dc.description.abstractEinige menschliche neurodegenerative Krankheiten, einschließlich Alzheimer und Parkinson, führen zur Ablagerung von proteinartigen Aggregaten, die Amyloidfibrillen genannt werden. Überraschenderweise aggregieren Proteine mit sehr verschiedenen Aminosäurereihenfolgen und dreidimensionale Strukturen zu Amyloid-Fibrillen, die ähnliche Eigenschaften aufweisen, wie eine ähnliche Morphologie und eine spezifische molekulare -Faltblatt Architektur. Dieser Befund legt nahe, dass die Fähigkeit Fibrillen-artige Strukturen zu formen eine intrinsische Eigenschaft der Polypeptidkette ist und dass die native Struktur nicht notwendigerweise die einzige geordnete Struktur ist, die jedes Protein annehmen kann. Weiterhin treten bei der Aggregation teilweise oder völlig entfaltete Zustände der fehlgefalteten Proteine auf. Während die Faktoren, die zu Aggregation führen, mehr und mehr verstanden werden, sind die strukturellen Charakteristiken von Intermediaten und die molekularen Details, die die Anordnung von fehlgefalteten Proteinen in Amyloidfibrillen führen nur ansatzweise bekannt. Hier in Kapitel 3 wird durch eine Kombination von Lösungs- und Festkörper-NMR-Spektroskopie gezeigt, dass die Struktur von Amyloidfibrillen von -synuclein ( S) direkt mit den Konformations-Eigenschaften des entfalteten Zustandes und des monomeren Zustandes der fehlgefalteten Proteine korreliert. Amyloidfibrillen haben sehr hohe Stabilität, die sie als eine der festesten bekannten biologischen Materialien auszeichnet. Sie können durch hohe Konzentrationen von denaturierenden chemischen Substanzen oder durch Hochdruck getrennt werden. In Kapitel 4 wird gezeigt, dass Amyloidfibrillen, die durch das Protein S geformet werden und als Ablagerungen im Gehirn von Parkinson-Patienten gefunden werden, in hohem Grade empfindlich für niedrige Temperatur sind. Trotz ihrer bemerkenswerten Stabilität bei hohen Temperaturen, dissoziieren sie äußerst schnell in Monomere und kleine Aggregate in schock-tiefgekühlten Wasser bei -15°C ohne einzufrieren. Schließlich befasst sich Kapitel 5 mit dem Verständnis der Mechanismen, welche die Aggregation und Toxizität bei Parkinson and verwandten Krankheitsformen bestimmen, indem NMR mit anderen spektroskopischen Methoden kombiniert wird. Es wird gezeigt, dass die Phosphorylierung an S129 und an S87 die Struktur von S erweitert, die konformationelle Flexibilität von S (wt) erhöht, und in vitro die Bildung von Fibrillen hemmt. Zusätzlich können die Phosphorylierungnachahmer (S129E und S129D) den Effekt von Phosphorylierung auf die strukturellen und Aggregations-Eigenschaften von S in vitro nicht reproduzieren. dc.description.abstractengA number of human neurodegenerative diseases, including Alzheimer"s and Parkinson"s diseases (PD), are associated with the deposition of proteinaceous aggregates known as amyloid fibrils. Surprisingly, proteins with very different amino acid sequences and three-dimensional structures aggregate into amyloid fibrils that share common characteristics, such as a similar morphology and a specific beta-sheet-based molecular architecture. This suggests that the ability to fibrillate is an intrinsic property of a polypeptide chain and that the native structure is not necessarily the only ordered structure that each protein can assume. An additional common property of aggregation into amyloid fibrils is the presence of partially or fully unfolded states of the misfolding proteins. Whereas there is increasing knowledge about the factors that drive aggregation, the structural characteristics of intermediates and the molecular details that determine the arrangement of misfolded prote! ins in amyloid fibrils are still only understood in outline.Here in Chapter 3, it is demonstrated, by a combination of solution-state and solid-state NMR spectroscopy, that the structure of amyloid fibrils of á-synuclein (aS) is directly correlated to the conformational properties of the unfolded state and monomeric state of misfolding proteins. Amyloid fibrils have very high stability placing them among the strongest known biological materials. It can be dissociated by high concentrations of chemical denaturants or by high pressure.In Chapter 4, it is showed that amyloid fibrils formed by the protein aS, which constitute the insoluble aggregates found in brains of patients suffering from PD, are highly sensitive to low temperature. Despite their remarkable stability to high temperature, mature amyloid fibrils of aS are rapidly dissociated into monomers and small aggregates in supercooled water at 258K, without freezing.Finally, Chapter 5 sheds light on understanding the mechanisms that govern aS aggregation and toxicity in PD and related a-synucleinopathies by NMR in combination with other spectroscopic methods. It is demonstrated that the phosphorylation at S129 and S87 expands the structure of aS, increases the conformational flexibility of wt aS and inhibits its fibrillogenesis in vitro. In addition, the phosphorylation mimics (S129E and S129D) do not reproduce the effect of phosphorylation on the structural and aggregation properties of aS in vitro. However, the phosphorylation mimic S87E is able to mimic the effect of phosphorylation on the aggregation properties of aS in vitro.de
