dc.description.abstract | In dieser Arbeit wurde die Funktion des bZIP-Transkriptionsfaktors BZI-1 aus Nicotiana tabacum untersucht. Aufgrund der Aminosäuresequenz, der Konservierung von Proteindomänen und der genomischen Exon-Intron Struktur kann BZI-1 einer Gruppe homologer bZIP-Transkriptionsfaktoren zugeordnet werden, zu denen sowohl Vertreter in monokotylen wie auch in dikotylen Pflanzen zu finden sind. Für BZI-1 konnte ein Aktivierungpotential in planta nachgewiesen werden, welches durch die 72 Aminosäuren der Domäne N vermittelt wird. Da BZI-1 in vitro an Promotorelemente der Chalcon-Synthase (CHS) und auch in vivo an G- und C-Box cis-Elemente bindet, wurde eine Regulation von Genen des Phenylpropanstoffwechsels wie CHS und PAL (Phenylalanin-Ammonium-Lyase) postuliert. Durch Analysen in transgenen Tabakpflanzen, in denen die Menge an funktionellem BZI-1 sowie die BZI-1-Aktivierungseigenschaften verändert waren, konnte jedoch nachgewiesen werden, daß CHS und PAL keine in vivo Zielgene von BZI-1 sind. Vielmehr konnte gezeigt werden, daß BZI-1 an der Vermittlung der Auxinantwort in der Pflanze beteiligt ist. Transgene Tabakpflanzen, die ein dominant negativ wirkendes BZI-1-Derivat exprimieren, dem die N-terminale Aktivierungsdomäne fehlt (BZI-1-DN), zeigen einen auxininsensitiven Phänotyp. BZI-1-DN-Pflanzen wachsen gestaucht und bilden vermehrt Seitensprosse. Auch auf molekularer Ebene wird die Fähigkeit, auf exogenes Auxin zu reagieren, von BZI-1 reguliert. So konnte GH3 als mögliches BZI-1-Zielgen identifiziert werden, da seine auxinabhängige Expression in BZI-1-DN-Pflanzen reduziert und in VP16-BZI-1-Pflanzen, die ein Fusionsgen aus der VP16-Aktivierungsdomäne und BZI-1 exprimieren, in Abhängigkeit von Auxin verstärkt wird. | de |

dc.description.abstracteng | The tobacco (Nicotiana tabacum) bZIP protein BZI-1 exhibits all typical characteristics of a transcription factor. BZI-1 binds to G-box and C-box cis-elements in vivo, and the N-terminus of BZI-1 functions as an activation domain in plant cells. With reference to amino acid sequence, conservation of protein domains, genomic exon-intron structure and expression pattern, BZI-1 is closely related to CPRF2, OHP1/2, BLZ1 or REB, a group of bZIP proteins which have been described in a number of dicot and monocot species. In order to analyse the function of BZI-1 in detail, transgenic tobacco plants ectopically overexpressing various derivatives of BZI-1 were engineered. Since BZI-1-related dicot transcription factors have been isolated by in vitro binding to chalcone synthase (CHS) G-box promoter elements, it has been suggested that phenylpropanoid pathway genes, such as CHS and PAL (phenylalanine ammonia-lyase), are in vivo target genes. However, no changes in CHS or PAL expression were observed in transgenic plants expressing increased levels of BZI-1 or a dominant negative BZI-1-DN protein. Plants expressing BZI-1-DN protein develop various phenotypical changes which implicate an effect of BZI-1 on plant hormone response. Auxin-induced root- or callus-development is strongly inhibited in BZI-1-DN plants whereas cytokinin mediated shoot-induction is unaffected. In BZI-1-DN plants the induction of the auxin-inducible genes PARA, PARC, and GH3 is reduced and delayed. Hence, BZI-1 is involved in plant development by regulating the response to auxin-mediated hormone signalling in planta. | de |
