Transcriptional regulation of the Human Zfm1/Sf1 Gene

dc.description.abstractDas erste Ziel der hier vorgelegten Arbeit war zu untersuchen, ob die Zytokine IL-1β/TNFα und/oder der Wachstumsfaktor PDGF, ähnlich wie in glatten Gefäßmuskelzellen (GMZ) der Ratte, auch in humanen GMZ die Expression des Gens für den transkriptionellen Repressor Zfm1 hemmen und so die Proliferation dieser Zellen induzieren. Tatsächlich zeigte sich, dass IL-1β/TNFα weder Proliferation auslösten, noch die Zfm1-Expression beeinflussten. Allerdings konnten diese Zytokine nach Hemmung der Zfm1-Expression durch ein spezifisches RNAi-Molekül doch die Zellteilung humaner GMZ stimulieren. Im Gegensatz hierzu konnte der Wachstumsfaktor PDGF wie in Rattenzellen beides, Proliferation induzieren und Zfm1-Expression hemmen. Diese Daten zeigen, dass Zfm1-Repression notwendig ist, um hohe Proliferationsraten in GMZ aufrecht zu erhalten und geben Hinweis, dass die Regulation der globalen Transkription durch Zfm1 auch beim Menschen den zellulären Phänotyp (mit-) bestimmt.Weiterführende Experimente mit Decoy-Oligonukleotiden (dODN) gegen die Transkriptionsfaktoren EGR-1 und SP-1 zeigten, dass beide Proteine be ider Expression von Zfm1 eine Rolle spielen; während SP-1 eine hohe basale Expression vermittelt, spielt EGR-1 eine ungewöhnliche Rolle. Im Falle des Zfm1-Gens führt Bindung der Faktors an den Promoter nicht zu einer Erhöhung der Expression, sondern zu einer deutlichen Hemmung. Um die Bindungsstellen von EGR-1 und SP-1 auf dem Promoter zu definieren, wurde der humane Genpromoter kloniert und in den Reportergenvektor pGL2 kloniert. Dieses Konstrukt führte, gemeinsam mit gezielten Deletionskonstrukten zur Definition eines Komplexes von Bindungsstellen für EGR-1 und SP-1 etwa 200 bp vor dem Nukleotid +1 (ATG) des Zfm1-Gens. Schließlich konnte mit Hilfe eines Koimmun-präzipitationsassays der Mechanismus der PDGF-induzierten EGR-1-ver-mittelten Hemmung der Zfm1-Expression gezeigt werden. Während unter basalen Bedingungen (wahrscheinlich 2 Moleküle) SP-1 (Bindung -191 bis -174) an den Promoter binden, verdrängt EGR-1 bei Aktivierung diese Proteine und führt so zu einer verminderten Expression der Gens.Parallel zu diesen Arbeiten wurden, um ein besseres Verständnis der Mechanismen der Zfm1-1-vermittelten Transkriptionskontrolle zu gewinnen, Ko-Immunpräzipitationen von Kernproteinkomplexen durchgeführt. Hierbei konnte gezeigt werden, das Zfm1 mit mindestens zwei Komponenten des basalen Transkriptionsfaktors TFIID assoziiert ist, namentlich hTAFII68 und hTAFII80. Zusammengefasst zeigen diese Ergebnisse, das Zfm1 seine Rolle als transkriptioneller Repressor auf der Ebene der basalen Transkriptions-maschinerie ausübt und dass seine EGR-1-vermittelte Repression wichtig für eine hohe Proliferationsrate ist. ein vertieftes Verständnis der Rolle von Zfm1 bei der Phänotypregulation humaner GMZ in arteriosklerotischen Gefäßen könnte in der Zukunft zur Formulierung eines therapeutischen Konzepts führen um die Bildung der Neointima in betroffenen Gefäßen zu verlangsamen oder zu verhindern.de

dc.description.abstractengThe first aim of the present study was to elucidate whether IL-1β/TNFα and/or PDGF down-regulate Zfm1 gene expression and induce proliferation also in human SMC. The results revealed that IL-1β/TNFα alone is not sufficient to induce cell proliferation, but only after down-regulation of the Zfm1 gene through siRNA treatment proliferation was increased. In contrast, PDGF alone maximally induced cell proliferation. Thus, it was investigated whether PDGF directly down-regulates the endogenous Zfm1 gene in order to induce cell proliferation. The findings presented here revealed that PDGF does down-regulate endogenous Zfm1 gene expression, suggesting that this decrease in Zfm1 abundance may be important to allow PDGF induced growth-promoting gene transcription.In order to understand the mechanism of PDGF induced down-regulation of Zfm1 expression in hSMC, the promotor of the human gene was cloned for the first time. Zfm1 promotor reporter gene analysis confirmed that PDGF down-regulates Zfm1 gene expression in hSMC.Experiments employing decoy ODN directed against the transcription factors Egr-1 and SP-1 revealed that both factors are involved in the regulation of the Zfm1 gene; while Egr-1 is the transcription factor responsible for PDGF-induced down-regulation of Zfm1, SP-1 is important for maintaining basal expression of Zfm1. In order to define the exact binding sites and transcription factors important for basal expression and PDGF-induced down-regulation of the Zfm1 gene, deletion constructs of a combined Egr-1/SP-1 consensus site (situated approximately 200 bp upstream of the transcriptional start site) of the Zfm1 promotor were generated. Together with a paralleled chromosome immunoprecipitation analysis, data obtained confirmed that under basal conditions, SP-1 predominantly interacts with the complex binding site located at position -191 to -174 of the Zfm1 gene and maintains the basal expression of Zfm1. Interestingly, under PDGF stimulation, Egr-1 activity was up-regulated, whilst SP-1 activity was decreased, confirming that under PDGF stimulation, Egr-1 displaces SP-1 from the binding site.Finally, analysis of Zfm1 protein-protein interactions through co-immunoprecipitation confirmed that Zfm1 interacts with both hTAFII68 and hTAFII80, suggesting that Zfm1 plays a role as a regulator of SMC phenotype at the transcriptional level.In summary, these findings reveal that Zfm1 is a repressor of growth promoting gene transcription possibly through modulating the transcriptome architecture via specific Zfm1-TAF interactions. Zfm1 expression is down-regulated by PDGF through the displacement of SP-1 by Egr-1. A deepened understanding of SMC proliferation in atherosclerosis and the role of Zfm1 therein, may lead to new therapeutic strategies involving the stabilization of a contractile and non-proliferating SMC phenotype, in order to prevent or slow down neo-intima formation in the course of the disease.de
