dc.description.abstractDer Ryanodin-Rezeptor (RyR) als größter bekannter Ionenkanal der Herzmuskelzelle gilt als Multiproteinkomplex. Durch die Interaktion mit Proteinen wie z.B. Kinasen und Phosphatasen wird er in seiner Aktivität gesteuert. Durch die Bindung von FKBP12.6 an den RyR2, die Isoform der Herzmuskelzelle, wird die Offenwahrscheinlichkeit des Kanals und damit der Calciumverlust in der Diastole herabgesetzt. Die durch FKBP12.6 koordiniertere Öffnung benachbarter RyR (coupled gating) bedingt zudem eine verbesserte relative Zellverkürzung der Zelle. Da die genaue Anzahl bzw. Funktion aller Bindungspartner am RyR derzeit noch nicht abschließend geklärt ist, liegt die Vermutung nahe, dass die Wirkung von FKBP durch andere Bindungspartner beeinflusst wird. Es ist bekannt, dass FKBP12.6 die Proteinphosphatase Calcineurin bindet und FK506-abhängig inhibiert. In dieser Arbeit wurde erstmals eine Calcineurin-bindungsdefiziente Mutante (FKBP12.6G89PV90K )erzeugt, um die funktionelle Bedeutung der FKBP12.6-Calcineurin-Interaktion zu untersuchen. Es konnte nachgewiesen werden, dass FKBP12.6G89PV90K Calcineurin bei erhaltener endogener Rotamaseaktivität weder binden noch inhibieren kann. Durch adenoviral vermittelte Überexpression in isolierten Kardiomyozyten adulter Kaninchen wurde die Auswirkung auf die Kontraktilität der Zellen untersucht. Bei nur gering ausgeprägter Expression der Mutante (MOI 10) fand sich kein Unterschied zu kontrolltransfizierten Myozyten, bei einer MOI 50 hingegen eine negative Inotropie der mit FKBP12.6G89PV90K-transfizierten Zellen. Im Vergleich zu FKBP12.6-überexprimierenden Zellen, die eine positive Inotropie zeigen, liegt so die Vermutung nahe, dass die bekannten Effekte von FKBP12.6 am RyR zum Teil durch Calcineurin vermittelt werden.de

dc.description.abstractengThe ryanodin receptor, the largest known ion channel of the heart muscle cell is known as a multi protein complex. It´s activity is regulated by proteins like kinases and phosphateses. The bond of FKBP12.6 to RyR2, the isoform of the heart cell, causes decreased open probability of the channel and therefore reduced diastolic calcium loss. The FKBP12.6 mediated enhanced coordination of neighboured RyR (coupled gating) leads to improved relative cell shortening. Since the definitive number and function of all binding partners of RyR is not fully known at this moment, it is has been speculated, that the effect of FKBP12.6 is mediated by other ligands of the channel. It is known, that FKBP12.6 binds to and inhibits Calcineurin, a protein phosphatase. To examine the functional relationship between both a FKBP12.6 mutant (FKBP12.6G89PV90K ) has been generated first time in this work. It has been proved that FKBP12.6G89PV90K can´t bind or inhibit Calcineurin while endogene rotamase activity is unchanged. Through adenoviral mediated gene transfer in isolated heart muscle cells of adult rabbits the effect on contractility has been examined. When the mutant is slightly overexpressed (MOI10) there is no difference in comparison to control transfected myocytes whereas cells overexpressing FKBP12.6G89PV90K (MOI 50) show a negative inotropy. In comparison to FKBP12.6 transfected myocytes, that show positive inotropy, it is likely, that the known effects of FKBP12.6 on RyR are partially calcineurin mediated.de
