The present study reports on responses of olfactory receptor neurons (ORNs) upon application of amino acids and pharmacological agents activating the cAMP-mediated transduction pathway (forskolin and membrane-permeable cAMP) using a novel slice preparation of the olfactory epithelium of Xenopus laevis tadpoles. Responses were measured using the patch-clamp technique and calcium imaging. Both amino acids and agents activating the cAMP-mediated transduction pathway proved to be potent stimuli. Interestingly, a number of ORNs that responded to amino acids did respond neither to forskolin nor to membrane-permeable cAMP. This suggests that some amino acids activate transduction pathways other than the well-known cAMP-mediated one. The differential processing of cAMP-mediated stimuli on one hand and amino acid stimuli on the other was further elucidated by calcium-imaging of olfactory bulb neurons using a novel nose-olfactory bulb preparation of Xenopus laevis tadpoles. The projection pattern of amino acid-sensitive ORNs to olfactory bulb neurons differed markedly from the projection pattern of forskolin-sensitive ORNs. Olfactory bulb neurons activated by amino acids were located laterally compared to those activated by forskolin, and only a small proportion responded to both stimuli. The ensemble of neurons activated by forskolin was also activated by IBMX and membran-permeable cAMP. On the basis of these results it can be concluded that sensory transduction of a number of amino acids is cAMP-independent, and amino acid- and cAMP-mediated responses are processed differentially at the level of the olfactory bulb.

Die vorliegende Untersuchung berichtet über Reizantworten olfaktorischer Rezeptorzellen (ORZ) in einer neuartigen Gewebeschnittpräparation des olfaktorischen Epithels von larvalen Xenopus laevis nach Applikation von Aminosäuren und pharmakologischen Wirkstoffen, die den cAMP-vermittelten Transduktionsweg aktivieren (Forskolin und membranpermeables cAMP). Die Reizantworten wurden mittels der "patch-clamp" Technik und "calcium imaging" gemessen. Sowohl Aminosäuren als auch die Wirkstoffe, die den cAMP-vermittelten Transduktionsweg aktivieren, erwiesen sich als wirksame Reize. Interessanterweise reagierten etliche ORZ, die auf Aminosäuren eine Reaktion zeigten, weder auf Forskolin noch auf membranpermeables cAMP. Dieses Ergebnis suggeriert, dass einige Aminosäuren andere Transduktionswege als den wohlbekannten cAMP-vermittelten Transduktionsweg aktivieren. Die unterschiedliche Verarbeitung von cAMP-vermittelten Reizen einerseits und Aminosäuren andererseits wurde zusätzlich durch "calcium imaging" von Neuronen des Bulbus olfactorius in einer neuartigen Präparation von larvalen Xenopus laevis, die Epithel und Bulbus olfactorius enthält, erläutert. Das Projektionsmuster von aminosäuresensitiven ORZ auf Neurone des Bulbus olfactorius unterschied sich merklich vom Projektionsmuster von forskolinsensitiven ORZ. Neurone des Bulbus olfactorius, die von Aminosäuren aktiviert wurden, lagen in einer lateralen Position verglichen mit jenen, die von Forskolin aktiviert wurden. Nur ein kleiner Anteil von Neuronen wurde von beiden Reizen aktiviert. Das Ensemble von Neuronen, das von Forskolin aktiviert wurde, wurde auch von dem Phosphodiesterasehemmer IBMX und von membranpermeablem cAMP aktiviert. Aufgrund dieser Ergebnisse kann gefolgert werden, dass die sensorische Transduktion von Aminosäuren weitestgehend cAMP-unabhängig ist und dass aminosäurenvermittelte und cAMP-vermittelte Reizantworten im Bulbus olfactorius unterschiedlich verarbeitet werden.
