dc.description.abstractSNARE-Proteine sind in allen Eukaryonten an der Fusion von Transport-Vesikeln mit Zielmembranen beteiligt, wobei sie Komplexe bilden in denen jeweils 3 Q-SNAREs mit einem R-SNARE interagieren. Im ersten Teil der Dissertation wurde über einen Multicopy-Suppressor-Screen versucht, zusätzliche Proteine aufzufinden, die mit dem Hefe-Q-SNARE Vti1p im Transport vom trans-Golgi zur Prävakuole oder zur Vakuole interagieren. Es gelang ein bislang uncharakterisiertes nicht-gessentielles Gen, welches als VTS1 bezeichnet wurde, und das R-SNARE YKT6 als Suppressoren des Wachstumsdefektes und des Defektes im Transport zur Vakuole der temperatursensitiven vti1-2-Mutante zu identifizieren. VTS1 zeigte dabei eine spezifische genetische Interaktion mit VTI1 ausschließlich im letzten Transportschritt zu Vakuole, nicht jedoch im Transport zur Prävakuole. YKT6 interagierte mit VTI1 zusätzlich zu seiner bereits beschriebenen Funktion im retrograden Transport zum cis-Golgi und in der homotypischen Vakuolenfusion auch im Transport zur Prävakuole und Vakuole. Dies deutet darauf hin, dass es sich bei Ykt6p um das fehlende R-SNARE in den SNARE-Komplexen des biosynthetischen Transportes zur Prävakuole (Ykt6p, Vti1p, Pep12p und Syn8p) und Vakuole (Ykt6p, Vti1p, Vam3p und Vam7p) handelt. Im zweiten Teil der Arbeit gelang es über eine Datenbanksuche ein neues Q-SNARE-Protein in Saccharomyces cerevisiae zu identifizieren und charakterisieren, welches als Use1p bezeichnet wurde und ER-lokalisiert ist. Mittels spezifischer Transport-Assays mit temperatursensitiven Mutanten des Proteins konnte gezeigt werden, dass Use1p am retrograden Transport vom cis-Golgi zum ER beteiligt ist. Dieses Ergebnis wurde auch durch starke genetische Interaktionen zwischen USE1 und Genen die für SNAREs des retrograden Golgi-zu-ER-SNARE-Komplexes codieren unterstützt. Anti-Use1p-Antikörper koimmunpräzipitierten außerdem spezifisch SNAREs des retrograden Golgi-zu-ER-SNARE-Komplexes, jedoch nicht solche des anterograden SNARE-Komplexes. Daher scheint der SNARE-Komplex des retrograden Transportes vom cis-Golgi zum ER aus den SNARE-Proteinen Use1p, Ufe1p, Sec22p und Sec20p aufgebaut zu sein.de

dc.description.abstractengSNARE proteins are involved in the fusion of transport vesicles with target membranes in all eukaryotes and form complexes in which 3 Q-SNAREs interact with one R-SNARE. In the first part of the work I tried to find additional proteins which interact with the yeast Q-SNARE Vti1p in the transport from the trans-Golgi to the prevacuole and vacuole via an multicopy suppressor screen. I was able to identify a previously uncharacterized non-essential gene, which was named VTS1, and the R-SNARE YKT6 both as suppressors of the growth and vacuolar transport defect of the temperature sensitive vti1-2 mutant. VTS1 showed a specific genetic interaction with VTI1 only in the last transport step to the vacuole, but not in the transport to the prevacuole. In addition to its previously described function in retrograde traffic to the cis-Golgi and in homotypic vacuolar fusion, YKT6 also interacted with VTI1 in traffic to the prevacuole and vacuole. This indicates, that Ykt6p is the missing R-SNARE in the SNARE-complexes of the biosynthetic transport to the prevacuole (Ykt6p, Vti1p, Pep12p and Syn8p) and vacuole (Ykt6p, Vti1p, Vam3p and Vam7p). In the second part of the work I was able to identify and characterize a new Q-SNARE protein, named Use1p, in Saccharomyces cerevisiae via a database search, which was localized to the ER. In specific transport assays with temperature sensitive mutants of the protein I could show that Use1p is involved in the retrograde transport from the cis-Golgi to the ER. This was also supported by strong genetic interactions between USE1 and the genes encoding SNAREs in retrograde traffic to the ER. In addition, anti-Use1p-antibodies specifically co-immunoprecipitated SNAREs of the retrograde Golgi-to-ER-SNARE complex but not those of the anterograde SNARE complex. Therefore it seems, that the SNARE complex of the retrograde transport from the cis-Golgi to the ER consists of the SNARE proteins Use1p, Ufe1p, Sec22p and Sec20p.de
