dc.description.abstractIm Unterschied zu den von anderen Gruppen entwickelten vielfältigen synthetischen Zugängen zu zahlreichen biologisch aktiven Peptidomimetika, gehen wir jeweils nur von einem einzigen gut zugänglichen Baustein aus, an den mittels einiger synthetischer Transformationen nahezu beliebige pharmakophore Information angebracht werden kann und der darum zum Einfrieren wichtiger Peptidsequenz-Motive genutzt werden kann. Dies kann die zeitintensive Entwicklung jeweils neuer peptidischer Leitstrukturen wesentlich erleichtern. Um dies zu erreichen, haben wir einen einfachen und ökonomisch sinnvollen Syntheseweg zum 2-Chlor-2-cyclopropylidenacetat und seinen Derivaten eröffnet, ferner zu einem Aminocyclopropylidenacetat, das synthetisch ähnlich vielseitig wie das 2-Chlor-2-cyclopropylidenacetat ist, jedoch den Bedürfnissen der Peptidchemie besser entgegenkommt. Von diesen Bausteinen ausgehend haben wir einen universellen Zugang zu 2-Oxopiperazinmotiven und anderen heterocyclischen Grundkörpern gefunden und in diese biologisch relevante kurze Peptidsequenzen, z. B. die RGD-Sequenz oder die inverse RGD-Sequenz (DGR) eingebaut.de

dc.description.abstractengIn contrast to the various synthetic accesses to biologically active peptidomimetics developed by other groups, we start from a single readily available class of building blocks. As in a few synthetic steps various pharmacophoric information can be added to these starting materials, they seem to be ideal to freeze important peptide-motifs. This can extremely facilitate the time-demanding development of new peptidic leads. To achieve this goal, we open a simple and economical way to methyl 2-chloro-2-cyclopropylideneacetate and its derivatives and to an aminocyclopropylideneacetate, which suits better to the demands of peptide-chemistry, but is synthetically as versatile as methyl 2-chloro-2-cyclopropylideneacetate is. Starting from these building-blocks, we paved synthetic ways to 2-oxopiperazinone-motifs and other heterocyclic scaffolds and incorporated in these biologically relevant short peptide-sequences, such as the RGD-sequence or the inverse RGD-sequence (DGR).de
