Several differently substituted monocyclic 3-spirocyclopropanated β-lactams, formally derived from α-cyclopropyl-modified β-alanine, have been prepared through the rearrangement of 4,5-dispirocyclopropanated isoxazolidines under acidic conditions. The latter had been obtained by means of 1,3-dipolar cycloadditions of acyclic nitrones on bicyclopropylidene. Furthermore, a new one-pot procedure for the synthesis of the same kind of β-lactam derivatives has been developed and optimized applying microwave heating. In particular, azaspiro[2.3]hexanones were prepared by a three-component cascade reaction, in which a N-alkylhydroxylamine hydrochloride, formaldehyde or an alkyl glyoxylate and bicyclopropylidene yielded the final products in the presence of sodium acetate. Some azaspiro[2.3]hexanones, protected at the nitrogen atom with electron-withdrawing groups, were coupled with tert-butyl glycinate and/or tert-butyl (S)-phenylalaninate by N-nucleophilic ring-opening of the azetidinone core to obtain dipeptides, in which the presence of the small ring, adjacent to the carbonyl function, introduces a conformational lock. N-Deprotected 3-spirocyclopropanated β-lactams were used to obtain spirocyclopropanated poly-β-propiolactams, applying an anionic polymerization protocol.de
