dc.description.abstractIn der vorliegenden Arbeit werden Modellsynthesen und Strukturaufklärungen von Polyketiden und Arbeiten zur Aufklärung der Biosynthese von Mimosin durchgeführt. Die Dissertation gliedert sich dabei in drei Teile. Der erste Teil beschäftigt sich mit Modellsynthesen von Polyketiden mit deren Hilfe versucht wurde Vorhersagen über Cyclisierungsarten zu machen. Es konnte gezeigt werden, daß die Cyclisierung zu Hemiketalen stärker bevorzugt wurde als die der Pyronringe. Der zweite Teil der Arbeit behandelt den in einem Screening auffällig gewordenen Stamm Streptomyces sp.106. Man kultivierte den Bakterienstamm, isolierte die Metaboliten aus den Extrakten und klärte deren Strukturen mit spektroskopischen Methoden auf. Es stellte sich heraus, daß es sich bei den Produkten um Vertreter der Substanzklasse der Makropolyene handelt. Die beiden Hauptkomponenten konnten als das neue Makropolyen (32) und Rimocidin (33) identifiziert werden, welche sich in den Seitenketten unterscheiden. Im dritten Teil wurde zur Aufklärung der Biosynthese von Mimosin, in Leucaena leucocephala, Einbauversuche mit einem hypothetischen Biosynthesevorläufer, DL-Lysin, in isotopenmarkierter Form durchgeführt. Bei den Fütterungen mit 13C-markierten Lysin kam es zu einem Einbau in die Seitenkette von Mimosin (40). Für weiterführende Fütterungsversuche mit 18O-markiertem Sauerstoff wurden Pflanzenzellkulturen entwickelt.de

dc.description.abstractengThe theme of the thesis is model syntheses and structure elucidation of polyketides and investigation of the biosynthesis of Mimosin. The thesis is divided thereby into three sections. The first section concerns itself with model syntheses of polyketides. The reason was to forecast the kind of cyclization of open polyketide chains. It could be shown that the cyclization was more strongly preferred to hemiketals than to pyrons-rings. The second section concerns itself with the bacteria Streptomyces sp.106 which showed active compounds in a screening. The work included fermentation of the bacteria, isolation of the metabolites and structure elucidation with spectroscopic methods. It turned out that it is a product representative of the substance class of polyene macrolids. The two main components could be identified as the new polyene macrolid (32) and Rimocidin (33), which differ in the side chain. The third section investigates the biosynthesis of Mimosin. The feeding experiments with 13C- lysine gave an incorporation of a label in the side chain of Mimosin (40). For further experiments with 18O- oxygen plant cell cultures of leucaena leucocephala were developed.de
