dc.description.abstractAminozucker sind essentielle Bausteine des Lebens, die in Bakterien vorallem als Vorläufer der Zellwandbiosynthese dienen. Ihre de novo-Synthese beginnt mit der Umsetzung von Fruktose-6-Phpsophat und Glutamin zu Glukosamin-6-Phosphat und Glutamat. Diese Reaktion wird durch die Glukosamin-6-Phosphat Synthase GlmS katalysiert. Escherichia coli ist in der Lage, externe Aminozuckerquellen zu verwerten, und unter diesen Bedingungen ist die Aktivität von GlmS nicht essentiell. Für eukaryotische Organismen und Gram-positive Bakterien sind Mechanismen beschrieben, die die Expression oder Enzymaktivität von GlmS an die Verfügbarkeit des eigenen Produktes oder von Metaboliten stromabwärts in der Zellwandbiosynthese koppeln. In E. coli war eine derartige Regulation bislang unbekannt. Die Beobachtung, dass Stämme mit einer Deletion des Gens yhbJ eine stark erhöhte GlmS-Menge aufweisen, gab eine ersten Ansatzpunkt für die Suche nach einer Kontrolle der GlmS-Aktivität in diesem Organismus. In dieser Arbeit konnte gezeigt werden, dass die spezifische Expression von glmS einer posttranskriptionellen Kontrolle unterliegt. Mittels Transposonmutagenese konnte die kleine, nicht-kodierende RNA GlmZ als notwendiger Faktor für diese Regulation identifiziert, sowie ein Einfluß von Rnase E und Hfq beschrieben werden. Diese Regulation stellt einen der ersten beschriebenen Fälle dar, in denen eine kleine RNA die Transkription eines Gens innerhalb eines Operons positiv beeinflusst. Des Weiteren konnte die intrazelluläre Verfügbarkeit von Glukosamin-6-Phosphat als Signal dieses Regulation identifiziert, werden. Somit erfolgt die Kontrolle der glmS-spezifischen Expression durch einen negativen Feedback-Mechanismus. Zudem wurde das Protein YhbJ analysiert und mehrere konservierte Motive in der Aminosäuresequenz identifiziert. Es wurde gezeigt, dass YhbJ in vitro an RNA binden kann. Ausserdem wurde die globale Auswirkung einer yhbJ-Deletion auf die Transkription mittels MicroArray Analysen untersucht.de

dc.description.abstractengAmino sugars are essential building blocks of life. In bacteria, they are mainly used as precursors for the biosynthesis of the cell wall. Their de novo synthesis starts with the transformation of fructose-6-phosphate and glutamine to glucosamine-6-phosphate and glutamate. This reaction is catalyzed by the glucosamine-6-phosphate synthase GlmS. Escherichia coli is able to utilize external sources of amino sugars, making the activity of GlmS non-essential. In eukaryotic organisms, the activity of GlmS is linked to the availability of its product or metabolites further downstream in the pathway of cell wall biosynthesis. Till now, no equivalent mechanism in E. coli was known. The observation that strains carrying a deletion of the gene yhbJ produce large amounts of GlmS gave a first hint for the search of the mechanism that controls the activity in of this enzyme in E. coli. In this work is shown that the specific expression of glmS is guided by a post-transcriptional control. By using transposon mutagenesis a small, non-coding RNA was identified as an essential factor in this regulation. Further, an influence of Rnase E and Hfq has been shown. Thsi regulation is one of the first cases in which a small RNA positively influences the transcription of a gene inside of an operon. Additionally, the level of glucosamine-6-phosphgate inside the cell was identified as the signal monitored by this regulation. Thereby the control of the glmS expression is a negative feedback loop. The protein YhbJ was analysed and several conserved motivs were identified in the amino acid sequence. Further it has been shown that YhbJ is able to bind RNA in vitro. The global effect of a yhbJ deletion upon tranbscription was examined by MicroArray analyses.de
