Untersuchungen zur Kombinationswirkung von Valproat und Bestrahlung in Bezug auf klonogenes Überleben, Apoptose, Polyploidie und chromosomale Instabilität in Prostatakarzinom- und Prostatahyperplasie-Zelllinien

dc.description.abstractValproat kann als Histondeacetylase-Inhibitor einen starken Antitumoreffekt auf Prostata-Karzinom-Zelllinien ausüben. Dieser Effekt ist jedoch nach Entfernung des Valproats wieder reversibel. Andere Histondeacetylase-Inhibitoren konnten in vitro einen radiosensibilisierenden Effekt auf andere Tumorzellen auslösen. Die Kombinationswirkung von Valproat und Bestrahlung auf Prostata-Karzinom- und Prostata-Hyperplasie-Zelllinien wurde in dieser Arbeit anhand des klonogenen Überlebens untersucht. Es zeigte sich eine deutliche radiosensibilisierende Wirkung bei den Prostata-Karzinom-Zelllinien (PC-3 und DU-145) und keine Radiosensibilisierung bei den Prostata-Hyperplasie-Zelllinien (BPH-1, PTN-2). Die Ursache dieses Effektes konnte nicht abschließend geklärt werden. Die DU-145-Zellen zeigten eine Erhöhung der Apoptose-Rate durch diese Kombinationsbehandlung, die PC-3-Zellen in diesem Versuchsaufbau jedoch nicht. Eine vermehrte Polyploidie oder eine chromosomale Instabilität ließen sich bei den Prostata-Karzinom-Zelllinien nicht nachweisen.de

dc.description.abstractengValproate, as a histone deacetylase inhibitor, has a strong antitumor effect on prostate cancer cells. This effect is reversible after removal of valproate. Other histone deacetylase inhibitors could induce an increased radiosensibility in other tomor cells. Thus we studied the combined effect of valproate and radiation on prostate carcinoma cells (PC-3, DU-145) and prostate hyperplasia cells (BPH-1, PTN-2) using clonogenic survival assays. Valproate could radiosensitize the prostate carcinoma cells but not the benign prostate hyperplasia cells. The cause of this effect could not be fully clarified. The combination of valproate and radiation caused an increase of the aoptotic rate in the DU-145-cells, but not in the PC-3-cells in this experimental setup. An increase in poliploidity or chromosomal instability could not be detected in the prostate carcinoma cells after the combined treatment with valproate and radiation.de
