dc.description.abstractDas as-1 Element und die TGA Transkriptionsfaktoren, der an das as-1 Element binden kann, vermitteln Salicylsäure- und Auxininduzierbare Genexpression in Pflanzen. Vorhergehende Experimente haben hergestellt, dass as-1 Element vom ASF1 / SARP Proteinkomplexes aus Nicotiana tabacum erkannt werden. Als Komponenten des ASF1 / SARP, konnten die Klasse II TGA Faktoren, TGA2.2 und TGA2.1, identifziert werden. Zur identifizierung potentieller Interaktionspatner von TGA2.2 wurde im Rahmen dieser Arbeit ein modifiziert `Hefe Hybrid System` hergestellt. TGA2.2 interagiert spezifisch in der Hefe und in vitro mit zwei Proteine, At1g50570 und At5g55530, die ein C2 Domäne in ihren Aminosäuresequenz enthalten. At1g50570 interagiert nur mit der Klasse II TGA- Faktoren. At1g50570 kann als Transkriptionsaktivator in der Hefe und BY-2 Protoplasten funktionieren. Bei mikroskopischen Analysen konnte die Fluoreszenz der At1g50570-GFP Fusionsprotein überwiegend im Kernmembran und ER beobachtet werden. Die Expression des At1g50570 Gens werden nach Pathogeninfektion induziert. Überexpression oder Unterdrückung des At1g50570 Gens in Arabidopsis änderte nicht die Expression des PR-1 Gen mit oder ohne SA behandlung.TGA2.2 interagiert spezifisch in der Hefe mit At1g28480 Protein, ein Glutaredoxin, das bekannt, um Disulfid-Brücke zwischen Proteine zu katalysieren. At1g50570 interagiert mit alle TGA- Faktoren aus Tabak. Die Interaktion zwischen At1g28480 und TGA2.2 wurde weder durch oxydierende Stresszustände beinflusst, noch durch Mutation in konservierte Cysteine im TGA2.2 und At1g28480. At1g28480 erhöht indirekt die Dimerization zwischen TGA2.2 Faktoren. Die Interaktion zwischen At1g28480 und TGA2.2 wurde in planta bestätigt. At1g28480 kann über TGA2.2 mit NPR1 in einem `Hefe Drie-Hybrid System` interagieren. Die Expression des At1g28480 Gens werden nach Pathogeninfektion und SA behandlung induziert. Überexpression oder Unterdrückung des At1g28480 Gens in Arabidopsis änderte nicht die Expression des PR-1 Gen mit oder ohne SA behandlung. Überexpression des At1g28480 Gens unterdrükte die Auxininduzierbare einer as-1-Elementgenexpression in Arabidopsis. Diese Ergebnisse zeigen an, daß At1g28480 Protein eine Rolle in den as-1 Elment induzierbare Genexpression in Pflanzen spielen könnte.de

dc.description.abstractengIn plants, the as-1 cis-element and its trans-acting factors, known as TGA transcription factors, play an important role in the transcriptional control of salicylic acid- and auxin-inducible gene expression. Previous experiments have established that TGA2.2 is the major component of the nuclear as-1-binding factor (ASF1) and cellular salicylic acid response protein (SARP) that bind to the as-1 element. Using TGA2.2 as bait in a modified yeast hybrid screen, several cDNA TGA2.2-interacting proteins were isolated.TGA2.2 interacts specifically in yeast and in vitro with two closely related proteins, At1g50570 and At5g55530 that contain a C2 domain in their amino acid sequences. At1g50570 interacts with class-II members of tobacco TGA factors. The At1g50570 and At5g55530 proteins function as transcriptional activators in yeast. At1g50570 protein activated the transcription of the â-glucuronidase reporter gene driven by the as-1 element in BY-2 protoplasts. At1g50570-green fluorescent protein fusion protein localized mainly to the nuclear envelope and endoplasmic reticulum in BY-2 protoplasts. Expression of the At1g50570 gene was induced in response to pathogen infection. Overexpression or repression of the At1g50570 gene in transgenic Arabidopsis plants did not alter the expression of the PR-1 gene under unstressed or salicylic acid -induced conditions. The role of At1g50570 TGA2.2 interaction in as-1-mediated gene activation has yet to be examined.TGA2.2 interacts specifically with At1g28480, a glutaredoxin protein, which is known to catalyze reductions of disulfides. At1g28480 interacts specifically with all identified members of the tobacco TGA transcription factors. The At1g28480-TGA2.2 interaction was neither affected by oxidative stress conditions nor by site-directed mutagenesis of the TGA2.2 and At1g28480 encoded conserved cysteine residues. At1g28480 enhances indirectly the TGA2.2 dimerization and in vitro DNA binding activity by increasing steady state TGA2.2 protein levels in yeast. The interaction between At1g28480 and TGA2.2 was confirmed in planta by a protoplast two-hybrid system. In addition, At1g28480 interacts via TGA2.2 with NPR1, a key regulator of systemic acquired resistance, in a yeast three-hybrid system. The At1g28480 gene is expressed in Arabidopsis plants in response to SA-inducing and pathogen infection. Overexpression or repression of the At1g28480 gene in transgenic Arabidopsis plants did not alter the salicylic acid-inducible gene expression. In contrast, overexpression of the At1g28480 repressed the auxin-induced expression of an as-1::GUS reporter construct in Arabidopsis transgenic plants. These results indicate that At1g28480 protein might function in signal transduction pathways involving TGA transcription factors.de
