dc.description.abstractIn dieser Arbeit wird die Synthese und Charakterisierung neuer zweikerniger Zinkkomplexe vorgestellt, welche die aktiven Zentren von Dizink-Metallohydrolasen nachbilden. Ein Pyrazol-basierter Ligand, welcher in Positionen 3 und 5 mit chelatisierenden Seitenarmen substituiert ist, dient dabei zur Nachahmung der dreidimensionalen Umgebung des Proteingerüstes der Enzyme. Als Modelle für Histidin werden aromatische Donorgruppen wie Imidazol oder Benzimidazol in den Seitenarmen verwendet. Zusätzlich werden Carboxylatgruppen genutzt, um Aspartat nachzubilden. Die synthetischen Modelle der aktiven Zentren wurden durch Komplexierung der dargestellten Liganden von Zink erhalten. Im Verlauf der Komplexsynthese konnte eine umfangreiche Untersuchung von Komplexen, welche auf dem Ligand mit chelatisierenden Imidazol-Seitenarmen basieren, erforscht werden. Zur Untersuchung der Koordinationschemie dieses Liganden wurde die Methode der potentiometrischen Titration eingesetzt. Dabei wurden in Abhängigkeit des pH-Wertes sechs verschiedene Spezies in Lösung bestimmt, wobei vier dieser Komplexe in festem Zustand isoliert und durch Röntgenkristallstrukturanalyse untersucht wurden. Geeignete Komplexe, welche die aktiven Zentren der Enzyme nachbilden, wurden in ihrer katalytischen Aktivität hinsichtlich der Spaltung von β-Lactam-Antibiotika (Penicillin G) und Phosphatestern untersucht. Weiterhin wurde die Aktivierung von Kohlenstoffdioxid in der CO2-Copolymerisierung und in der Darstellung von zyklischen Carbonaten erforscht. Bei den Untersuchungen zur Hydrolyse-Aktivität von β-Lactam-Antibiotika wurde außerdem die Koordination von verschiedenen Substraten an die Komplexe analysiert. Übereinstimmend mit den Ergebnissen aus NMR- und IR-spektroskopischen Untersuchungen wurde ein Komplex/Substrat Addukt erhalten und mittels Röntgenkristallstrukturanalyse charakterisiert. Diese Struktur weist eine bidentate Koordination der Carboxylatgruppe des Substrates an beide Zinkatome des Komplexes auf, was zur Hemmung der Hydrolyse-Aktivität des Komplexes führt. Resultierend aus diesen Ergebnissen können nun neue bioinspirierte Ligand-Systeme entwickelt werden, um andere zweikernige Komplexe darzustellen, welche eine erhöhte Aktivität in der β-Lactam-Antibiotika Spaltung aufweisen. Dadurch können die Spaltungsmechanismen der Substrate näher untersucht werden, was einen wichtigen Aspekt in der Erforschung der Mechanismen der Enzyme darstellen könnte.de

dc.description.abstractengThis work presents the synthesis and characterisation of new binuclear zinc complexes that mimic the active sites of dizinc metallohydrolases. To emulate the three-dimensional protein scaffolds of these enzymes, a pyrazole based ligand is used that is substituted in positions 3 and 5 with chelating side chains. Bearing aromatic donor groups, side chains containing imidazole and benzimidazole moieties are a common motif to mimic histidine. In addition, carboxylate groups are used to emulate aspartate. Complexation of the designed ligands with zinc leads to the synthetic analogues of the active sites of the above mentioned enzymes. In the course of complex synthesis, a comprehensive investigation of complexes based on the ligand that bears imidazole as chelating side chains is reported. The coordination chemistry of this ligand is studied by potentiometric titrations. Depending on the pH, six different species are determined in solution and four of them are isolated in solid state and characterised by X-ray diffraction. Suitable complexes that mimic the active sites of dizinc metallohydrolases are investigated in their hydrolytic activity to cleave β-lactam antibiotics (i.e., Penicillin G) and phosphate esters. In addition to this, the activation of carbon dioxide is studied in CO2 copolymerisation and in the formation of cyclic carbonates. Within the investigations on β-lactam substrates, the binding mode of various substrates to the complex is explored. In accordance to the results obtained by NMR and IR spectroscopy, one complex/substrate adduct is analysed by X-ray diffraction showing a bidentate coordination of the carboxylate group of the substrate to both zinc atoms of the complex. This binding fashion inhibits the activity of the complex in hydrolytic cleavage reactions. Based on these findings, new bioinspired ligand scaffolds can be developed to enhance the hydrolytic activity of the complexes. In this way the mechanism of the cleavage reaction of the substrates can be investigated to gain insight into the mechanistic pathways of the enzymes.de
