wiesen die Existenz zweier protonentranslozierender Pyrophosphatasen nach. Die korrespondierenden Polypeptide wurden als MVP1 und MVP2 (Methanosarcina vacuolar type Pyrophosphatase) bezeichnet. Expressionsanalysen mittels Northern Blots ergaben, dass in methylothroph angezogenen Zellen in der logarithmischen Phase nur MVP2 exprimiert wurde. Gewaschene Membranen dieser Wachstumsphase zeigten eine spezifische Pyrophosphatase-Aktivität von 0,34 U pro mg Protein und Versuche mit invertierten Membranvesikeln ergaben, dass die Hydrolyse von einem Molekül Pyrophosphat die Translokation von einem Proton über die Cytoplasmamembran treibt. Dies lässt des Schluss zu, das die Aktivität dieses Enzyms zu Enegiekonservierungsprozessen in Ms. mazei beiträgt.de

dc.description.abstractengMethanogenesis from methanol and H2 plus CO2 is coupled to the generation of a transmembrane ion gradient, which is the driving force for ATP formation from ADP plus Pi.In this work three proton-translocating enzyme systems from the methanogenic archaeon Methanosarcina mazei Gö1 were analyzed: The F420H2:heterodisulfide oxidoreductase, the H2:heterodisulfide oxidoreductase and a proton-translocating pyrophosphatase.It was shown, that 2-hydroxy phenazine was able to mediate the electron transport between the key enzymes of the H2:heterodisulfide and the F420H2:heterodisulfide oxidoreductase. 2-hydroxy phenazine is a water soluble analogon of the physiological electron carrier, methanophenazine. Washed inverted vesicles of this organism were able to couple the electron transfer of both electron transport chains with the transfer of protons across the cytoplasmic membrane The maximal H+/2e- was found to be 4.0 for each electron transport chain. The electrochemical proton gradient thereby generated was used for ATP synthesis by an A- type ATP synthase.It was found, that 2-OH phenazine-dependent reactions were inhibited by diphenyliodoniumchloride (DPI). Several experiments lead to the hypothesis that DPI reacts as a competitive inhibitor due to its structural similarity to 2-OH phenazine.Sequence data from the genome sequencing project of Ms. mazei Gö1 revealed the existence of a gene cluster comprising 12 genes referred to as fpo (F420H2:phenazine oxidoreductase) organized in one operon. For eleven proteins homologues exist in the bacterial NDH-1 and the mitchondrial Complex I (NADH:ubiquinone oxidoreductase). A NADH dehydrogenase module does not exist in Ms. mazei, Gö1, instead a polypeptide with high homologies to Archaeal F420 dependent hydrogenases was copurified with the F420H2 dehydrogenase leading to the assumption that this protein serves the dehydrogenase as an F420H2 oxidizing input module. The corresponding gene to this subunit, fpoF was not located in the operon, but on a different part of the chromosome.The structure and chemical synthesis of methanophenazine, the first phenazine from Archaea, was reported. Experiments on the function of the new cofactor in Ms. mazei Gö1 demonstrated that it is also the first phenazine whatsoever involved in the membrane bound electron transport in biological systems. Furthermore, it was shown that methanophenazine is able to mediate the electron transport between the membrane-bound enzymes of the electron transport chains mentioned above.Further bioinformatical studies showed the existence of two open reading frames in the Ms. mazei genome encoding proton translocating pyrophosphatases. The corresponding polypeptides were referred to as Mvp1 and Mvp2. Northern blot analysis using RNA from mid growth phase harvested methanol grown cells revealed that only Mvp2 was produced under these conditions. Washed membranes showed a specific pyrophosphatase activity of 0.34 U per mg membrane protein and inverted vesicles were found to couple a translocation of one proton to the hydrolysis of one molecule of pyrophosphate. These findings indicate that this type of enzyme might contibute to the energy conservation processes in Ms. mazei Gö1.de
