dc.description.abstractIn der vorliegenden Studie wurden Expressions- und Funktionsanalysen an murinem Brunol1 und Brunol4 durchgeführt. Murines Brunol1 ist ein neues Mausgen, welches zur elav/Bruno Familie gehört, die für RNA-bindende Proteine codieren. Die Expression von Brunol1 beginnt an Tag 9.5 dpc und setzt sich in späteren Stadien der embryonalen Entwicklung fort. In adulten Mäusen ist die Expression von Brunol1 auf Gehirn und Testis beschränkt. Weiterhin konnte eine cytoplasmatische und eine Kernlokalisation von Brunol1 mit Hilfe eines GFP-Fusionsproteins nachgewiesen werden. Für Funktionsanalysen des Brunol1 Proteins wurde der Brunol1 Lokus der Maus durch homologe Rekombination zerstört. Männliche und weibliche Brunol1+/- und Brunol1-/- Mäuse aus Hybrid-Verpaarungen (C57BL/6J x 129/Sv ) und inbred-Verpaarungen (129X1/SvJ) zeigten einen normalen Phänotyp und waren fertil, obwohl die Anzahl und die Motilität der Spermien signifikant reduziert waren. Eine intensive phänotypische Analyse zeigte keine Abnormalitäten in der Testismorphologie. Zusammenfassend zeigen die Ergebnisse, dass Brunol1 nicht essentiell für die embryonale und neuronaleEntwicklung und die Spermatogenese der Maus ist.Die Expression und Funktion von Brunol4 wurde in ähnlicher Weise wie bei Brunol1 untersucht. Durch RT-PCR konnte eine Expression von Brunol4 in Gehirn, Ovar und allen Stadien der embryonalen Entwicklung, inclusive Präimplantations-Stadien, nachgewiesen werden. Durch Northern Blot Versuche von adulten Geweben konnte eine Brunol4 Expression im Gehirn nachgewiesen werden. Für Funktionsanalysen wurde eine knockout Maus für Brunol4 generiert. Während heterozygote Brunol4-Mäuse gesund und fertil waren, konnten keine homozygoten Brunol4-Mäuse identifiziert werden. Um das Stadium herauszufinden, indem die Entwicklung der homozygoten Tiere arretiert, wurde eine detailierte Analyse von Embryonen verschiedener Entwicklungsstadien, inclusive Einzell-Stadium, durchgeführt. Es wurde keine homozygote Brunol4-Mutante gefunden. Um homozygote Brunol4 Mäuse zu identifizieren, wurde daraufhin eine Verpaarungsstrategie angewandt. Fünfzehn Brunol4-Mäuse (die heterozygot genotypisiert worden waren) wurden mit Wildtyp-Mäusen verpaart und die Nachkommen wurden mit Hife von PCR genotypisiert. Bei vier Paaren waren alle Nachkommen heterozygot, dies bedeutet, dass die Brunol4-Mäuse homozygot waren. Die Ergebnisse zeigten deutlich, dass es homozygote unter den heterozygoten Tieren gab, die durch Genotypisierung nicht identifiziert werden konnten. Dies deutet darauf hin, dass es mehr als ein Brunol4 Gen gibt, wobei ein Brunol4 Gen, welches nicht transkribiert wird, deletiert wurde. Ausserdem wurde der Zeitpunkt der Aktivierung von Brunol4 in den frühen Stadien der embryonalen Entwicklung untersucht. Die zygotische Aktivierung von Brunol4-Transkripten beginnt im 4-Zellstadium.Weiterhin haben wir Brunol6 als ein neues Mitglied der elav/Bruno Familie identifiziert. Die Expression von Brunol6 beginnt an Tag 9.5dpc und setzt sich während der gesamten Embryonalentwicklung fort, wärend in adulten Mäusen die Expression auf Gehirn und Testis beschränkt ist.de

dc.description.abstractengIn the present study, Expression and functional analysis of murine Brunol1 and Brunol4, were performedMurine Brunol1 is a novel mouse gene related to elav/Bruno family of genes encoding for RNA-binding proteins. Brunol1 expression starts at 9.5 dpc and continues to the later stages of embryonic development. In adult mice, the Brunol1 expression is restricted to brain and testis. Furthermore, we showed a cytoplasmic and nuclear localization of Brunol1 using a GFP fusion protein. For functional analysis of the Brunol1 protein, we disrupted the Brunol1 locus in mouse by homologous recombination. Male and female Brunol1+/- and Brunol1-/- mice from hybrid C57BL/6J x 129/Sv and inbred 129X1/SvJ genetic backgrounds exhibit normal phenotype and were fertile, although number and motility of sperm were significantly reduced. An intensive phenotypic analysis showed no gross abnormalities in testis morphology. Collectively our results demonstrate that Brunol1 may not be essential for embryonic viability, spermatogenesis and development of neural tissue.Similarly like in the case of Brunol1, expression and function of Brunol4 was analysed. By RT-PCR, the expression of Brunol4 was observed in brain, ovary and at all the stages of embryonic development including preimplantation and postimplantation stages. By Northern blot, Brunol4 expression was found to be restricted to brain only. For the functional analysis, knockout mice for Brunol4 were generated. While Brunol4 heterozygous mice were apparently healthy and fertile, no Brunol4 homozygous mice were identified. To determine the stage when development of homozygous embryos is arrested, a detailed analysis of embryos at different stages of development, including up to one-cell stage embryos was done, but no Brunol4 homozygous mutant was found. To identify the Brunol4 homozygous mice among heterozygous mice breeding strategy was used. Fifteen Brunol4 so-called heterozygous mice were bred with wild type and the offspring of all the fifteen breeding pairs were genotyped by PCR. Four pairs among these fifteen breeding pairs gave all heterozygous offspring and it was then clear that they are Brunol4 homozygous mice Breeding results clearly indicated that there are in fact Brunol4 homozygous mice among heterozygous but they could not be identified by genotyping. So the present study shows that there might be more than one copy of Brunol4 gene and the one, which is not transcribed, was deleted. We also studied the activation of mouse Brunol4 gene during early embryonic stages. Zygotic activation of Brunol4 gene transcript starts at 4-cell stage of embryonic development.We also identified Brunol6 as a novel member of elav/Bruno family. Expression of Brunol6 starts at 9.5 dpc and continue to the later stages of embryonic development while in adult mouse tissues Brunol6 expression is restricted to brain and testis.de
