dc.description.abstractEine neue Möglichkeit zur Erforschung der großen metabolischen und genetischen Diversität von Mikroorganismen ist die Konstruktion von Genbanken direkt aus Umweltproben. Zur Identifizierung neuer Gene für Koenzym B12-abhängige Glycerin-und Diol-Dehydratasen wurden vier sogenannte Metagenom-Banken hergestellt. Als Proben wurde Boden eines Zuckerrübenfeldes, Sediment des Flusses Grone (beide nahe Göttingen, Deutschland), Sediment des Solar Lake (Ägypten) und Sediment des Golfs von Eilat (Isarel) verwendet. Die Durchmusterung auf Glycerin- und Diol-Dehydratasen basierte auf der Komplementation eines konstruierten Dehydratase-negativen E. coli Stammes. Des weiteren wurde die Methode der Koloniehybridisierung mit Dehydratase-spezifischen DNA-Fragmenten genutzt. Die Untersuchung von ca. 720 000 E. coli-Klonen erbrachte sieben positive Klone (E. coli pAK2 - pAK8). Zwei der Plasmide beinhalteten Dehydratase-Gene, identisch zu denen aus Citrobacter freundii und wurden daher nicht weiter untersucht. Drei der verbleidenden fünf Plasmide codierten für Glycerin-, zwei für Diol-Dehydratasen. Mit zwei der Glycerin-Dehydratasen und einer komplementierten Diol-Dehydratasen wurde eine Überproduktion und eine Reinigung der Enzyme durchgeführt. Die Enzyme unterlagen einer sogenannten suicide -Inaktivierung durch Glycerin, konnten aber durch den Reaktivierungsfaktor (DhaFG) der Glycerin-Dehydratase aus C. freundii kreuz-reaktiviert werden. Alle untersuchen Dehydratasen unterlagen außerdem einer Inaktivierung durch das Glycerin-Fermentationsprodukt 1,3-Propandiol.Etwa 300 000 E.coli-Klone der oben genannten Genbanken wurden auf Indikatoragar auf die Bildung von Carbonylen aus Polyolen untersucht. 16 aus ursprünglich 24 positiven E. coli-Klonen beinhalteten Plasmide (pAK101 bis pAK116) die einen stabilen Phänotyp vermittelten. Acht der positiven Klone zeigten NAD(H)-abhängige Alkohol-Oxidoreduktase-Aktivität mit Polyolen oder Carbonylen als Substrat. Durch die Sequenzierung konnten auf den Inserts der Plasmide 36 vollständige und 17 unvollständige, potentielle Gene identifiziert werden. Die für die Carbonyl-Bildung verantwortlichen Gene konnten für neun der Plasmide identifziert werden. Die Analyse der abgeleiteten Aminosäuresequenzen ergab, dass drei Gene (orf12, orf14 und orf22) neue Alkohol-Dehydrogenasen kodierten, vier (orf5, sucB, fdhD, und yabF) kodieren zwei mögliche Oxidoreductasen, die nicht zu den Alkohol-Dehydrogenasen gehören, eines (glkP) kodierte eine Glycerin-Kinase und eines (orf1) kodierte für eine Protein, das keinerlei Homologie zu bisher bekannten Genprodukten zeigte. Weitere 24 positive Klone wurden bei der Durchmusterung von drei bereits bestehenden Genbanken erhalten. 15 Klone enthielten Plasmide, die einen stabilen Phänotyp in E. coli vermittelten. Diese wurden mit pAK201-215 bezeichnet. Die weitere Analyse konzentrierte sich auf 7 Plasmide, die eine NAD(H)-abhängige Alkohol-Oxidoreduktase-Aktivität vermittelten. Durch Subklonierung konnten drei für den Phänotyp verantwortliche Gene (orf6, orf24 und orf25) identifiziert werden. Die Sequenzen zeigten keine signifikante Ähnlichkeit zu bekannten Alkohol-Oxidoreduktasen, sie beinhalteten jedoch Glycin-reiche Regionen, die typisch für die Bindung von Nikotinamid-Kofaktoren sind.de

dc.description.abstractengA new way to exploit the metabolic and genetic diversity of microorganisms is the construction of DNA libraries directly from environmental samples. To isolate new genes encoding coenzyme B12-dependent glycerol and diol dehydratases and genes conferring oxidation of short-chain (C2 to C4) polyols or reduction of the corresponding carbonyls four so called metagenomic DNA libraries were construct. As samples were used a sugar beet field, sediment from the river Grone (both near Göttingen, Germany), sediment from Solar Lake (Egypt), and sediment from Gulf of Eilat (Israel). The screening for glycerol- and diol dehydratase genes was based on complementation of a constructed dehydratase-negative Escherichia coli strain. In addition, screening was performed by colony hybridization with dehydratase-specific DNA fragments as probes. The screening of about 720,000 E. coli clones by these methods yielded seven positive clones (E. coli pAK2 - pAK8). Two of the plasmids recovered contained genes identical to those encoding the glycerol dehydratase of Citrobacter freundii and were not studied further. Three of the remaining five plasmids coded for glycerol dehydratases and two coded for diol dehydratases, which were all similar to the corresponding regions of enteric bacteria. High-level production and purification could be performed with two glycerol and one complemented diol dehydratase. The enzymes were subject to suicide inactivation by glycerol and but could be cross-reactivated by the reactivation factor (DhaFG) for the glycerol dehydratase of C. freundii. Furthermore all examined dehydratases were inhibited in the presence of the glycerol fermentation product 1,3-propanediol. Approximately 300,000 E. coli strains of the above mentioned libraries were screened for the production of carbonyls from polyols on indicator agar. Sixteen out of initially detected 24 positive E. coli clones contained a plasmid (pAK101 to pAK116), which conferred a stable phenotype. Eight of the positive clones exhibited NAD(H)-dependent alcohol oxidoreductase activity with polyols or carbonyls as the substrates. Sequencing revealed that the inserts of the plasmids encoded 36 complete and 17 incomplete presumptive protein-encoding genes. The genes responsible for the carbonyl formation of E. coli were identified for nine of the plasmids. Analyses of the deduced amino acid sequences revealed that three genes (orf12, orf14, and orf22) encoded novel alcohol dehydrogenases, four (orf5, sucB, fdhD, and yabF) encoded novel putative oxidoreductases belonging to groups distinct from alcohol dehydrogenases, one (glpK) encoded a putative glycerol kinase, and one (orf1) encoded a protein which showed no similarity to any other known gene product. Another 24 positive E. coli clones were obtained in a screening of three already existing libraries. Fifteen of them contained recombinant plasmids, designated pAK201-215, which conferred a stable phenotype on E. coli Sequencing revealed that the inserts of pAK201-215 encoded 26 complete and 14 incomplete predicted protein-encoding genes. The further analysis was focused on the 7 plasmids recovered from the positive clones, which exhibited an NAD(H)-dependent alcohol oxidoreductase activity. Three genes (orf6, orf24, and orf25) conferring this activity were identified during subcloning. The sequences of the three deduced gene products revealed no significant similarities to known alcohol oxidoreductases, but contained putative glycine-rich regions, which are characteristic for binding of nicotinamide cofactors.de
