Zur sequentiellen Umwandlung von Bicyclobutylidenen in Bicyclooctene: Synthese und Umlagerung funktionalisierter Spiro[3.4]octane

dc.description.abstractMit der vorliegenden Arbeit wurde versucht, das Potential einer sequentiellen Transformation von Bicyclobutylidenen in Bicyclo[3.3.0]octene auszuloten. Die zu prüfende Reaktionssequenz umfaßte eine Folge aus Epoxidierung, Oxaspirohexan-Cyclopentanon-Umlagerung, Methylierung und/oder Reduktion bzw. Addition von Methyllithium. Im ersten Teil der Arbeit lieferte unsubstituiertes Bicyclobutyliden (4) auf diese Weise über Spiro[3.4]octan-5-one 14, 50 und 51 sowie die Spiro[3.4]octan-5-ole 21 - 25 die mono- bis trimethylierten Bicyclo[3.3.0]octene (Hexahydropentalene) 56, 58, 59, 60 und 61. Das Ziel des zweiten Teils der Arbeit war eine Synthese des Bicyclo[3.3.0]octens 31 und des bicyclischen Ketons 30 als Partialstrukturen der Sesquiterpene (±)-Ptychanolid (33) und (±)-Ceratopicanol (32). Dazu durchgeführte Arbeiten zur Synthese und sequentiellen Transformation von 2,2-Dimethylbicyclobutyliden (26) brachten nicht den gewünschten Erfolg. Bei der Cyclobutylidenierung von 29 traten neben 26 mit 62 und 63 auch Kondensationsprodukte auf. Das aus 26 durch Epoxidierung erhaltene Oxaspirohexan 64 öffnete nicht regiospezifisch und lieferte neben dem benötigten Spiroketon 27 auch das Isomer 67. Daraufhin durchgeführte Versuche zur selektiven Darstellung von 27 und 67 über die Hydroxyselenide 49 bzw. 76 brachten zwar eine deutliche Verbesserung, aber bei 27 immer noch nicht genügend Material um die angestrebte Synthese von 28 und dessen Umlagerung zu 31 als potentiellen Vorläufer von 33 führen zu können. Die Versuche zur säurekatalysierten Umlagerung der Ketone 51 und 67 dagegen verliefern erfolgreich. Unter der Einwirkung des stark sauren Ionenaustauschers Nafion R SAC-13 lieferten beide Verbindungen das bicyclische Keton 30 und damit die gewünschte Partialstruktur von (±)-Ceratopicanol (32). Das ohnehin nur in geringer Menge vorhandene 27 verharzte. Nach Bicyclobutyliden (4) und 2,2-Dimethylbicyclobutyliden (26) wurde im dritten Teil der Arbeit mit 40 ein Bicyclobutyliden dargestellt und sequentiell verändert, das bei günstigem Verlauf der abschließenden Umlagerungen mit (±)-endo-Capnellen (39) ein vollständiges Sesquiterpengerüst hätte liefern können. Allerdings zeigte sich hier, daß in komplexeren Fällen selbst das Ergebnis einer Oxaspirohexan-Cyclopentanon-Umlagerung nicht mehr vorhergesagt werden konnte. So lieferte 40 nach Epoxidierung durch Bortrifluorid-katalysierte Umlagerung neben den stereoisomeren Cyclopentanonen 87 und 88 das hier unerwünschte Cyclopentanon 93 als Hauptprodukt. Die Ausgehend von 87 und 88 durchgeführten Transformationen in Form einer erschöpfenden Methylierung gefolgt von einer Methylenierung bzw. einer Addition von Methyllithium verliefen problemlos und lieferten mit 44, 45 und 103 sowie 41, 42 und 104 Vorläufer der tricyclischen Kationen 46 und 107 bzw. 105 und 106. Von diesen Kationen konnte lediglich 46 über eine 1,2-Methylverschiebung gefolgt von einer 1,3-Hydridverschiebung in (±)-endo-Capnellen (39) übergehen. Aber auch dieser Fall trat nicht ein. Zwar lieferte die säurekatalysierte Umlagerung von 44, 45 und 103 unter konkurrierender 1,2-Verschiebung der höher (a) und niedriger substituierten Cyclobutanbindung (b) neben 107 auch 46, doch blieb die erhoffte Bildung von (±)-endo-Capnellen (39) aus. Die beobachteten Produkte waren 118, 119 und 120. Im Gegensatz zu 44, 45 und 103 war bei der säurekatalysierten Umlagerung von 41, 42 und 104 eine Bildung von (±)-endo-Capnellen (39) von vornherein ausgeschlossen. dc.description.abstractengThis dissertation deals with the synthetic potential of sequential transformation of bicyclobutylidene derivatives to bicyclo[3.3.0]octenes. For that reason bicyclobutylidene is first epoxidised and rearranged giving rise to spiro[3.4]octanone, which is subsequently modified via alkylation and/or addition reactions. After a second rearrangement one to three methylated bicyclo[3.3.0]octenes are obtained. In the first part, using the above reaction pattern, bicyclobutylidene (4) yielded the mono to trimethylated bicyclo[3.3.0]octenes 56, 58, 59, 60 and 61 via intermediates spiro[3.4]octan-5-ones 14, 50 and 51 and spiro[3.4]octane-5-oles 21-25. The aim of the second part was the synthesis of tetramethylated bicyclo[3.3.0]octene 31 and the bicyclic ketone 30, which present partial structures of (±)-ptychanolid (33) and (±)-ceratopicanol (32). Synthesis and sequential transformation of 2,2-dimethylbicyclobutylidene (26) was just partially successful. Reaction of 2,2-dimethylcyclobutanone 29 with cyclobutylidene-triphenylphosphorane yielded besides expected 26 the condensation products 62 and 63. Epoxidation of 26 yielded oxaspirohexane 64, which did not open regiospecifically and formed a mixture of the required spiroketone 27 and the isomeric ketone 67. Experiments for the selective synthesis of 27 and 67 via hydroxyselenides 49 and 76 were more successful, but in the case of 27 not enough material was obtained to continue the planned synthesis of spiro[3.4]octane-5-ole 28 and rearrangement to 31 as a partial structure of 33. The acid catalysed rearrangement of the ketones 51 and 67 by treatment with Nafion R SAC-13 yielded in both cases the bicyclic ketone 30 as the expected partial structure of (±)-ceratopicanol (32). Acid-catalysed rearrangement of 27 failed. Goal of the third part was the synthesis and sequential transformation of bicyclic bicyclobutylidene 40 and final rearrangement to (±)-endo-capnellene (39), a complete sesquiterpene. Unfortunately, in this complex case the product distribution of the oxaspirohexane-cyclopentanone-rearrangement was not predictable. Epoxidation of 40 and rearrangement of the resulting oxaspirohexanes 89 and 92 with boron trifluoride yielded the unexpected cyclopentanone 93 as the major product beside the expected stereoisomeric ketones 87 and 88. Successful dimethylation of 87 and 88 followed either by reaction with methyene-triphenylphosphorane or addition of methyllithium yielded 44, 45 and 103 as well as 41, 42 and 104, which are potential precursors for the tricyclic intermediate cationes 46 and 107 as well as 105 and 106. Only cation 46 would be able to form (±)-endo-capnellene (39) via 1,2-methylshift followed by a 1,3-hydridshift. Actually, the acid-catalysed rearrangement of 44, 45 and 103 yielded a mixture of isomeric products 118, 119 and 120 by 1,2-shift of the higher as well as the lower substituted cyclobutane bond. However, the formation of (±)-endo-capnellene (39) failed. From a stereochemical point of view, the formation of 39 by acid catalysed rearrangement of 41, 42 and 104 was impossible. The products 110, 111 and 112 were formed.de
