dc.description.abstractZiel der vorliegenden Arbeit war es, die Transkriptions-Regulation der 11 humanen replikationsabhängig-exprimierten H3-Histongene aufzuklären und Beziehungen zur in vivo-Expression der Gene zu untersuchen. Im ersten Teil der vorliegenden Arbeit wurde die in vivo-Expression der 11 H3-Histongene in drei humanen fötalen Geweben (Blase, Leber und Lunge) und acht humanen Zelllinien mit quantitativen RNase-Protektionsassays bestimmt. Die drei fötalen Gewebe und die diploide Zelllinie IMR-90 zeigten ein ähnliches H3-Expressionsmuster. Im Gegensatz dazu zeigten die sieben untersuchten Tumorzelllinien ein stark abweichendes, aberrantes H3-Expressionsmuster, das sich vor allem in einer Vielzahl von nicht oder nur schwach exprimierten H3-Genen zeigte. Die Aberration ließ sich weder auf genetische Mutationen noch auf variierende Transkriptionsfaktor-Konzentrationen zurückführen. Das aberrante H3‑Expressionsmuster in den Tumorzelllinien ist daher sehr wahrscheinlich auf epigenetische Mechanismen zurückzuführen, wie z.B. DNA-Methylierung, Histon-Modifikationen oder Chromatin-Strukturen.Im zweiten Teil der vorliegenden Arbeit wurde die Trankriptionsregulation der H3-Histongene untersucht. Dazu wurden zunächst in Reportergenassays mit Promotor-Deletionsreihen die Struktur und die funktionale Länge der Promotoren der Gene H3/d, H3/h, H3/i, H3/k, H3/m und H3/n bestimmt. Mit Ausnahme von H3/i hatten alle Promotoren schon bei Längen von 99-228 bp nahezu maximale Aktivität: H3/m 228 bp, H3/k 99 bp, H3/h 115 bp, H3/n 186 bp und H3/d 176 bp. Darüber hinaus wurde eine Aktivierung des H3/m-Gens durch das upstream gelegene H2A/m-Gen beobachtet und näher charakterisiert. Es handelte sich um einen Histongen-spezifischen Effekt, der eine mögliche Erklärung für die Clusteranordnung der replikationsabhängig-exprimierten Histongene ist. Um die relevanten cis-regulatorischen Elemente in den Histon H3-Promotoren zu identifizieren, wurden die funktionalen Bereiche des H3/m- und der H3/k-Promotors mit Hilfe der absuchenden Mutagenese untersucht. Im H3/m-Promotor wurden die TATA-Box, die proximale und distale CCAAT-Box und eine ATF-Box identifiziert. Im H3/k-Promotor wurden als wesentliche funktionelle Elemente die TATA-Box und die proximale und distale CCAAT-Box identifiziert. Der funktionelle Grundaufbau der H3‑Promotoren besteht aus einer TATA-Box, der proximalen CCAAT-Box und der distalen CCAAT-Box. Weitere Mutationsexperimente im H3/k-Promotor konnten zeigen, dass das zentrale CCAAT"-Motiv der CCAAT-Boxen und die Natur der umliegenden Basen für die Funktion der CCAAT-Boxen entscheidend waren. In EMSA-Analysen band der Transkriptionsfaktor CBF/NF‑Y mit unterschiedlicher Intensität an die CCAAT-Boxen des H3/k- und des H3/m-Promotors. Somit ist CBF/NF-Y vermutlich der primäre DNA-bindende Faktor in der Histon H3-Promotoraktivierung. Die Länge der Abstände zwischen proximaler und distaler CCAAT-Box, sowie zwischen proximaler CCAAT-Box und TATA-Box waren für die volle Aktivität des H3/k-Promotors entscheidend. Offenbar bilden sich an den H3‑Promotoren Aktivierungskomplexe mit definierter Struktur und begrenzter Flexibilität.de

dc.description.abstractengThe intention of the present thesis was to elucidate the transcriptional regulation of the 11 replication-dependent histone H3 genes and to investigate relationships to the in vivo expression of the histone H3 genes. In the first part of the present thesis, the expression of the 11 replication-dependent histone H3 genes was determined in three fetal human tissues (bladder, liver and lung) and eight human cell lines with a quantitative RNase-protection assay. In the three fetal human tissues and in the diploid cell line IMR-90 the histone H3 expression pattern was similar. In contrast, in the examined seven tumor cell lines the histone H3 expression pattern was aberrant, with many histone H3 genes being not expressed or expressed in very low amounts. This aberrant expression pattern was neither the cause of genetic mutations nor the cause of varying transcription factor concentrations. Therefore, it is very probably caused by epigenetic mechanisms, such as DNA methylation, histone modifications or chromatin structure.In the second part of the present thesis the transcriptional regulation of the histone H3 genes was investigated. Initially, the promoters of the histone H3 genes H3/d, H3/h, H3/i, H3/k, H3/m and H3/n were analyzed using stepwise deletion coupled reporter gene assays. With the exception of H3/i all promoters showed almost maximal activity at lengths between 99 and 228 bp: H3/m 228 bp, H3/k 99 bp, H3/h 115 bp, H3/n 186 bp und H3/d 176 bp. In addition, a distal activation of the H3/m gene by the upstream H2A/m gene was observed and further characterized. The effect was histone gene specific and is a possible explanation for the cluster organisation of the replication dependent histone genes. To identify the relevant cis-regulatory elements in the histone H3 promoters, the functional regions of the H3/m- and the H3/k-promoter were analyzed using scanning mutagenesis. In the H3/m promoter, a TATA-box, a proximal and a distal CCAAT-box and an ATF-box were the essential functional elements. In the H3/k promoter, a TATA-box, a proximal and a distal CCAAT-box were the essential functional elements. The functional framework of the histone H3 promoters consists of a TATA-box, a proximal and a distal CCAAT-box. Further mutagenesis experiments in the H3/k promoter showed, that the central CCAAT"-motif and the nature of the surrounding bases were essential for the function of the CCAAT-boxes. In EMSA-experiments, the transcription factor CBF/NF-Y bound to the CCAAT-boxes of the H3/k and the H3/m gene with varied intensity. Therefore, CBF/NF-Y is probably the primary DNA-binding factor in histone H3 promoter activation. The length of the spacings between the proximal and the distal CCAAT-box, as well as between the proximal CCAAT-box and the TATA-box were essential for full activity of the H3/k promoter. Evidently, activation complexes form on the histone H3 promoters that have a defined structure and limited flexibility.de
