dc.description.abstractDer Hefepilz C. glabrata ist ein opportunistischer humanpathogener Organismus, der die zweithäufigste Ursache für systemische Candidosen bei immunsupprimierten Patienten darstellt. Typische Charakteristika des Erregers sind neben einer intrinsischen Resistenz gegenüber Azolantimykotika, die sehr dynamische Zellwandstruktur und eine hohe genomische Flexibilität. Es wird angenommen, dass diese Eigenschaften es C. glabrata ermöglichen, sich in verschiedenen Wirtsnischen perfekt an das umgebende Millieu anzupassen. Darüber hinaus könnten beide Faktoren wichtige Einflußgrößen für die Virulenz und Resistenz des Erregers sein. Um eine Aussage über die tatsächliche Bedeutung dieser Mechanismen sowohl für die Entstehung und Therapie von Candidosen, als auch für die Interpretation wissenschaftlicher Daten treffen zu können, wurden zwei C. glabrata-Kollektive untersucht. Das erste Kollektiv bestand aus in der Multilokus-Sequenztypisierung (MLST) isogenen Klonen des Labor-Referenzstamms ATCC2001/CBS138, das andere Kollektiv aus 124 heterogenen klinischen Isolaten, die aus unterschiedlichen europäischen Forschungs- und Diagnostikzentren stammten. Die Charakterisierung beider Gruppen erfolgte genotypisch mittels Pulsfeld-Gelelktrophorese (PFGE) und MLST, phänotypisch durch standardisierte Mikrodilutionstests auf Zellwand-destabilisierende und antimykotisch wirksame Substanzen und Zellwand-biochemisch, durch die quantitative Analyse des Chitin- und Glukangehalts mittels Durchflusszytometrie. Innerhalb des Laborstamm-Kollektivs konnten drei unterschiedliche Karyotyp-Gruppen identifiziert werden, die jeweils mit spezifischen Phänotypen und einer charakteristischen Zellwandkomposition assoziiert waren. Die klinischen Isolate zeigten in der PFGE insgesamt 32 unterschiedliche Karyoytpen und 9 Haupt-Kladen in der MLST. Anhand des elektrophoretischen Karyotyps der Isolate konnte eine Einteilung der Isolate in zwei Hauptgruppen vorgenommen werden, die ebenfalls mit charakteristischen Sequenztypen in der MLST einhergingen. Im Gegensatz zu den Laborstämmen zeigten phylogenetisch eng verwandte klinische Isolate desselben Karyotyps größtenteils eine breite Variabilität bezüglich phänotypischer Charakteristika und in der Zellwandzusammensetzung. Jedoch konnte auch eine MLST-Klade identifiziert werden, deren Isolate alle eine charakteristische chromosomale Aberration in der mittleren Bandenfraktion in der PFGE besaßen. Des Weiteren zeigten die Isolate dieser Klade eine vergleichsweise reduzierte Suszeptibilität gegenüber Fluconazol und Zellwand-destabilisierenden Substanzen. Alle durchflusszytometrisch analysierten Isolate dieser Klade verfügten darüber hinaus über einen erhöhten Glukangehalt der Zellwand. Diese Ergebnisse deuten an, dass komplexe Zusammenhänge zwischen geno- und phänotypischen Charakteristika, sowie einer spezifischen Zellwandzusammensetzung bestehen. Die aufgezeigten Abhängigkeiten stellen somit die Grundlage für neue Erklärungsmodelle zum Verständnis der fungalen Virulenz, der Entstehung von Resistenz, und von essentiellen Adaptionsmechanismen von klinischen C. glabrata-Stämmen dar. Die nachgewiesene hohe genetische Variabilität und die resultierende signifikante Veränderung verschiedener Phänotypen der untersuchten isogenen Laborstämme macht außerdem deutlich, mit welch großer Vorsicht die Interpretation wissenschaftlicher Daten von erzeugten Deletionsmutanten erfolgen muss.de

dc.description.abstractengCandida glabrata is an opportunistic fungal pathogen and the second leading cause of systemic candidosis in immunocompromised patients after Candida albicans. Its intrinsic resistance to clinically used azol antimycotics, the very dynamic cell wall structure and its highly flexible genome, conferring adaptive plasticity when invading the human host, renders C. glabrata a fungal pathogen of rising importance. To investigate possible relationships between genotype, phenotyp and the biochemical structure of the cell wall and possible consequences for molecular biological research as well as for clinical antimycotic therapy regimen and diagnostic tools, we established two C. glabrata collectives. One containing 10 isogenic typed laboratory reference strains and the other consisting of 124 heterogenous clinical Isolates, obtained from various european hospitals. Both groups were characterized phenotypically by using standardized broth microdilution testing towards cell wall perturbing and antimycotic agents, genotypically via electrophoretic karyotyping and MLST and in terms of the quantitative biochemical composition of the cell wall using FACS analysis. The isogenic laboratory strains showed three groups, each displaying a characteristic karyo- and phenotype, as well as a specific cell wall composition. The clinical isolates displayed a broad heterogenicity in every analysed aspect. We could distinguish 32 different Karyotypes, 9 major MLST clades and observed a phenotypic division in three main categories. Unlike the laboratory strains, isogenic clinical isolates of the same karyotype showed a rather wide variability in terms of phenotypic characteristics and cell wall composition. Nevertheless, one MLST clade consisted mostly of fluconazole hyposusceptible isolates, which shared a specific chromosomal aberration and an elevated glucan content of the cell wall. Additionally, most isolates clustered in this clade showed hyposusceptibility towards the cell wall perturbing agents blancophor and congo red. Interesting ly, the characteristic chromosomal alteration of these isolates could be found more frequently among all hyposusceptible phenotypes than other ones. This data strongly suggests, that there could be a linkage between geno- and phenotypic characteristics, as well as a spezific cell wall composition, that might contribute to fungal resitance, adaptability and virulence. Further Investigation is needed, to confirm this hypothesis on a molecular level regarding involved gene activity in possible underlying mechanisms. The Observation, that aneuploidy of isogenic typed clones of the laboratory reference strain CBS138/ATCC2001 led to significant differencies in phenotypes to common stress tests and in the cell wall composition has a huge impact on the C. glabrata research community. Before and during production of C. glabrata deletion mutants and revertants the use of the standard CBS138/ATCC2001 background karyotype needs to be confirmed either by using PFGE or comparable methods like CGH and Microarrays.de
