dc.description.abstractDie F420H2: Chinon Oxidoreduktase aus dem hyperthermophilen sulfatreduzierenden Archaeon Archaeoglobus fulgidus und die F420H2: Phenazin Oxidoreduktase des methanogenen Archaeons Methanosarcina spec. sind initiale Enzyme eines membrangebundenen Elektronentransportsystems und an der Energiekonservierung beteiligt.Die F420H2: Chinon Oxidoreduktase wird von den 11 Genen des fqo Operons kodiert (fqo J, K, M, L, N, A, BC, D, H, I, F). Die F420H2:Phenazin Oxidoreduktase hingegen wird von 13 Genen codiert, von denen nur 12 in dem fpo Operon organisiert sind (fpo A, B, C, D, H, I, J, K, L, M, N, O). Das Gen, welches für die Untereinheit FpoF kodiert ist an einer anderen Stelle im Genom lokalisiert. Mit Ausnahme der Gene fqoF/fpoF und fpoO, weist die abgeleitete Aminosäuresequenz aller Gene Homologien zu Untereinheiten der NADH: Ubichinon Oxidoreduktase (Komplex I) der Pro- und Eukaryonten auf. Daher liegt der Schluss nahe, dass es sich bei dem F420H2-abhängigen und dem NADH-abhängigen Enzym um funktionelle Äquivalente handelt.Ein Schwerpunkt dieser Arbeit lag in der Charakterisierung und Kristallisation, der als Input-Modul fungierenden FqoF/FpoF Untereinheiten, welche an die Oxidation von reduziertem F420H2 angepasst sind. Des Weiteren wurde die FpoO Untereinheit von Methanosarcina mazei, die weder im Komplex I noch in der F420H2: Chinon Oxidoreductase von A. fulgidus vorkommt und vermutlich an der Reduktion des Methanophenazins beteiligt ist, während dieser Arbeit charakterisiert. Hierzu wurden die Untereinheiten FqoF, FpoF und FpoO in Escherichia coli heterolog überproduziert und aufgereinigt. Die gereinigten F Untereinheiten waren in der Lage reduziertes F420 unter Verwendung des Elektronen-akzeptorsystems Methylviologen und Metronidazol, zu oxidieren. Alle heterolog produzierten Untereinheiten wiesen die aus der Primärsequenz vorausgesagten prosthetischen Gruppen auf. Das gereinigte FqoF Protein aus A. fulgidus wies einen Nicht-Häm-Eisen-Gehalt von 14 mol, einen säurelabilen Schwefelgehalt von 7 mol und 0.8 mol FAD pro mol Protein auf. Im Fall der F Untereinheit aus Methanosarcina acetivorans betrug der Gehalt von Nicht-Häm-Eisen, säurelabilen Schwefel und FAD 8, 7 und 0.9 mol pro mol Protein. Die FpoO Untereinheit aus Methanosarcina mazei wies einen Nicht-Häm-Eisen-Gehalt von 1,6 mol und einen Gehalt an säurelabilem Schwefel von 1,5 mol pro mol Protein auf. EPR Analysen der aufgereinigten Proteine ergaben Hinweise auf das Vorkommen von mindestens zwei [4Fe-4S] Zentren in den Input Modulen (FqoF/FpoF) und auf ein [2Fe-2S] Zentrum in der FpoO Untereinheit. In Kristallisationsexperimenten konnten die Input Module, FqoF und FpoF kristallisiert werden, jedoch wiesen die Kristalle nicht die erforderliche Streuung im Röntgenstrahl auf.de

dc.description.abstractengThe F420H2: quinone oxidoreductase from the hyperthermophilic sulfate reducing archaeon Archaeoglobus fulgidus and the F420H2: phenazine oxidoreductase from the methanogenic archaeon Methanosarcina spec. are both initial enzymes of membrane-bound electron-transport systems and are involved in energy conservation.The F420H2:quinone oxidoreductase is encoded by the fqo gene cluster which comprises 11 genes (fqo J, K, M, L, N, A, BC, D, H, I, F). The F420H2:phenazine oxidoreductase is encoded by 13 genes, 12 of them organized in the fpo gene cluster (fpo A, B, C, D, H, I, J, K, L, M, N, O). The gene encoding the subunit FpoF is not part of the operon and is located at a different part of the chromosome. With the exception of fqoF/fpoF and fpoO, the deduced amino acid sequences of all other genes show homologies to distinct subunits of the NADH-quinone oxidoreductases (Complex I) from prokaryotes and eukaryotes. Thus, it can be concluded that the F420H2-dependent and the NADH-dependent enzymes are functional equivalents.A main focus of this work was the characterisation and crystallisation, of the FqoF/FpoF subunits, which function as input device adjusted to the oxidation of reduced cofactor F420H2. Furthermore our interest focuses on the characterisation of the FpoO subunit from Methanosarcina mazei. Because the FpoO subunit from Methanosarcina species is neither found in Complex I nor in the F420H2: quinone oxidoreductase from A. fulgidus, it is possible that the polypeptide participates in the reduction of methanophenazine.In the course of this work the subunits FqoF, FpoF and FpoO were heterologous overproduced in Escherichia coli and purified. The purified F subunits were able to oxidise reduced cofactor F420 using the electron-acceptor system methyl viologen plus metronidazole. The heterologous produced subunits contained the prosthetic groups as predicted from the amino acid sequences. The purified FqoF protein from A. fulgidus contained 14 mol non-heme iron, 7 mol acid-labile sulfur and 0.8 mol FAD per mol protein. In case of the F subunit from Methanosarcina acetivorans the content of non-heme iron, acid-labile sulfur and FAD was 8 mol, 7 mol and 0.9 mol per mol protein, respectively. The FpoO subunit from Methanosarcina mazei contained 1.6 mol non-heme iron and 1.5 mol acid-labile sulfur. EPR analyses of the purified proteins indicate the presence of at least two [4Fe-4S] clusters in the input modules (FqoF/FpoF) and one [2Fe-2S] cluster in the FpoO subunit from Methanosarcina mazei. The input modules FqoF and FpoF were crystallised but the crystals showed no x-ray diffraction.de
