dc.description.abstract | Die verholzte Zellwand stellt den höchstentwickelten biologischen Verbundwerkstoff dar, der bekannt ist. Ihre Bestandteile, Phenole und Kohlenhydrate, wurden bereits in der Antike zusammen mit einer Phenoloxidase für die Herstellung von Lackarbeiten benutzt. Um diese Prozesse in industriellem Maßstab zu nutzen, benötigt man kostengünstige Phenole, wie beispielsweise technische Lignine aus der Papierindustrie, welche mit Hilfe des Enzyms Laccase in aktive Radikale überführt werden können. Bei den Untersuchungen wurde mit aktivierten Ligninen gearbeitet, die eine hohe Dichte der Phenoxyradikale aufwiesen. Diese waren reaktiver und reagierten sogar mit Nucleophilen wie Zellulose und Stärke. Diese Reaktion eröffnet neue Möglichkeiten für den biotechnologischen Einsatz von Lignin. Im Rahmen dieser Arbeit wurde die Reaktion zwischen aktiviertem Lignin, wasserlöslicher Ligninsulfonsäure und wasserunlöslichem Indulin, mit Wachsmaisstärke, im Hinblick auf eine Verbesserung der Wasserfestigkeit, untersucht. Zudem wurde der Einfluss der Zugabe eines Mediators auf die Radikaldichte und die Ligninablösung des Holzfasereigenen Lignins untersucht. Während die Ligninablösung und die Polymerisation gebremst wurden hatte der Mediator eine gesteigerte Reaktivität der Faseroberfläche durch eine erhöhte Dichte der Phenoxyradikale bewirkt. In weiteren Versuchen wurden faserverstärkte Kunststoffe aus Zellulosefasern und Acrylmonomeren untersucht. Zunächst wurden Holzfasern mit aktiviertem Ligninsulfonat zur Reaktion gebracht. Danach erfolgte eine Laccase-katalysierte Aufpolymerisation von Acrylamid auf die aktivierte Faseroberfläche. Es konnte gezeigt werden, dass das aufpolymerisierte Acrylamid fest an die Faseroberfläche gebunden wurde und auf dieser eine Polymerschicht bildet. Um zu überprüfen ob die für Lignin gefundenen Ergebnisse auch auf Humus übertragbar sind wurde Huminsäure mit Laccase umgesetzt, wobei eindeutig gezeigt werden konnte, dass eine Polymerisation erfolgte. Mit Hilfe von ESR-Messungen wurden Phenoxyradikale gefunden. Humus verhielt sich in seinen Reaktionen mit Laccase völlig analog zu denen des Lignins. Des Weiteren wurde festgestellt, dass die für die Reaktion zwischen Ligninen und Kohlenhydraten gefundenen Ergebnisse auch auf Lignine und aliphatische Amine sowie Aminosäuren übertragen werden können. Auch reifer Humus selbst ist noch zu diesen Reaktionen fähig. | de |

dc.description.abstracteng | The woody cell wall is the most developed biological composite material existing today. Its components, phenols and carbohydrates, in combination with phenoloxidase were already used in the antique to produce lacquerwork. To allow the use at an industrial scale of these processes today, there must be a cheap phenol, as for example technical lignins produced in the paper industry that can be transformed into active radicals by using the enzyme laccase. This study used activated lignins that had a high density of phenoxy radicals. They were used because they proved to be more active than non-activated lignin and could even react with nucleophiles as cellulose and starch. The chemical properties leading towards this reaction allows for innovative use of lignin in biotechnology. The thesis analysed the reaction of activated lignin, water-soluble lignin and water insoluble indulin with maize starch to allow an improvement of the water solubility. Furthermore, the influence of an addition of mediators on the radical density of wood fibres were studied. While the addition of mediators resulted into an inhibition of both the release of fibre lignin into the incubation medium and the polymerization of the released lignin, an increased reactivity of the surface of the wood fibres could be observed owing to a higher density of the phenoxy radicals. In addition, it was tried to build fibre reinforced polymers out of wood fibres and acrylic monomers. In a first step, a reaction of woody fibres with activated lignin was initiated. In a second step,a laccase catalysed polymerization of acrylamide onto the activated fibre surface was conducted. It could be proven that the polymerized acrylamide was strongly bound to the surface of the fibre, building a polymer layer. In addition, it was shown that activated lignin was reacting the same way with aliphatic amines and amino acids as was shown before for the reaction between lignin and carbohydrates. To find out, if the results observed for lignin could be transferred on humus, humic acid was reacted with Laccase. It could be shown, that a polymerisation took place there as well. The existence of phenoxy radicals could be shown via ESR spectra. Even mature humus showed exactly the same reactions towards laccase as lignin. | de |
