dc.description.abstract | Das erste Projekt beschäftigt sich mit der Untersuchung der Biosynthese des neuen mikrobiellen Metaboliten Spirodionsäure und der Aufklärung der absoluten Konfiguration dieses Naturstoffs. Innerhalb der Studien mit 13C-markierten Vorläufern konnten die Kultivierungsbedingungen durch Variation des Mediums, des Volumens, der Kultivierungsgefäße und der Fermentationszeit optimiert werden. Alle Ergebnisse deuten auf einen biogenetischen Ablauf hin, in dem 6-Ethyl-2H-pyrone und Sarkomycin über eine intermolekulare Diels-Alder-artige Reaktion zu Spirodionsäure reagieren. Für die Aufklärung der absoluten Konfiguration des Spiro[4,5]decen-Gerüsts und seiner drei stereogenen Zentren wurde in einer Kooperation mit dem Arbeitskreis von P. v. Zezschwitz die Totalsynthese von Spirodionsäure durchgeführt. Die Herstellung von Camphersultam-Amiden synthetischer Diastereomere und des Naturstoffs führten zur Aufklärung der absoluten Konfiguration von Spirodionsäure mit der Struktur (1R,5R,9S)-6-Ethyl-7,9-dimethyl-4,8-dioxospiro[4,5]deca-6-en-1-carbonsäure.Das zweite Projekt beschäftigt sich mit der Biosynthese von Kirromycin. Kirromycin ist ein komplexes, lineares Polyketid, welches als Inhibitor der Proteinbiosynthese an den bakteriellen Elongationsfaktor Tu bindet. Die Identifizierung und Analyse des Biosynthese-Gen-Clusters von Kirromycin durch den Arbeitskreis von W. Wohlleben und T. Weber führten zu der Annahme, dass bei der Pyridonring-Bildung PKS- und NRPS-Schritte beteiligt sind. Diese Hypothese wird hier experimentell durch chemisch-analytische Arbeiten mit Deletionsmutanten von Streptomyces collinus Tü 365 und Fütterungsexperimenten mit [U-13C315N]β-Alanin untersucht und zeigt erstmals β-Alanin als Baustein der Pyridonring-Bildung. | de |

dc.description.abstracteng | The first project is about the investigation of the biosynthesis of the novel microbial metabolite spirodionic acid and its absolute stereoconfiguration. Within the scope of these biosynthetic studies with 13C-labeled precursors the cultivation parameters were optimized varying media, volume, vessel and incubation time. All results of the feeding experiments indicate a biogenetic pathway with 6-ethyl-2H-pyrone and sarkomycin as precursors for the formation of spirodionic acid by an intermolecular Diels-Alder-type reaction. To elucidate the structure of the spiro[4,5]decene and the configuration of the three stereogenic centers, the total synthesis of spirodionic acid was envisaged in cooperation with the group of P. v. Zezschwitz. The preparation of camphersultam amides of the synthetic diastereomeres and of the natural product led to the elucidation of the absolute stereoconfiguration by spectroscopic and HPLC analysis and emphasized the structure to be (1R,5R,9S)-6-ethyl-7,9-dimethyl-4,8-dioxospiro[4,5]deca-6-ene-1-carboxylic acid.The second project deals with the biosynthesis of kirromycin. Kirromycin is a complex linear polyketide that acts as a proteinbiosynthesis inhibitor by binding to the bacterial elongation factor Tu. The working group of W. Wohlleben and T. Weber identified the biosynthetic gene cluster, which was isolated from the strain Streptomyces collinus Tü 365. Based on in silico analysis, a route to pyridonering formation involving PKS and NRPS steps was postulated This hypothesis is experimentally proven by chemical-analytical experiments with certain knock-out mutants of the strain Streptomyces collinus Tü 365 and feeding studies with [U-13C315N]β-alanine. The NMR and MS analyses of the isolated pure kirromycin showed β-alanine as a precursor for the pyridonering-formation for the first time. | de |
