dc.description.abstractDie Informationsverarbeitung im Gehirn basiert darauf, dass sich die synaptischen Verbindungen zwischen den Nervenzellen in ihrer Stärke ständig anpassen können. Die Stärke der exzitatorischen Synapsen im Hippocampus hängt von der Anzahl der AMPA Rezeptoren (AMPARs) in der postsynaptischen Verdichtung (PSD) ab. PSD-95 ist das prototypische Ankerprotein in der PSD, dem eine wichtige Rolle für die Funktion von AMPARs zugesprochen wird. PSD-95 gehört zu der Proteinfamilie der PSD-MAGUKs, zu der auch die Proteine SAP97, SAP102 und PSD-93 gehören. Die PSD-MAGUKs haben die gleiche Abfolge von Proteindomänen, existieren aber in unterschiedlichen Isoformen, die sich in der N-terminalen Aminosäuresequenz unterscheiden. Es wurde zuvor gezeigt, dass diese N-terminalen Unterschiede die Funktion von PSD-95 und SAP97 diktieren. PSD-93 existiert in sechs unterschiedlichen Isoformen. Die alpha1, alpha2 und beta-Isoformen sind analog zu den alpha- und beta-Isoformen von PSD-95 und SAP97. Zusätzlich hat PSD-93 drei Isoformen, die einzigartig unter den PSD-MAGUKs sind, und die bisher nicht analysiert wurden. In der vorliegenden Arbeit wurde das Expressionsmuster der PSD-93 Isoformen im Hippocampus analysiert und ihre Funktion mit Hilfe von Elektrophysiologie charakterisiert. Mit Massenspektrometrie und Western Blotting konnten PSD-93beta, gamma1, delta sowie eine oder beide alpha-Isoformen im Hippocampus nachgewiesen werden, wobei die alpha-Isoformen am Stärksten exprimiert sind. Überexpressionsstudien in den CA1-Pyramidenzellen in Schnittkulturen vom Hippocampus ergaben, dass nur PSD-93delta und epsilon AMPAR EPSCs um 50-100% erhöhen und somit einen ähnlichen Phenotyp wie die alpha-Isoformen von PSD-95 und SAP97 haben. Obwohl das N-terminale alpha-Exon von PSD-93 sich prinzipiell wie ein klassisches alpha-Exon verhält, kann es die alpha-Funktion nicht ausüben, weil der PSD-93 N-terminus eine Sequenz enthält, die inhibitorisch auf das alpha-Exon wirkt. Da PSD-93delta und epsilon lediglich 11% der PSD-95 Expressionslevel ausmachen ist es nicht verwunderlich, dass der Verlust von PSD-95 durch RNA Interferenz, jedoch nicht der Verlust von PSD-93, die basale synaptische Transmission beeinträchtigen. Sowohl PSD-93beta als auch PSD-93delta, jedoch nicht die anderen PSD-93 Isoformen, reagierten auf Aktivitätsveränderungen in der Schnittkultur. Es ist daher möglich, dass die Rolle der PSD-MAGUKs in Plastizität nicht, wie zuvor vermutet, auf die beta-Isoformen limitiert ist, sondern dass auch PSD-93delta für Plastizitätsmechanismen verantwortlich ist.de

dc.description.abstractengChanges in the strength of synaptic transmission are thought to underlie information processing in the brain. At excitatory synapses in the hippocampus, synaptic strength is determined by the number of AMPA receptors (AMPARs) at the postsynaptic density (PSD). PSD-95 is the prototypical scaffolding protein in the PSD, and has been proposed to be important for synaptic AMPAR function. PSD-95 belongs to the PSD-MAGUK protein family that includes SAP97, SAP102 and PSD-93. PSD-MAGUKs share a common domain structure, but they exist as different isoforms with distinct N-terminal amino acid sequences. These N-termini have previously been shown to govern the function of PSD-95 and SAP97 isoforms, implying selective roles in modulating synaptic strength. PSD-93 has six alternative N-termini, including alpha1, alpha2, and beta, which are similar to the alpha- and beta-isoforms of PSD-95 and SAP97, and three unique ones, which have not been analyzed so far. In this thesis, I analyzed the expression pattern of PSD-93 isoforms in the hippocampus and characterized their function by electrophysiology. Using mass spectrometry and western blotting, PSD-93beta, gamma1, delta, and one or both alpha-isoforms of PSD-93 were detected in the hippocampus. The alpha-isoforms were the most abundant of all. Gain-of-function studies in CA1 pyramidal cells of hippocampal slice cultures demonstrated that only PSD-93delta and epsilon increased AMPAR EPSCs by 50-100%, similar to the alpha-isoforms of PSD-95 and SAP97. Although the alpha-N-terminal exon of PSD-93 in principle behaved like a classical alpha-isoform, an inhibitory domain was discovered that blocked the positive action of endogenous PSD-93alpha1/2 on AMPARs. Consistent with this and the fact that PSD-93delta and epsilon make up at most 11% of PSD-95, knockdown of PSD-95, but not PSD-93, affected basal synaptic transmission. Activity manipulations revealed differential effects of the individual isoforms, and provide evidence that the activity-dependence, and potentially the role in long-term plasticity, may be governed by several PSD-MAGUKs, including the beta-isoforms and PSD-93delta.de
