The BRUNO-like proteins are family members of the RNA Binding proteins. These proteins play an important role in development, cell differentiation and regulation of the nervous system, likewise for the mRNA transport, mRNA localization and mRNA translation. A characteristical common structure are three RNA-Recognition-Motifs (RRM), which represent the core of a functional RNA-Binding-Domain (RBD). In this paper a new member of the Bruno-like gene family, the mouse Metr-1 gene was isolated and characterized. The Metr-1 transcript contains 2843 nucleotides and codes for a 481 amino acids protein METR-1 which shows these characteristic three RRM. In nothern blot analysis on the one hand we detected in murine brain mRNA two Metr-1 transcripts, 2.8 kb and 3.5 kb, and on the other hand a 3.0 kb transcript in testis mRNA. Expression analysis performed with the aid of RT-PCR showed at least a week Metr-1 expression in 9.5 days old mouse embryos. Additionaly we demonstrated that the gene is also expressed in pachytene-spermatocytes. By the whole mount in situ technique we pointed out that possibly Metr-1 plays a role in the embryonic development, differentiation and dorso-ventral orientation of the embryonal nervous system. We demonstrated that Metr-1 is expressed in 10.5 and 11.5 days p.c. old mice embryos in the dorsal regions of the telencephalon, mesencephalon, diencephalon, myencephalon, neuro-epithelium, neural tube and the spinal ganglions. In order to determine the cell specific expression in adult mice brains, using in situ hybridisation technique, we detected an expression exclusively in the cytoplasma of Purkinje cells of the cerebellum. Using the genomic southern blotting method we confirmed that Metr-1 is a single copy gene. We sequenced the whole coding region of an isolated genomic Metr-1 gene and showed at least that the gene is 18 kb large, besides we identified and characterized 13 exons.

Die Proteine der BRUNO-like Familie gehören der Familie der RNA-Bindungsproteine an. Diese Proteine spielen eine wichtige Rolle in der Entwicklung, Zelldifferenzierung und Regulation des Nervensystems, aber auch für den mRNA-Transport, die mRNA-Lokalisation und die mRNA-Translation sind sie von großer Bedeutung. Charakteristisch für die Struktur dieser Proteine sind drei RNA-Recognition-Motifs (RRM), die eine Kernstruktur einer funktionellen RNA-Bindungs-Domäne (RBD) repräsentieren. Im Rahmen dieser Arbeit wurde ein neuer Vertreter der Bruno-like Gen Familie, Metr-1 der Maus, isoliert und anschließend charakterisiert. Metr-1 ist ein 2843 Nukleotide großes Transkript mit einem offenen Leserahmen für ein 481 Aminosäuren kodierendes Protein METR-1, das die charakteristischen drei RRM Motive aufweist. Durch Northern Blot Analysen konnten man Transkripte einerseits in der Gehirn mRNA mit einer Größe von 2,8 kb sowie 3,5 kb und andererseits ein 3.0 kb großes Transkript in der Testis RNA nachweisen. Expressions-Analysen mit Hilfe der RT-PCR Technik zeigten, daß Metr-1 schon in der RNA 9,5 Tage p.c. alter Maus-Embryonen schwach zu detektieren ist. Außerdem konnte eine Expression in den Keimzellen, in den Pachytän-Spermatozyten, nachgewiesen werden. In whole mount in situ"- Analysen wurde gezeigt, daß Metr-1 wahrscheinlich in der embryonalen Entwicklung, der Differenzierung und der dorsoventralen Orientierung des embryonalen Nervensystems eine Rolle spielt. So wurde Metr-1 in den dorsalen Bereichen des Telencephalons, Mesencephalons, Diencephalons, Myencephalons, Neuroephitels, Neuralrohrs und in den Spinalganglien 10,5 und 11,5 Tage p.c. alter Embryonen identifiziert. Durch in situ Hybridisierungs-Analysen von Gehirnschnitte adulter Mäuse wurde eine Metr-1 Expression ausschließlich im Zytoplasma der Purkinje-Zellen des Cerebellums aufgezeigt. Mittels genomischer Southern Blot Analyse wurde nachgewiesen, daß Metr-1 als Einzelkopie im Genom vorliegt. Das Gen der Maus wurde mit Hilfe eine Cosmid Klons charakterisiert; es ist ca. 18 kb groß und weist insgesamt 13 Exone auf.
