Funktionelle Charakterisierung der Transkriptionsfaktoren Nkx2.2 und Arx in der Entwicklung der endokrinen Zellen im murinen Pankreas

dc.description.abstractDie Bauchspeicheldrüse lässt sich gemäß ihrer funktionalen Eigenschaften in zwei Kompartimente unterteilen: Exokrine Zellen zur Produktion von Verdauungsenzymen und endokrine Zellen zur Produktion Blutzucker-regulierender Hormone. Endokrine Zellen aggregieren typischerweise in kugelförmigen Anordnungen, den Langerhans'schen Inseln, die im exokrinen Gewebe eingebettet sind. Es werden fünf endokrine Zelltypen aufgrund ihrer Hormonproduktion unterschieden: Beta-, Alpha-, Delta-, PP- und Epsilon-Zellen welche Insulin, Glucagon, Somatostatin, Pankreatisches Polypeptid bzw. Ghrelin produzieren. Die Differenzierung pankreatischer Vorläuferzellen in diese endokrinen Subtypen unterliegt der Steuerung durch diverse Transkriptionsfaktoren. Knockout- und Überexpressions-Studien haben offenbart, dass der Transkriptionsfaktor Arx für die Ausbildung Glucagon-produzierender Alpha-Zellen essenziell ist. Arx-Knockout Mäuse entwickeln keine Alpha-Zellen wohingegen die Populationen der Beta- und Delta-Zellen vergrößert ist (Collombat et al., 2003). Die Überexpression von Arx in pankreatischen Vorläufern resultiert in einer Zunahme von Alpha- aber auch PP-Zellen auf Kosten der Beta-und Delta-Zellpopulation (Collombat et al., 2007). Nkx2.2, ein Transkriptionsfaktor aus der Klasse der NK-Homeobox-Gene, In Nkx2.2-Knockout-Mäusen sind die Beta- und Alpha-Zell Populationen stark reduziert, wohingegen ein Ghrelin-exprimierender Zelltyp dominiert. Interessanterweise sind in Nkx2.2-defizienten Pankreata fünf-fach erhöhte Arx-Transkriptmengen zu detektieren (Sussel et al., 1998, Chao et al., 2007). Daraus ergab sich die Frage nach der funktionellen Verbindung von Nkx2.2 und Arx. Detaillierte Analysen der Pankreata von Wildtypen, Nkx2.2-/- / Arx- Einfach- und Doppelmutanten, lassen den Schluss zu, dass die kombinierte Aktivität von Nkx2.2 und Arx zur Entwicklung der Alpha-Zellen notwendig ist. Im Gegensatz dazu erscheint Nkx2.2 in entstehenden Beta-Zellen notwendig zur Kontrolle der Arx-Expression. Wie bereits angesprochen resultiert die Überexpression von Arx in pankreatischen Vorläufern in der Bildung zahlreicher Glucagon- aber auch PP-exprimierender Zellen (Collombat et al., 2007). Daraus ergaben sich die Fragen ob (1.) Arx ausreichend ist um diese Differenzierungsprogramme zu aktivieren und (2.) in welchem spatio-temporalen Kontext diese Programme initiiert werden. Zur Beantwortung dieser Fragen wurde eine transgene Linie erzeugt, mit Hilfe derer ein chemisch induzierbares ArxER-Fusionsprotein in pankreatischen Vorläuferzellen Arx-defizienter Tiere aktiviert werden konnte. Die Aktivierung von ArxER zu frühen Stadien der Pankreatogenese resultierte dabei in der vornehmlichen Bildung Glucgon+ Zellen wohingegen die Aktivierung ab Embryonaltag 13.5 zusätzlich in der Bildung zahlreicher PP+ Zellen resultiert. Diese Ergebnisse zeigen, dass Arx ausreichend ist um diese Differenzierungsprogramme zu aktivieren. Ein weiterer Aspekt dieser Arbeit war die Funktion des Transkriptionsfaktors Pax4 in Beta-Zellen adulter Mäuse. Mehrere Studien offenbarten, das Pax4 zur Bildung der Beta-Zellen essenziell ist. In Pax4-Knockout-Tieren ist die Entwicklung von Beta-Zellen fast vollständig verhindert (Sosa-Pineda et al., 1997). Aufgrund der daraus resultierenden frühen Letalität dieser Mutanten (2 Tage nach der Geburt) blieb jedoch offen ob Pax4 auch in reifen Beta-Zellen adulter Tiere eine Funktion zukommt. Zur Beantwortung dieser Frage wurde eine konditionale Pax4-Knockout Linie erzeugt. Die Beta-Zell spezifische Deletion von Pax4 mit Hilfe einer Insulin-Cre Linie führte zu keiner offensichtlichen Beeinträchtigung normaler Beta-Zellfunktionen in der Maus. Diese Ergebnisse zeigen, dass Pax4 zwar maßgeblich an der Spezifizierung endokriner Vorläufer in Richtung eines Beta-Zellschicksals beteiligt ist, aber mit beginnender Insulin-Produktion redundant wird.de

