Wind belonging to the PDI-family is located in the ER and is involved in dorso ventral patterning in drosophila. It is responsible for the correct targeting of Pipe, a transmembrane protein in Golgi. In colaboration with Dept. of Structural CHemistry University of Göttingen we succeeded in cristallizing Wind. As it turned out, Wind is a homodimer- constisting of a N-terminal thioredoxin- and C-terminally located D-domain. Both domains are connected via a linker region. By point mutagenesis of Wind I could identify a binding site for Pipe. It is mainly a cluster of tyrosines (Y53, Y55).By deletions of domains in Wind I characterized all domains involved in interacteion with Pipe.

Wind ist ein ER luminales Protein der Protein Disulfid Isomerase-Familie, welches seine Bedeutung in der Dorsoventral-Entwicklung von Drosophila hat. Es ist für die korrekte Lokalisation eines weiteren Proteins Pipe, einem Golgi-Transmembranprotein, verantwortlich. In Zusammenarbeit mit der Abt. f. Strukturchemie der Uni Göttingen konnte Wind bei einer Auflösung von 1,9 A kristallisiert werden. Es liegt als Homodimer vor- bestehend aus einer N-terminalen Thioredoxin- und C-terminalen D-Domäne. Beide Domänen werden über einen flexiblen Linker verbunden.Über Punktmutationen an Wind konnte eine Bindestelle für Pipe identifiziert werden. Dabei handelt es sich um ein Tyrosin-Cluster (Y53, Y55).Über Deletionsmutagenese von Wind konnten die an der Interaktion mit Pipe beteiligten Domänen charakterisiert werden.
