dc.description.abstract | BI-1 (Bax-Inhibitor-1) konnte in Hefen, Pflanzen und verschiedenen Säugetierzellen vorwiegend als anti-apoptotisches Protein charakterisiert werden.Um die Funktion von BI-1 in Neuronen zu untersuchen, wurde sowohl HA- als auch GFP-getaggtes BI-1 transient und stabil in CSM14.1- (nigro-striatale Rattenzelllinie) und SH-SY5Y-Zellen (humane Neuroblastomzelllinie) überexprimiert.Überraschenderweise zeigte sich nach Bax-induzierter Apoptose kein verbessertes Überleben von transient BI-1-exprimierenden Zellen. Im Gegensatz dazu konnte nach stabiler Überexpression von BI-1, wobei sich ein gleichmäßiges BI-1-Expressionsmuster zeigte, ein neuroprotektiver Effekt abhängig vom Zelltodstimulus belegt werden. BI-1 konnte in beiden neuronalen Zelllinien vor ER-Stress durch den Ca2+-ATPase-Inhibitor Thapsigargin schützen, während Mitochondrien-assoziierter Zelltod, ausgelöst durch den Kinaseinhibitor Staurosporin nicht inhibiert werden konnte. Außerdem zeigte sich im Rahmen eines Modells zur Induktion eines Ischämie-ähnlichen Zustands durch den Entzug von Sauerstoff und Glukose bzw. Serum ebenfalls ein neuroprotektiver Effekt von BI-1.Eine Betrachtung der subzellulären Lokalisation von BI-1 hat zusätzlich ergeben, dass BI-1 vorwiegend mit dem ER und dem perinukleären Randsaum, jedoch nicht mit den Mitochondrien assoziiert ist.Zusammenfassend zeigt sich, dass eine im ER lokalisierbare BI-1-Expression auf niedrigem Niveau neuroprotektiv gegenüber ER-assoziiertem Zelltod und Ischämie wirkt. BI-1 ist somit ein interessantes Kandidatengen für die Therapie von neurodegenerativen Erkrankungen und Schlaganfällen. In der Literatur finden sich Hinweise auf eine Beteiligung von BI-1 an Prozessen der Ca2+-Freisetzung aus dem ER. Weitere Studien mit dem Ziel, den Signaltransduktionsweg von BI-1 in Neuronen aufzudecken, der ursächlich für die beobachtete ER-assoziierte Neuroprotektivität ist, sind somit notwendig. | de |

dc.description.abstracteng | BI-1 (Bax-Inhibitor-1) has been characterized as anti-apoptotic protein in yeast, plants and various mammalian cell lines.To explore the function of BI-1 in neurons, HA- as well as GFP-tagged BI-1 was transiently and stably overexpressed in rat nigro-striatal CSM14.1 and human SH-SY5Y neuroblastoma cells.Surprisingly, increased survival of transiently BI-1 overexpressing cells could not be observed for bax-induced apoptosis. In contrast, after stable overexpression of BI-1 on a moderate level, showing a smooth expression pattern, a neuroprotective effect depending on the cell death stimulus could be documented. In both neuronal cell lines BI-1 protected against ER-stress induced by thapsigargine, blocking the Ca2+-ATPase of the ER, but failed to inhibit mitochondria-associated cell death induced by the kinase inhibitor staurosporine. Moreover, a neuroprotective effect of BI-1 could be observed in a model mimicking ischemia-like conditions, namely oxygen-glucose or serum deprivation.In addition, examination of the subcellular BI-1-localization revealed that BI-1 is predominantly associated with the ER and the nuclear envelope but not with mitochondria.Taken together, BI-1-expression located to the ER on a moderate level is neuroprotective towards ER-associated cell death and ischemia. Thus, BI-1 appears as an interesting therapeutic target in neurodegenerative diseases and stroke. There is increasing evidence for BI-1 being involved in processes of Ca2+-release from the ER. Future experiments may aim at the discovery of the signal transduction pathway of BI-1 in neurons underlying the observed ER-associated neuroprotectivity. | de |