dc.description.abstractengThe pancreas is a compound-gland structured into two functional units exhibiting exocrine and endocrine properties. Exocrine cells produce and secret digestive enzymes, whereas endocrine cells produce blood sugar regulating hormones. Endocrine cells are typically organized in clusters termed Islets of Langerhans that are embedded in the exocrine tissue. Five different endocrine cell types are specifiable due to the production of distinct hormones: Beta-, Alpha-, Delta-, PP- and Epsilon-cells that produce insulin, glucagon, somatostatin, pancreatic polypeptide and ghrelin, respectively. The differentiation of pancreatic progenitor cells into an endocrine cell subtype is controlled by the concerted interplay of numerous transcription factors. Loss- and gain-off function studies revealed that the homeobox-containing factor Arx is essential for the differentiation of the alpha-cell lineage. Arx-knockout mice lack glucagon-producing alpha-cells and exhibit increased beta- and delta-cell populations (Collombat et al., 2003). Conversely, the forced expression of Arx in pancreatic progenitors results in the formation of alpha- but also PP-cells at the expense of the beta- and delta-cells (Collombat et al., 2007). The NK-class homeobox factor Nkx2.2, has been shown to act in the differentiation of the alpha- and beta-cell lineages as these cell types lack in Nkx2.2-deficient animals and are replaced by ghrelin-expressing cells (Prado et al., 2004, Sussel et al., 1998). Notably, Arx mRNA levels are fivefold increased in Nkx2.2-deficient pancreata, suggesting a genetic link between both factors (Chao et al., 2007). Comparative analysis of pancreata of Nkx2.2-/- /Arx- single- and double-mutant animals suggest that the combined activity of Nkx2.2 and Arx is necessary for the formation of alpha-cells. However, Arx directly co-localizes with beta-cell specific markers like iAPP, PC1/3 and Pdx1 in Nkx2.2-deficient mice, suggesting an involvement of Nkx2.2 in the repression of Arx in committed beta-cells. As mentioned above, the forced expression of Arx in pancreatic progenitors results in the development of numerous alpha- but also PP-cells. These findings provoked the questions whether (1) Arx is sufficient to initiate the alpha- and PP-cell differentiation programs and (2) in which spatio-temporal manner. To address these questions a transgenic and inducible Arx-rescue model was established. Thereby, the early activation of Arx in Arx-deficient pancreata results in the recovery of glucagon-producing cells, whereas the activation at later developmental stages induces glucagon as well as PP-expressing cells. Hence, Arx is sufficient to induce the programs of alpha- and PP-cell differentiation in a time-dependent manner. Furthermore the role of the transcription factor Pax4 was investigated. Pax4 has been shown to be essential for the differentiation of beta-cells. Pax4-deficient animals lack almost all beta-cells (Sosa-Pineda et al., 1997). Due to the resulting lethality nothing was known about the role of Pax4 in mature beta-cells in adult mice. To address this question we generated a Pax4-conditional knockout line. The subsequent beta-cell specific deletion of Pax4, using an insulin-cre line, did neither affect normal beta-cell differentiation nor function. Thus although Pax4 is essential for the specification of the beta-cell, it is redundant after insulin-activation.de
