dc.description.abstract | Die Aufnahme von Nitrat als wichtigste Stickstoffquelle der meisten Pflanzen erfolgt durch konstitutive und induzierbare Nitrat-Transportsysteme in der Plasmamembran (PM). Während auf molekulargenetischer Ebene Nitrat-Transporter codierende Gene eingehend untersucht sind, sind die Kenntnisse über die eigentlichen, die Transportsysteme konstituierenden Proteine sehr begrenzt. Dies gilt auch für weitere Plasmamembran-Proteine, die ebenfalls durch Nitrat induzierbar sind. Aus der einzelligen Grünalge Chlorella saccharophila als Modellsystem für niedere Pflanzen sollten Nitrat-Transporter und weitere Nitrat-induzierbare Proteine der Plasmamembran aufgereinigt und charakterisiert werden. Dazu wurden Verfahren etabliert zur Isolierung und Auftrennung der PM-Proteine nach Hydrophobizitätseigenschaften und ihrer anschließenden zweidimensionalen Gelelektrophorese. Basierend darauf wurden durch in vivo-Markierung mit 35S-Methionin, Photoaffinitätsmarkierung mit einem 14C-markierten Nitrat-Analogon und immunologische Nachweise (Western Blot) Nitrat-induzierte PM-Proteine isoliert, teilweise ansequenziert und charakterisiert. Nach RT-PCR einer mit Nitrat-induzierten Transkripten angereicherten mRNA wurde eine PCR mit degenerierten Primern durchgeführt und amplifizierte Fragmente ebenfalls ansequenziert. | de |

dc.description.abstracteng | The uptake of nitrate as the most important nitrogen source for higher and lower plants is mediated through constitutive and inducible nitrate-transport systems in the plasma membrane (PM). While the molecular genetics of nitrate transporters are well examined, knowledge about the actual proteins constituting nitrate-transport systems and further plasma membrane proteins that are also inducible by nitrate is very limited. Using the unicellular green alga Chlorella saccharophila as a model system for lower plants, object of this thesis was to characterize nitrate transporters and further nitrate-inducible PM proteins. Methods were established for isolation and separation of PM proteins according to their hydrophobicity, followed by subsequent two-dimensional gel electrophoresis. Based on that different approaches were made to identify nitrate-induced PM proteins by in-vivo labelling with 35S-methionine, photo-affinity labelling with a 14C-radiolabelled nitrate analogon and immunodetection (Western blot). Few nitrate-induced PM proteins were isolated, partly sequenced and characterized by data base comparison.
