dc.description.abstract | Im Mittelpunkt dieser Arbeit stand das 2005 erstmals beschriebene murine lysosomale Matrixprotein 66.3-kDa-Protein. Die Interaktion des 66.3-kDa-Proteins mit der lysosomalen Aspartylprotease Cathepsin D, welche während der ausführlichen molekularen Charakterisierung vermutet wurde, konnte im Rahmen der vorliegenden Arbeit mit verschiedenen Interaktionsstudien (Ko-Immunpräzipitation, Pepstatin-A-Chromatographie, Vernetzung mit heterobifunktionalen Quervernetzern) bestätigt werden. Das zuvor beschriebene Prozessierungsmuster für das 66.3-kDa-Protein konnte durch die Identifizierung eines weiteren Fragments mittels MALDI-TOF-MS/PMF und Western-Blot-Untersuchungen vervollständigt werden. Die Prozessierung des 66.3-kDa-Proteins stellt sich demnach als zweistufiger autokatalytischer Prozess dar. Die drei entstandenen Fragmente bleiben nicht-kovalent miteinander verknüpft und stellen vermutlich die aktive Form des 66.3-kDa-Proteins dar. Zudem konnte mittels Gelfiltration und Querv! ernetzung gezeigt werden, dass je zwei 66.3-kDa-Proteine als funktionell stabiler Homodimer aus jeweils 2-3 Untereinheiten vorliegen. Durch Kristallisation des 66.3-kDa-Proteins konnten Röntgenbeugungsmuster generiert werden, mit deren Hilfe die dreidimensionale Struktur des Proteins aufgelöst wurde. Der Vergleich der dreidimensionalen Struktur des 66.3-kDa-Proteins mit bereits bekannten Proteinstrukturen konnte auf Grund der charakteristischen Anordnung der Peptidkette in einer αββα-Konformation die Zugehörigkeit des 66.3-kDa-Proteins in die Superfamilie der Ntn-Hydrolasen (Enzymklasse 3.5.1) zeigen und eine hydrolytische Funktion im Lipid-Stoffwechsel bei der Spaltung von linearen, nicht-peptidischen Amidbindungen vorhersagen. Mögliche Substrate für das 66.3-kDa-Protein wären somit u. a. verschiedene N-Acylethanolamide oder Sphingosine, sowie hydrophobe Proteinmodifikationen mit Amidbindungen wie die N-Myristoylierung, Glypiation oder Lysin-Acetylierung. | de |

dc.description.abstracteng | The focus of this study was the murine lysosomal matrix protein 66.3-kDa protein, first described in 2005. The suggested interaction of the 66.3-kDa protein with the lysosomal aspartyl protease cathepsin D was confirmed by various interaction studies (co-immunoprecipitation, pepstatin A chromatography, cross-linking with heterobifunctional cross-linkers). The former described processing pattern for the 66.3-kDa protein could be completed by identification of a further fragment using MALDI-TOF-MS/PMF and western-blotting. Maturation of the 66.3-kDa protein therefore performs as a two-step autocatalytic process. The three resulting fragments remain non-covalently linked and presumably present the active form of the 66.3-kDa protein. Additionally, gel filtration and cross-linking revealed that two 66.3-kDa proteins present as a functionally stable homodimer, each consisting of 2-3 subunits. Through crystallization of the 66.3-kDa protein, X-ray diffraction patterns could be gen! erated, assists to reveal the three-dimensional structure of the protein. Due to the characteristic arrangement of the peptide chain in a αββα conformation and comparison of the 66.3-kDa protein s three-dimensional structure with already known protein structures allowed its classification to the enzyme superfamily of Ntn hydrolases (Enzyme Class 3.5.1) and thus predict a hydrolytic function in lipid metabolism in the dissociation of linear, non-peptide amide bonds. Possible substrates for the 66.3-kDa protein would therefore include various N-acylethanolamide or sphingosine, and hydrophobic protein modifications with amide bonds such as the N-myristoylation, glypiation or lysine acetylation. | de |
