dc.description.abstractChronisch progressive Nierenerkrankungen münden, begleitet durch tubulointerstitielle Fibrose, in eine terminale Niereninsuffizienz. Bedingt durch die Ausschüttung von Chemokinen werden Entzündungszellen rekrutiert, proinflammatorische Mediatoren freigesetzt und darüber interstitielle Fibroblasten zur Matrixsynthese angeregt. Im Rahmen der Fibrogenese gehen Tubulusepithelzellen apoptotisch zu Grunde oder durchlaufen via epithelial-mesenchymaler Transition eine phänotypische Umwandlung zu ECM produzierenden Zellen. Durch tubuläre Atrophie und exzessive Matrixsynthese kommt es zur progressiven Fibrosierung und folglich zu renalem Funktionsverlust. Entscheidend für die Fibrogenese ist die Imbalance zwischen profibrogenen und antifibrogenen Zytokinen. Chronische Schädigungen führen zu einer gesteigerten Expression von TGF-β1, dem potentesten profibrogen wirkenden Zytokin. HGF ist ein endogenes Zytokin, mit renoprotektiven Fähigkeiten. Es besitzt als Antagonist von TGF-β1 ein hohes antifibrotisches Potential.In der vorliegenden Arbeit wurden die Effekte einer HGF-Überexpression an murinen Tubulusepithelzellen (NP1) und kortikalen Fibroblasten (TFB) hinsichtlich der Proliferation, Matrixsynthese, -Degeneration, und Apoptose untersucht. Es konnte gezeigt werden, dass eine HGF-Überexpression die Proliferation proximaler Tubulusepithelzellen und renaler Fibroblasten stimuliert. In den aufgeführten BrdU-Assays steigerte HGF die Proliferation der TFB-Zellen und NP1-Zellen. Anhand von Western Blots und ELISA konnte gezeigt werden, dass eine HGF-Überproduktion die Fibronektinsekretion der TFB-Zellen um bis zu 44% und die der NP1-Zellen bis zu 40% senkt. Zur Ermittlung des Einflusses einer HGF-Überexpression auf die Matrixdegeneration wurde die proteolytische Aktivität der Proteasen MMP-2 und MMP-9 in Zellkulturüberständen zymographisch ermittelt. Bei den NP1-Zellen konnte eine durch HGF-Überexpression gesteigerte Aktivität für MMP-2 (28%) und MMP-9 (32%) nachgewiesen werden. Im Gegensatz dazu zeigte sich bei den HGF-überexpremierenden TFB- Zellen eine verminderte Aktivität sowohl von MMP-2 (13%) als auch MMP-9 (32%). Nach Cyclosporin-A-induzierter Apoptose konnte mittels FACS-Analysen, nach Annexin-V- und PI-Färbung, ein zytoprotektiver Effekt durch HGF- Überexpression nachgewiesen werden. Es konnte für die TFB-Zellen eine 42% geringere und für die NP1-Zellen eine 59% geringere Apoptoserate gegenüber den HGF-negativen Zellen ermittelt werden.Diese Beobachtungen lassen auf ein antifibrotisches Potential durch HGF-Überexpression schließen. dc.description.abstractengChronic renal disease is affected by tubular interstitial fibrosis and lead to end stage renal failure. Mediated by chemokines inflammatory cells are recruited, proinflammatory mediators are released and interstitial fibroblast are stimulated to produce matrix components. Tubular epithel cells undergo apoptosis or via epithelial-mesenchymal transition a phenotypic conversion into extracellular matrix producing cells. Progressive fibrosis caused by tubular atrophy and excessive matrix synthesis lead to loss of renal function. Essential for renal fibrosis is the imbalance between profibotic and antifibrotic cytokines. Chronic renal damage causes the release of TGF-ß1, the most potent profibrotic cytokine. HGF is a cytokine with renal protective properties. As an antagonist of TGF-ß1 it has a high antifibrotic potential. In this work we examined the effects of HGF overexpression in tubular epithelia cells (NP1) and renal fibroblasts (TFB) looking at proliferation, matrixsynthesis, matrixdegeneration and apoptosis. We could demonstrate that HGF overexpression stimulates proliferation. Using BrdU-assays we found that HGF enhances Prolifertationrate in TFB and NP1. With the help of Western blot analysis and ELISA we showed that HGF overexpression reduces Fibronektinsythesis and secretion up to 44% in TFB and up to 40% in NP1. To investigate the effect HGF overexpression regarding matrixdegeneration we used zymography assays to measure the proteolytic activity of MMP-2 and MMP-9. Here we demonstrated that HGF enhances the activity of MMP-2 (28%) and MMP-9 (32%) in NP1. In contrast to TFB cells. Here HGF overexpression lead to diminished activity of MMP-2 (13%) and MMP-9 (32%) compared to the control group. Furthermore we found a cytoprotective effect of HGF overexpression after Cyclosporin-A induced apoptosis. After marking cells with Annexin-V and PI and using FACS analysis we demonstrated a lower rate of apoptosis in the HGF overexpressing NP1 (59%) and TFB (42%) compared with the control group. These findi ngs suggest a antifibrotic potential of HGF overexpression in renal cells . They are interesting for further investigations to develop innovative therapystrategies.de
